وادریغا!دودمان قاسمی کالعل شب چراغ گم ہوگیا۔چمن زار دارالعلوم دیوبند کا گل سرسبد مرگ کی باد صرصر سے نذرخزاں ہوگیا،بزم علم وعرفان کی شمع فروزاں بجھ گئی،حسن بیان وخطابت کے ایوان میں زلزلہ آگیا،مسند وعظ ومصطبۂ ارشاد وہدایت بے رونق ہوگئے، یعنی ۱۷/جولائی کوحضرت مولانا قاری محمد طیب صاحب کم وبیش۸۸برس کی عمر میں عالم آب وگل کوخیرآباد کہہ کر عالم آخرت کی طرف منتقل ہوگئے۔اناﷲ واناالیہ راجعون۔ شب میں عشاء کی نمازکے بعد ہزاروں ماتم گساروں کے مجمع میں نمازجنازہ دارالعلوم کے احاطۂ مولسری میں ادا کی گئی اورپھرتدفین جدامجد نوراﷲ مرقدہ کے پہلو میں ہوئی، اس طرح گویا:
پہنچی وہیں پہ خاک کہ جس کاخمیر تھی
کل من علیہا فانoویبقیٰ وجہ ربک ذوالجلال والاکرام۔
حضرت مرحوم جب پیداہوئے، یہ دارالعلوم دیوبند کے اوج شباب کا زمانہ تھا۔ اساتذہ کرام اپنے اپنے فن میں یگانۂ روزگار تھے جن کے علم وفضل اور مہارت فن کا آوازہ ممالک غیر میں بھی دور دور تک پہنچا ہواتھا۔پھر اس دور کی ایک بڑی اوراہم خصوصیت یہ تھی کہ اصحاب درس وتدریس خود بھی روحانی اور باطنی کمالات کے حامل اورجامع ہوتے تھے، اوران کے علاوہ تھانہ بھون، سہارنپور اور دیوبند میں مستقل طورپرطریقت ومعرفت کی درس گاہیں قائم تھیں اور دارالعلوم جس کانام تھا وہ درحقیقت انہی دونوں قسم کے علوم وفنون کی تعلیم وتربیت گاہ تھا، غرض کہ ایک طرف یہ سرچشمہ ہائے فیض تھے جوپوری آب وتاب کے ساتھ رواں دواں تھے اوردوسری جانب حضرت مرحوم خانوادۂ قاسمی کے چشم و چراغ ہونے کے باعث ہرایک کے نور نظر اورلخت جگر تھے اورخود بھی ذاتی طورپر نہایت ذہین اور طباع، روشن ضمیر و نکتہ رس تھے اورطلب علم کاجوہر فطری رکھتے تھے، پھر کسر کس بات کی تھی، جوان ہوئے توحافظ قرآن اورقاری خوش الحان ہونے کے ساتھ ایک پختہ استعداد کے بالغ...

Development and Validation of a Self-Concept Scale for College Students Using Comics Superhero Characters

Self-concept refers to the domain of self-descriptions that have self-evaluative connotation. Though many researchers embarked in the study of self-concept, and some even developed tests that measured self-concept, majority of these instruments had methodological and theoretical problems due to lack of systematic instrument development and presentation. The objective of the study is to develop a reliable and valid alternative approach to measuring the self in a semi-structured undisguised comics-type test that directly accounts for the way college students consider their choices of superheroes’ traits that characterizes their own. A preliminary survey on self-concept, in a form of open-ended statements was conducted to five hundred ninety-eight (598) college students of selected schools in Manila and Bulacan to know how college students see themselves indicatory of their self-concepts. Results of which, were collated to form the preliminary form. The preliminary form of the SCSS was administered to five hundred ninety-five (595) college students of different universities and colleges. Eighty-eight (88) items under eight (8) components were subjected to item analysis by identifying factors through a series of exploratory factor analysis (EFA) and confirmatory factor analysis (CFA). Descriptive results were also calculated, as well as the exact reliability coefficient through split-half and Cronbach’s alpha. For the validity, content analysis was applied using two groups of experts who ascertain the suitability of each item in terms of content, relevance, clarity, appropriateness and their representations. They include three (3) experts who have a long experienced in comics industry and another three (3) experts in the field of college students’ self-concept formation. From the total of 88 items, 30 items were eliminated. However, the items that constitute the final form of the SCSS was concentrated into 55 items under six (6) factors upon post-analysis consideration. Statistical analysis revealed that the experts’ ratings were consistent and has high reliability with a generated r value of.894. The SCSS final form was administered to 809 respondents following the same procedures that were used for the preliminary form. The test scores were subjected to reliability facility, such as Alpha Coefficient and Split-Half, computing the reliability coefficients of the final form. Validity was established through convergent analysis, tested in a sample of 419 respondents who took the Tennessee Self-Concept Scale (TSCS: 2) Adult Form. The test was found to have high reliability with r =.792.

Cloning and Expression Studies on Abiotic Stress Tolerance Gene S in Solanum Tuberosum L .

The three dehydrin genes of Eucalyptus species, the dehydrin-1 (EgDHN-1), dehydrin-2 (EgDHN-2) and dehydrin-10 (EgDHN-10) were isolated from local Eucalyptus globulus species for transformation in different varieties of potato for developing abiotic stress tolerance. These three lysine rich dehydrin genes of Eucalyptus have not been reported earlier to be manipulated for genetic engineering or for transformation into a crop plant. The genes were amplified from DNA as well as mRNA (through cDNA). The amplified genes were cloned in TA cloning vector. Cloned dehydrin genes were sequenced and detailed in silico analyses were conducted. Complete sequence of EgDHN-10 gene was obtained from DNA including introns. Coding sequence from its mRNA was also complete and found 92% similar with EgDHN-10 of Eucalyptus globulus after DNA BLAST analyses. The sequencing data of other genes obtained incomplete that showed 99% and 98% homology, after DNA BLAST analyses, with Predicted Eucalyptus grandis Rab18 gene and Eucalyptus globulus EgDHN-1 gene respectively. 297bp long sequence of EgDHN-10 that codes for mature peptide was further sub-cloned in E. coli expression vector pET30a. The sequence was submitted to the NCBI (MG948256.1) and it had extra integration of five electrically charged amino acids (two positively charged Histidine, one negatively charged Glutamate, and two negatively charged Aspartate residues) and one Lysine (K). The presence of one extra K residue is very important for enhanced stress tolerance activity of the transformed dehydrin protein due to its role in functions of K-segments. Presence of more charged amino acids in the binding surface of dehydrin protein enhances its capability to bind with other macromolecules, reactive oxygen species and free metal ions.IPTG induced successful expression of EgDHN-10, cloned in pET30a, was detected in E. coli BL21 ii DE3 strain by protein dot blot analysis. After getting excellent expression of Eucalyptus globulus EgDHN-10 gene outside Eucalyptus cell’s environment, the gene was sub-cloned in pCAMBIA 1301 plant binary vector and transformed into Agrobacterium tumefascience. The nodal regions of three potato cultivars, the Cardinal, Sante and Lady Rosetta, were infected with transformed Agrobacterium (through co-cultivation) carrying EgDHN-10. Putative transgenic plants were first screened by GUS assay and PCR. The successful integration of EgDHN-10 gene in potato genome was confirmed by DNA dot blot analysis. The EgDHN-10 gene’s expression was confirmed from potato mRNA by PCR and real time quantitative PCR. In some plants up to 12 folds DHN 10 expression was observed through qPCR. The acquired abiotic stress tolerance was quantified in stressed transformed potato plants by ion leakage assay (ILA) of cell rupture to select the stable transgenic plants. The stress tolerant transformed plants of Cardinal, Sante and Lady Rosetta were maintained in plant tissue culture setup and pots. The transgenic Sante variety was multiplied and maintained in CAMB trial field in an experiment of growth promoting effect of biofertilizer on trial field plants. The stress tolerant Sante plants showed significant difference over control plants in studied plant growth parameters like plant height, maximum root length, number of roots and number of multiple shoots formation, number and weight of tuber formation.
