Over 9 million new active tuberculosis (TB) cases emerge each year from an enormous pool of 2 billion individuals latently infected with Mycobacterium tuberculosis (M. tb.) worldwide. About 3 million new TB cases per year go undiagnosed, and 1.5 million die (equivalent to AIDS related deaths). Involvement of various host related and environmental risk factors increasing the susceptibility of the disease and emergence of MDRTB has made the control and eradication of the disease difficult. TB, however, is generally curable if diagnosed correctly and in a timely manner. The current diagnostic methods for TB and MDRTB, including state-of-the-art molecular tests, have failed in delivering the capacity needed to curtail this ongoing pandemic. Highly efficient, accurate, cost effective and scalable diagnostic tests are critically needed. We report a multiplex TB serodiagnostic panel for TB infection and a DNA based tests for MDRTB, based on microbead suspension array. Our serodiagnostic panel demonstrates sensitivity of 91% for confirmed TB cases on serum/plasma samples from patients. Sensitivities of this new test format for sputum smear positive and negative cases are 95%, and 89%, respectively, and this test has specificity of 96%. Similarly our DNA based test for MDRTB mutation has 86% sensitivity for Rifampicin, 92% for Isoniazid, 62% for Streptomycin and 63% for Ethambutol resistant samples. This test has shown 100% specificity and 100% correlation between sputum and culture samples of MDRTB. These diagnostic criteria for the multiplex TB & MDRTB test are greatly superior to that of the frontline sputum smear test (30-70% sensitivity) and conventional culture and DST. The test can be run on either blood plasma/serum or dried blood spots (DBS) for TB infection and on DNA isolated from sputum/culture of MDRTB sample, enabling use in almost any setting with scalability from 1 to 360 patients per day, and is amenable to automation for higher (1000s per day) throughput. We also have reported the involvement of well-established host related, environmental and social risk factors in susceptibility of the disease. For host-related factors, TB was found to be significantly associated with male gender, marital status, smoking, drinking, personal and family history of TB, asthma and diabetes (OR: 1.08, 1.96, 1.21, 4.26, 2.07, 3.16, 3.43 and 3.67) respectively with P-value <0.001. For environmental and socio economic factors TB was found to be associated with adult crowding, increased family size, poor ventilation and use of biofuels (OR: 4.60, 1.75, 3.29 and 3.90) illiteracy, unawareness of the disease, migration and presence of animals in the house (OR: 1.74, 0.07, 1.83 and 1.60) respectively with p-values of <0.005. Thus in combination with information of demographic details, risk factors information and use of the proposed test, diagnosis of TB and MDRTB can be possible in one day without any delay in initiation of the treatment. This approach can help TB control program to control the disease by diagnosing it efficiently, accurately, and timely.

Pakht┴n society has its own justice system which has different types of penalties and remedies to maintain the justice in the society. This study concentrates to investigate the nature of pecuniary punishment’s practice in distressing issues like killing, civil injuries and criminal offences. Perpetrators are punished to resolve the disputes. On one hand, this paper aims to find out answer to the methods of inflicting decisions in Pakhtan’s cult and on other hand, to shed light on the legal status of arbitration regarding resolving such issues in the light of Qur’an and Sunnah. Study results illustrate that in some cases the offenders are charged in term of money to facilitate the victims, while in other cases both of the parties, perpetrators and victims, are called upon on meal for reconciliation of their dispute. Besides this, sometimes it is observed that the offenders are not only awarded pecuniary punishment but they are exiled as well.

Diversity of Vascular Plants, Ethnobotany and Their Conservation Status in Ushairy Valley, Distt. Dir Upper Nwfp Northern Pakistan

The present studies were carried out to explore the vascular plant diversity and investigate the ethnobotanical potential and their conservation status in Ushairy Valley. The study area is located in District Dir (Upper) of the North West Frontier Province (NWFP), Pakistan. The area is gifted with diverse and unique flora because of the extension and representation of all the three mountain ranges i.e. Himalaya, Hindu Kash and Karakorum. The people are mostly poor and depend on plants for their domestic needs. Extensive exploration of Ushairy Valley shows that 562 angiosperm species are found here representing by 107 families (17 families of monocots and 90 families of dicots) and 343 genera. Among these families Asteraceae is the leading one with 50 species followed by Rosaceae with 35 species and Brassicaceae with 32 species. The gemnosperm are represented by 4 families with 9 genera and 11 species in which Pinaceae is the leading one with 6 species followed by Cupressaceae with 3 species. The Pteriodphyte represented by 37 species, 16 genera and 10 families in which the Dryopteridaceae is the leading one with 7 species and followed by Aspleniacaceae and Pteridaceae with 6 species each in Ushairy Valley. The collected species were identified, preserved and voucher specimens were deposited in the herbarium of Quaid-i-Azam University, Islamabad and Department of Botany, Islamia College Peshawar. The ethnobotanical information revealed that there are 26 different usage of 274 plant species in which 151 used as medicine, 57 fuelwood in which 16 species are preferred as a best quality fuel, 35 vegetable and pot herb, 16 fencing and hedges, 83 fodder in which 53 are herb, 11 are shrub and 19 are tree species, 15 furniture and 19 species are used as timber. Other ethnobotanical uses of plants in Ushairy Valley as, for making agricultural appliances used 12 spp., in thatching and roofing 14 spp., in making tooth brushes, 7 spp., as insect repellent, 5 spp., honeybee attractants, 19 spp., 8 spp. are used in spices and condiments, 9 spp., used in scorpion sting, dog and snake bite, 7 spp., used in snuff preparation and 19 spp., used in fish poisoning or considered as poison plants. 20 spp., are used as dye and for yielding oil, 36 spp., are edible fruits and seed producing plants, Ornamental plants are 9 and 10 spp., are used as veterinary medicines. The species Abies pindrow, Aesculus indica, Cedrus deodara, Ficus palmata, Pinus Roxburghii, Pinus wallichiana, Quercus spp., and Salix babylonica exhibit multiple uses. The part used data shows that mostly the leaves, whole plant, fruit and stem are in common use. There are 66 species belonging to 45 families used in different monoherbal and polyherbal recipes. It has been discovered that some species are no more available in their respective habitats due to over collection and has been threatened by unsustainable extraction, over grazing, cutting, deforestation and unawareness. One of the major causes of the degradation of forest is poverty that most of the people live below poverty line in hilly areas. Sustainable harvest of natural resources may help to partially alleviate poverty. Programmes for natural resources management must become integral parts of over all developments strategies. The conservation status of Ushairy Valley was evaluated through IUCN (1994-2001) modified criteria. Among these species, 18 are critically endangered, 14 are endangered, 16 are vulnerable and 8 are near threatened.
