Leishmaniasis a group of parasitic diseases related to a major health problem in Asian sub-continent, Africa and rest of world. New discovery and isolation of drug against different types of leishmaniasis is one of the researcher priorities. This works describes an analytical platform based on a semi-high resolution Antileishmanial bioassays in combination with hyphenation of high performance liquid chromatography, solid phase extraction, and tube-transfer nuclear magnetic resonance spectroscopy. i.e., HPLC-SPE-ttNMR/semi-high resolution Antileishmanial assay. The platform enables fast screening for individual leishmania tropica inhibitory analytes in complex matrices, followed by structural identification targeted these L. tropica inhibitors via in vitro, in vivo and cytotoxic test. The plant Lawsonia Inermis was collected from territory of Dera Ismail Khan. Methanol extracts of plant material at concentrations of 10 - 100 µg/ml were tested for them in vitro anti-leishmanial effects on L. tropica KWH23 promastigotes for 24-48 h, relative to negative control and Amphotericin-B (standard drug). For in vivo anti-leishmanial activity, the extract was tested against L. tropica-infected albino mice, while cytotoxicity was investigated against mammalian cells (lymphocytes). For Lawsonia Inermis leaves, mean % inhibition of extracellular promastigotes at 10 µg/ml, 25 µg/ml, 50 µg/ml, and 100 µg/ml after 48 h were 98.2 ± 0.06, 98.75 ± 1.09, 99.31 ± 0.00 and 100.00 ±0.00 respectively. After 8 weeks, mean lesion size decreased from 0.8 ± 0.2mm to 0.3 ± 0.1mm (p < 0.01), and % cure at 150mg/kg against intracellular amastigotes in albino mice was 97.02 (95% C.I = 96.14-98.10). IC50 for Lawsonia Inermis leaves was 12.22 µg/ml (95% C.I = 11.54-13.84) against lymphocytes. The results obtained in this study show that leaves of Lawsonia Inermis are safe and possess potent anti-leishmanial activity. Fractional separation of Lawsonia Inermis leaves was done by applying polarity based solvent system comprised of n-Hexane, Chloroform, Ethyl acetate and Methanol. Phytochemical screening declared that Flavonoids, Coumarins, Phenol and Alkaloids are present in Lawsonia Inermis. Further characterization including identification and evaluation of isolated fractions and compounds was performed via hyphenated HPLC-HRMS-SPE-ttNMR spectroscopic techniques. Active analytes were cumulatively trapped on SPE cartridges and the structures identified by 1H NMR experiments obtained in the HPLC-SPE-ttNMR mode showed six known compounds from the leaves of Lawsonia Inermis; Luteolin (1), Lalioside (2), Luteolin-4’-O-β-D-glucopyranoside (3), Apigenin 4-O-beta-D-glucopyranoside(4), Apigenin (5) and 2, 4, 6-trihydroxyacetophenone-2-O-β-D-glucopyranoside (6). In vitro results showed that luteolin having IC50 of 4.15 value; eliminate the clinically isolated L. tropica promastigotes within 48 hours which is a promising result against leishmaniasis while Amphotericin-B was the reference drug. Lesions sizes were determined in BALB/c mice when luteolin were given to mice while proceeding in vivo study. After 60 days’ treatment, most of mice in Group I, II, III, IV and V were recovered; lesions of few mice were completely healed at 90th day in same groups. In negative control study, 1 mouse died on 45th day and 2 on 90th day. For compound 1, mean lesion size decreased from 0.82 ± 0.12 up to 0.10 ± 0.01 after 120 days, with 97% cure of intracellular L. tropica amastigotes at dose 15mg/kg, compared to Amphotericin B which had 95% cure at a dose of 30 mg/kg. IC50 for compound 1 was 4.15 µg/ml against lymphocytes. The results indicate that luteolin is a potent inhibitor of L. tropica promastigotes and amastigotes, but it had higher cytotoxic effects against lymphocytes, relative to luteolin-4’-O-β-D-glucopyranoside. To eliminate leishmaniasis through discovery of potent drug, further research work should be done to check the efficacy of luteolin and luteolin-4’-O-β-D-glucopyranoside on dogs. Next step should be monkeys and human beings later on.
مولانا سید مرتضیٰ حسن افسوس ہے کہ گزشتہ ماہ میں جناب سیدمرتضیٰ حسن صاحب کم و بیش پچانوے سال کی عمر میں اپنے وطن چاندپور ضلع مرادآباد میں اورجناب نہال سیوہاروی نے کراچی میں وفات پائی۔ مولانا مرحوم اکابر علمائے دیوبند میں سے تھے۔ علاوہ علم و فضل کے بڑے خوش بیان مقرر، کامیاب مناظر اور واعظ تھے۔ تحریکِ خلافت کے زمانہ میں مرحوم کی تقریروں کی جن میں حقیقت وظرافت دونوں کاخوش گوار امتزاج ہوتاتھا ملک بھر میں دھوم تھی۔حضرت مولانا شاہ رفیع الدین صاحبؒ مہتمم اوّل دارالعلوم دیوبند سے نسبتِ روحانی تھی اور اس تقریب سے قطبِ وقت حضرت مفتی اعظم مولانا عزیز الرحمن صاحب سے تعلق ِخاص رکھتے تھے اورقطب ِعالم حضرت مولانا گنگوہیؒ کی مجلس علمی وروحانی کے مخصوص ہم نشینوں میں داخل تھے، اس لیے ذکرومراقبہ کاشغل بھی رکھتے تھے۔ ایک عرصہ تک مدرسۂ امدادیہ مرادآباد کے روح رواں رہے۔۱۹۲۰ء میں پھر دارالعلوم دیوبند کے ناظم تعلیمات ہوکرچلے گئے۔ اب ادھر پندرہ سولہ سال سے عملاً خانہ نشین ہوگئے تھے۔ خود بزرگ تھے اوربزرگوں کی نشانی تھے، سینکڑوں ہزاروں علما جن میں مولانا سید سلیمان ندوی ایسے بلند پایہ عالم بھی شامل ہیں، ان کے فیض تلمذ سے مستفید ہوئے۔حق تعالیٰ انھیں جنت الفردوس میں مقام جلیل عطافرمائے۔آمین ثم آمین۔ [جنوری۱۹۵۲ء]
Ahmad Showqi is an eminent poet in this era, and he is also leader in Egypt, Arabic World and Europe. He is pioneer in Modern Arabic History. He followed his mentor Mahmood Saami Al-Baroodi. He spent his life in grooming. Arabic thinking and was beginner and elevator of Arabic poetry. He highlighted great civilization of Arabs. This modern movement of Arabic literature is zeal and zest of this period. This article reveals a profound academic and research study of Ahmad Showqi’s poetry. The main focus of the study is the Words and Verses of Holy Quran used by Ahmad Showqi in his poetry. Ahmad Showqi has presented before us a very fine interpretation and true meanings of the Quranic words. Similarly through his poetry he has presented before us the Quranic verses and passages in an effective way. So we may describe his poetry as an interpretation of the Holy Quran.
Regionalism emerged in international politics after the World War II, but the end of Cold War has significantly impacted on global politics and regionalism. It is a new phenomenon and nowadays it is difficult to find a country that is not a member of at least one regional organization. Even some of them are the members of more than one regional organizations and international organizations. Economic Cooperation Organization (ECO) as a regional organization established by Pakistan, Iran and Turkey in1985, but after the Cold War Central Asian countries also joined the Organization and ECO expanded with a new situation. In this study to better explain and analyze regionalism in ECO, nefunctionalism, interdependence and liberal intergovernmentalism theories are examined and used together in the context of globalization. It seems that non of theory alone can explain the perfect situation of the region. The advance of technology and information revolution have considerably accelerated globalization and vice versa. Consequently interdependence amongst nations also has been increasing. Such conditions have affected to rise regionalism as a requirement of nations to promote their interests through regionalism particularly after the Cold War. Thus globalization is a main driver of regionalism, however there is interaction between globalization, regionalism and nation-states. The nation-states can manage to take the best advantages of globalization age and remain as an important actor. In globalization age, economic interdependence between the members on the one hand and interdependence between the regions and countries on the another hand have considerably affected on the shape and progress of ECO. Common historical heritage, culture, social and many other factors also have impacted on ECO. Geo-economic and geostrategic locations of ECO also have had important role in the establishment of the organization. The region is very rich in oil and gas which are the main factors of development. Globalization, growth of technology and economic development of EU, China and India''s have impacted on the requirement of energy. These countries have been suffering from shortage of energy and ECO can help them out their energy crisis. Moreover ECO members are also dependent on manufacturing and technology of advance countries like EU, China and other countries. Thus although the process of regionalism in ECO is not fast like EU, interdependence in energy, trade and many other factors within the region and outside the region can drive an push integration not only in the region but also with the other regions and countries.
