Historic Mughal gardens of Pakistan along with their contents are an important part of its national heritage. The Mughals are claimed to be the first to introduce the formal garden design to Indio-Pak sub-continent. Many changes have occurred overtime in the landscape of these gardens. This situation necessitates the need to restore and conserve the original landscape of these gardens. In this regard present research study had been proposed to be carried out on three purposively selected Mughal gardens namely Shalamar Garden, Tomb of Jahangir and Wah Garden where original landscape plants were searched out through available documentary records, and a list of present plants was prepared and documented. The perceptions of visitors and the information-rich persons were recorded and analyzed for inferring the results. The scanty documentary records and the absence of any landscape plan of these gardens imposed major difficulties in finding out the complete list of original plants. Many factors were found contributing towards the deterioration and destruction of landscape of these historic gardens. These included time factor, invasion of armies, neglect and carelessness, changing government policies, temporality of plant material, insufficient documentary records, and political and communal influence of residents. It was also observed that new plant varieties were being introduced in the landscape of these historic gardens which was a deviation from originality. Lack of consistent policies of the concerned department, lack of sufficient funds and technical staff and the changing priorities of the changing governments were the main reasons of unsatisfactory landscape conservation and restoration work of these gardens. Majority of the visitors expressed their wish to see the original landscape plants of these gardens. The respondents suggested various measures to increase the income of the department for proper landscape conservation and restoration. The major measures suggested by the respondents were an increase in the amount of entry ticket and imposing small tax on exports and imports. In the light of the findings of present study it is recommended that landscape conservation and restoration of these historic gardens should be carried out on scientific bases by following the international charters so that this valuable heritage asset could be saved from the threats mentioned above.

ملیشیا میں مرحوم فخر الدین علی احمد صاحب کے بیمار ہونے کی اطلاع اخبارات میں پڑھی توان سے جو تعلق خاطر تھااس کے باعث تشویش پیداہوئی اور خصوصاً اس لیے کہ وہ دل کے مریض تھے، لیکن۱۱/فروری کی صبح کو انگریزی اخبارات میں مرحوم کے بخیریت ہندوستان واپس پہنچ جانے کی خبر کے ساتھ ان کا وہ فوٹو بھی دیکھا جس میں وہ ہشاش و بشاش اورمسکراتے ہوئے وزیراعظم اور کابینہ کے بعض وزراء کے ساتھ پالم پرہوائی جہاز سے اترکر کھڑے ہیں تودل کو اطمینان ہوااوراﷲ کاشکراداکیا، یہ آٹھ ساڑھے آٹھ بجے کی بات ہے۔ اس کے بعد سوا نوبجے حسب معمول انسٹیٹیوٹ آیا، سب سے پہلے انسٹیٹیوٹ کے ڈائریکٹر کرنل تاج الدین صاحب سے ملاقات ہوئی توعلیک سلیک کے بعد انھوں نے گلوگیر آواز میں کہا:’’سخت افسوس ہے کہ صدر کاانتقال ہوگیا۔‘‘یہ سننا تھا کہ جیسے بجلی گرپڑی اورجی دھک سے ہوکررہ گیا، میں اوروہ فوراً ٹیلی فون پرآئے اورآخر جس خبر پریقین کرنے کے لیے دل ہرگز آمادہ نہیں تھا اُس پریقین کرناپڑا اورعالم یہ ہواکہ:

Efforts in Connection With Education in the Era of the Holy Prophet Muhammad SAW

The Prophet Muhammad (SAW) was the most effective orator of all the times. He knew how to address the hearts, and thus he became the beloved one of the hearts. He first made himself loved, and then sought the ways to educate people. He used a style in his speeches that affected people and made them think, so he achieved permanent behavior change in people by means of the best educational methods. It is fact that in the present times those nations who give priorities to knowledge are ruling over the world in one way or the other way i.e militarily or economically. A very important questions strikes the minds that what is the importance of Duniavi(world) knowledge in Islam. Objectives of this study were to investigate the importance of duniavi(world) knowledge in the light of Quran and hadith and provide a guide line to our young generation for getting knowledge. There are many hadiths that encourage education and criticizes ignorance. Being a member of the Muslim nation it is our responsibility to train and guide our young generation for better future.

Biochemical Characterization of in Vitro Salt Tolerant Cell Lines and Regenerated Plants of Potato Solanum Tuberosum L.

The present investigation deals with the establishment of an efficient in vitro selection strategy to produce salt-tolerant cell lines and subsequent regeneration protocols in potato (cvs. Cardinal and Desiree). The activities of antioxidant enzymes (peroxidase, catalase and superoxide dismutase) and total soluble protein contents of various tissues under stress were evaluated to understand their possible role in salinity tolerance. Exogenous application of ascorbic acid and salicylic acid were also tested for salt stress alleviation. In order to proceed with these objectives, the initial focus was to establish protocols for micropropagation, callus induction and maintenance, plant regeneration, establishment of cell suspension cultures and ex vitro acclimatization of regenerated plants. Three different concentrations of TDZ (10-8, 10-9, or 10-10 M) in MS medium were tested for the purpose of in vitro clonal propagation. MS basal medium fairly supported micropropagation of both the tested potato cultivars followed closely by MS medium supplemented with TDZ (10-10 M). For callus induction and proliferation in dark, internodal segments proved to be a good explant source whereas MS medium fortified with 2, 4-D (18.09 μM) was the best medium composition equally effective for both the potato cultivars. A combination of NAA (2.64 μM) and TDZ (1.00 μM) supplemented to MS medium was the best choice for shoot initiation from callus cultures after 20 and 21 days in Cardinal and Desiree, respectively. Rooting of regenerated shoots was achieved on MS medium supplemented with 8.87 μM BAP, 2.64 μM NAA and 0.123 μM IBA. Cell suspension cultures using friable calluses were developed successfully using MS2 medium for the two cultivars. The best supporting medium for ex-vitro transplantation of potato plants was vermiculite. It was observed in this study that different in vitro growth parameters, i.e., shoot/root length and numbers of roots decreased while number of shoots increased with an increase in NaCl (20-140 mM) concentration in the medium. In Desiree, rosette-type of shoot development initiated at 100 mM whereas in Cardinal it was evident at 120 mM NaCl level. During this investigation, a direct recurrent selection procedure was employed to select salt- tolerant cell lines in potato (Cvs. Cardinal and Desiree) on the basis of sub-lethal concentration of salt. Results have shown more than 50% reduction in relative fresh weight in both the cultivars above 100 mM NaCl. Callus morphology correspondingly changed from off-white to blackish-brown above 100 mM to acutely-necrotic at 140 mM NaCl. Regeneration potential of recurrently-selected callus cultures (100 mM NaCl-treated) on salt- free medium was more pronounced in Desiree as compared to Cardinal. When well- acclimatized recurrently-selected plants were treated with 100 mM NaCl and compared with control plants to check their acquired salinity tolerance, it was observed that recurrently- selected plants showed higher fresh/dry weight and number of tubers in both the cultivars. A slight decrease in protein contents of in vitro Cardinal cultures was observed as the concentration of NaCl (20-140 mM) gradually increased in the media. However, there was an increase in protein contents in Desiree plants when subjected to increasing salt concentrations. In case of in vitro recurrently-selected plants, protein contents were higher as compared to control (non-selected ones) in both the cultivars. The peroxidase activity exhibited a slightly decreasing trend in Cardinal though an increasing one was observed in Desiree with an increasing NaCl level in the medium. In the present investigation, recurrently-selected plants had higher POD, CAT and SOD activities as compared to the control ones in both the cultivars.
