Identification of high yielding stable genotypes is an integral objective of plant breeding programs. Testing of genotypes across environments is required to determine yield stability of genotypes. The specific objective of the current study was to analyze genotype by environment interaction (GEI) of grain yield for 50 genotypes using the additive main effects and multiplicative interaction (AMMI) model. Experiments were planted in an alpha lattice design with two replicates in Peshawar (E-1 and E-3), Hangu (E-2 and E-4) and Kohat (E-5) Khyber Pakhtunkhwa province, Pakistan during 2011/12 and 2012/13. Analysis of variance revealed significant differences among genotypes for all traits, while interactions due to genotype by environment were significant for all traits except days to emergence and 1000-grain weight. Significant GEI justified environment-specific as well as AMMI analysis to identify genotypes with specific and wider adaptation. The AMMI analysis revealed that the first interaction principal component analysis (IPCA 1) captured 64.0% of GEI sum of squares while the second interaction principal component analysis (IPCA 2) explained 25.8% of the interaction sum of square. The AMMI biplot identified G30 as a high yielding genotype followed by G19 and G49, whereas low yielding genotypes were G13, G8 and G7. Being close to IPCA1 axis, the most stable genotype with wider adaptability was G30 followed by G31 and G25. Based on AMMI stability value (ASV), genotypes G18 (2.15), G5 (2.78), G27 (3.72), G44 (4.31), G25 (4.43), G42 (4.57), G43 (5.78), G11 (5.82), G1 (7.66) and G29 (7.81) were found in the given order of relative stability. GGE biplot analysis explained 79.9% (PCA1=56.6 and PCA2= 23.3%) of the total variation. Genotype G19 positioning on vertex in sector E-3, E-4 and E-5, while G30 in sector E-1 and E-2 revealed their specific suitability to respective environments. GGE biplot identified environment E-4 as the most representative environment, whereas G49, G30, G22 and G45 as the high yielding genotypes. Shifted multiplicative model (SHMM) grouped genotypes into four clusters based on similarity/dissimilarity index for grain yield. High yielding and stable genotypesG19, G49 and G30 were placed in group B. Grain yield had positive association with tillers m-2 (r =0.73**), grain weight spike-1 (r =0.57**), biological growth rate (r =0.44**), grain growth rate (r = 0.80**), biological yield (r = 0.41**) and harvest index (r = 0.55*). The SHMM clustering and correlations of yield with other traits inferred that tillers m-2, grain weight spike-1, biological growth rate, grain growth rate, biological yield and harvest index contributed towards higher grain yield. Therefore, these traits could be used as selection criteria for the improvement of grain yield in bread wheat. Stability analysis identified G49 (Wafaq × Ghaznavi-98-3) as a high yielding stable genotype among breeding lines which can be commercialized after fulfilling procedural requirements

افسوس ہے کہ گزشتہ اپریل کی ۲۹؍ تاریخ کو علم و عمل اور فضل و کمال کی ایک اور شمعِ فروزاں گل ہوگئی، مولانا مفتی سید محمد مہدی حسن شاہجہانپوری نے ۹۶ سال کی عمر میں بعارضہ فالج داعی اجل کو لبیک کہا مرحوم اس عہد میں اگلی صحبتوں کی چند بقیۃ السلف یادگاروں میں سے تھے، وہ اتباع سنت، تبحر علم، وسعت نظر، طہارت و تقویٰ زہد و ورع اور کتاب و سنت کی تفسیر و تعبیر میں یگانہ عہد تھے، دارالعلوم دیوبند کی مسند درس و افتا ان کے فیضان کمال سے ایک عرصہ تک بارونق رہی ہے اور سیکڑوں تشنگان علم ان کے منبع فیض سے سیراب ہوئے، وہ بلاشہ معلومات کے دریا، حافظہ کے بادشاہ اور وسعت علم کی ایک نادر مثال تھے، ہر مجلس و محفل میں یکساں ان کی قدر و منزلت تھی، حدیث کے ساتھ فقہ کے جزئیات پر ان کی وسعت نظر مسلم خیال کی جاتی تھی۔

THE TEACHERS AND STUDENTS PERCEPTION ON ENGLISH TEXTBOOK WHEN ENGLISH RINGS THE BELL

Abstract The purpose of this study is to compare the student's and teacher’s perceptions about their current English textbook they used. This study categorized as a descriptive qualitative study. The population of VIII grade students (45 students) of junior high school was involved in this study and an English teacher of them too. The questionnaire and semi-structured interview were used as the instrument of this research. Besides, the guideline of the interview and questionnaire was from [1]Cunningsworth's (1995) theory which is explained about the book evaluation. The data were analyzed through Google form percentage presentation for the questionnaire while transcription, coding was used for the interview section. The result showed, 75% of students believed that the book they used has good quality. While the rest 25% felt it did not fulfill their expectation and need. Besides, the teachers’ perception supports it with some of the books’ part lacks organization and employed too many vocabularies. Whereas the students felt their current level was not suitable with the teachers’ beliefs, but the teacher believed that it was appropriate with the students’ level. So, there were few different perceptions between them.  However, the teacher believed that her role in helping students with textbook usage could help the main point of students’ need due to the teacher has lack of knowledge and awareness to do coursebook evaluation. Therefore, further research needs to be done to make this study more comprehensive.    

Characterizaton and Immobilization of Dextranase from Bacillus Species Using Different Synthetic and Non-Synthetic Polymers

Hydrothermal springs are renowned as ideal ecological niche for wide species of thermophilic microbes. Thermozymes of thermophiles have enticed commercial application owing to their stability against inimical industrial approaches. Current study is designed to explore hot springs of Gilgit, Pakistan for the isolation of thermophile capable of degrading complex polymer dextran. Dextranase not only plays a vital role in sugar processing industry but the enzymatic fractions of polymer are also of significant interest in cosmetics, pharmaceutical and food industries. Besides their vast commercial application, thermostable dextranase from non-toxin producing microbe is still a dilemma in food processing industry in order to develop an efficient and cost effective process. Therefore, efforts are being made to explore new source of thermostable dextranase.Bacillus megaterium KIBGE-IB31 [GenBank accession: KF241867] was identified as thermophile accountable for enhanced production of valuable extracellular dextranase under specific fermentation conditions. Strain characterization was based on phenotypic and genotypic analysis. The distinctive feature of this strain classified it as a GRAS and aerobic thermophile. Maximum production of 464.68 U ml-1 with a specific activity of 160.2 U mg-1 was achieved when 10.0 kDa dextran (15.0 g L-1) was amalgamated with various trace elements and nitrogen sources at 60°C up to 24 hours of fermentation time. Inoculum size and agitation speed also had a positive impact on maximum enzyme yield. Gradient precipitation resulted in 61% recovery of partially purified dextranase from crude sample. Whilst, steady state kinetics of dextranase exhibited high catalytic activity within 5.0 minutes at 50°C in 50 mM sodium phosphate buffer of pH: 07 with Vmaxand Km value of 5191 U ml-1 min-1 and 4.38 mg ml-1 respectively. The behavior of different metal ions revealed activating as well as inhibitory action on the catalytic performance of dextranase however, none of the metal ion was found to be essential for enzymatic reaction. Similar findings was observed with surfactant and solvents. SDS (10 mM) was found to be strong inhibitor while triton X-100 (10 mM) decreased 77% activity of dextranase. Data of stability studies proved thermophilic origin of dextranase as it showed stability against wide range of temperature and pH. Hydrolytic action of dextranase was observed by studying surface topology of dextran through scanning electron microscopy. Thin layer chromatography suggested that the hydrolytic response resulted in the formation of various isomalto-oligosaccharides and these oligosaccharides could be utilized as commercially important prebiotics. Immobilization of dextranase using different interactions with different matrices, bestowed covalent cross linking as influential contact of dextranase with matrix in contrast, to adsorption and entrapment methods. However, cross linking without any matrix that is CLEAs was proven to be an efficient immobilization protocol in terms of stability, reusability as well as reduced the cost due to absence of any matrix. Although the reaction time after immobilization remains same except in entrapment but the temperature and pH optima was shifted after covalent cross linking in both protocols. Being a thermostable biocatalyst the stability of dextranase was high at extreme temperature and pH, but was further improved after immobilization. The recycling efficiency of dextranase in the forms of CLEAs was highest (10 cycles with 67% residual activity) than that of other three immobilized form.
