Oil and fats whether for human consumption or for industrial purposes are largely derived from plant sources. To meet the increasing demand for edible oils and oilcakes, improvements are being made with conventional crops, as well as with other new sources of plant species, that have the ability to produce unique desirable oils. Therefore, several plants are now grown not only for food and fodder but also for a striking variety of products, including oils with nutritional and pharmaceutical attributes. This necessitates the search of new sources of indigenous oils. In the present study new native resource of oil i.e. Bauhinia seeds and apple seeds have been explored. The study is divided into five parts. In first and second part the physiochemical characteristics, fatty acid composition, lipid bioactive, unsaponifiable content of extracted oil of three locally grown Bauhinia species (B. purpurea, B. variegata and B. linnaei) were evaluated. Analysis of fatty acid composition of oil samples revealed 13 fatty acids with chain length C14 to C24. The major fatty acids were Myristoleic acid (C14:1) and lignoceric acid (C24:0), linoleic, oleic and palmitic acid. Tocopherols (α-tocopherol, γ+β-tocopherol and δ-tocopherols) were identified and α-tocopherol is reported first time in this study. The unsaponifiable lipid fraction of Bauhinia species ranged 1.8-3.2%, β-sitosterol, campesterol and stigmasterol were the major sterols which accounted for 84-92%. The proximate compositions of meal residue of all samples were also analyzed to determine the suitability of these seeds meal in animal feed formulations. The results revealed that Bauhinia species could be helpful in understanding the influence of cultivar / variety on the quality of oil. The study revealed that the seed oils of the Bauhinia species grown in Pakistan were found nutritionally important with higher amount of PUFA, tocopherols and sterols. In the third part of study the oxidative stability assessment was done by Differential scanning calorimetry (DSC) and oxidative stability index (OSI) method among three Bauhinia species (B. purpurea, B. variegata and B. linnaei), rice bran and cotton seed oil. B. purpurea oil showed highest oxidative stability. Excellent calibration was achieved between ivDSC T 0 and OSI measurements. The coefficients of correlation were highly significant (P < 0.01) for each evaluation. The coefficient of the determination (R 2 ) for analyzed oils was above 0.9956, showing good linear regression, which revealed that oxidative stability of the oils can be accurately determined by DSC in a short time as compared to OSI method. In fourth part of study Infraspecific variation in composition of Bauhinia purpurea Linn. (B. purpurea L.) seed oil was assessed for regional discrimination. Samples were collected from five cities of Pakistan (Hyderabad, Tandojam, Multan, Pakpattan and Abbotabad). Linoleic acid, α-tocopherol, and β-sitosterol contents were used to find variability and significant difference among five regions and was found to be p<0.0001. On the basis of fatty acid composition, five regions could not be discriminated using PCA, LDA on fatty acids discriminated the regions and cross-validation was found to be 99%. Using tocopherols only one PCA component was extracted and LDA on tocopherols discriminated within the regions and cross-validation was found to be 100% perfect. PCA and LDA plots for sterol composition showed five distinct groups for both statistical protocols and all cases were 100% correctly classified. The results of present study indicated that tocopherols and sterols are better chemotaxonomic marker as compared to fatty acids for regional discrimination of B. purpurea L. In fifth part of study the extracted oil from four apple seed varieties (Royal Gala, Red Delicious, Pyrus Malus and Golden Delicious) from Pakistan, total forty two samples were investigated for their physiochemical characteristics, fatty acids profile and lipid bioactive by GC-MS. The oil content in the seeds of apple varieties ranged from 26.8-28.7%. The results revealed that linoleic acid (40.5-49.6%) was the main fatty acid. The unsaponifiable lipid fraction of apple seed oils ranged from 1.8-2.1%, squalene, β-tocopherol, α-tocopherol, campesterol, avenasterol, β-sitosterol, 9,19-Cyclolanost-24-en-3-ol and Stigmast-4-en-3-one were identified, which accounted for 98- 100%. The variation among the results of both fatty acids and lipid bioactive for four varieties was assessed by principal component analysis, discriminant analysis and cluster analyses. The results conclude that both oil fractions could be applied as a useful tool to discriminate the apple seed varieties.

مولوی عبدا لحق برِ صغیر پاک ہند کے عظیم اردو مفکر، محقق، ماہر لسانیات، معلم، بانی انجمن ترقی اردو اور اردو کالج کے بانی تھے، ا?پ کو بابائے اردو کے لقب سے یاد کیا جاتا ہے۔آپ کی تاریخ پیدائش کے حوالے سے کافی اختلاف پایا جاتا ہے ،لیکن بقول ممتاز حسین مولوی عبدالحق 20 اپریل،1870ء کوبرطانوی ہندوستان کے ضلع میرٹھ کے ہاپوڑ کے قریب سراوہ نامی ایک گاؤں میں پیدا ہوئے۔مولوی صاحب کے والد شیخ علی حسین نے عہد مغلیہ میں اسلام قبول کیا۔ان کے سپرد محکمہ مال کی اہم خدمات رہیں۔مغلیہ دور میں انہیں جو مراعات حاصل تھیں وہ انگریز وں کے دور میں بھی برقرار رہیں۔

مولوی عبدالحق نے ابتدائی تعلیم گھر پرحاصل کی۔ثانوی تعلیم میرٹھ سے حاصل کی۔ 1894ء میں علی گڑھ سے بی اے کیا۔علی گڑھ میں سر سید احمد خان کی شخصیت سے بہت متاثر ہوئے۔1895ء میں حیدر آباد کے ایک سکول میں ملازمت اختیار کی۔اورنگ آباد میں صدر محتتم تعلیمات کے عہدے پر فائز رہے۔عثمانیہ کالج اورنگ آباد میں 1920ء تک بحیثیت پرنسپل فرائض سر انجام دیتے رہے۔
انجمن ترقی اردو:
جنوری 1902ء میں آل انڈیا محمڈن ایجوکیشن کانفرنس علی گڑھ کے تحت ایک علمی شعبہ قائم کیا گیا جس کانام انجمن ترقی اردو تھا۔ مولانا شبلی نعمانی اس کے سیکرٹری رہے تھے۔ عزیز مرزا کے بعد 1912ء میں مولوی عبدالحق سیکرٹری منتخب ہوئے جنھوں نے بہت جلد انجمن ترقی اردو کو ایک فعال ترین علمی ادارہ بنا دیا۔ مولوی عبدالحق اورنگ آباد (دکن ) میں ملازم تھے ،وہ انجمن کو اپنے ساتھ لے گئے اور اس طرح حیدر آباد دکن اس کا مرکز بن گیا۔

Language Localization of Foreign Dramas in Pakistan: Transmogrifying Cognizance Against Zealotry and Bigotry

Language localization codes modern world to dilate trade and surplus. Capitalists approach far off markets by localizing their products to tantalize autochthonous consumers. Creative arts are immensely influenced by dubbed localization. Films and dramas are dubbed and localized in target cultures and languages to amplify the global market. This is proliferating a hybrid popular culture. Localization can be adapted as a tool to educate people and to broaden their approach towards life. This paper is exploring positive impact of localized foreign dramas in conservative and hidebound societies. This study presents a basic survey of culturally and linguistically sensitive localization of foreign dramas and its clout on Pakistani society. Three dramas from different cultures and ethnicities have been selected to analyze the power of localized visual arts to influence the thought of masses. The Korean drama “Dae Jang Geum, A Jewel in the Palace”, The Iranian drama “Roz e Hasrat” and the Turkish drama “Fatima Gul Mera kasur kia ha” are part of this study. This paper focuses on two basic questions. How localization influence a target culture in this global \capitalist world? How can we apply this power of localization to reduce bigotry in Pakistani society?

Rna Interference Based Resistance Against Chilli Leaf Curl Disease Complex

Chilli (Capsicum annuum), a member of the family Solanaceae, is an important spice crop cultivated in tropical and subtropical countries. Chilli leaf curl disease (ChLCD) is a limiting factor for chilli yield across Pakistan and India. Symptoms of ChLCD include severe upward leaf curl with cup-shape, yellowing and stunted plant growth. This disease is caused by begomoviruses (single-stranded DNA viruses (family Geminiviridae) that are transmitted by whiteflies). All three different types of begomoviruses are already reported from chillies. In this study chilli samples showing typical disease symptoms were collected from Faisalabad in the Province of Punjab (Pakistan) during the year 2006. All samples were positive for begomoviruses and Pepper leaf curl Lahore virus (PepLCLV) along with Tomato leaf curl New Delhi virus DNA B and Chilli leaf curl betasatellite (ChLCB) were identified. The DNA of Pepper leaf curl Lahore virus consisted of 2747 nucleotides and had the highest sequence identity (99%) with PepLCLV-[PK: Lah: 04] AM404179). Agrobacterium-mediated inoculation of the partial repeat construct of PepLCLV clone obtained in this study to Nicotiana benthamiana induced very mild symptoms and very low flow of viral DNA were detected in infected plant leaves. Co-inoculation of ChLCB with PepLCLV to N. benthamiana did not affect the symptoms severity or the virus titre. However neither the PepLCLV alone or with ChLCB was able to induce any symptoms on N. tabacum L. and C. annuum. Inoculation of PepLCLV with DNA B of ToLCNDV induced very severe symptoms in N. benthamiana, N. tabacum and symptoms typical of ChLCD in C. annuum. Southern hybridization analysis showed very high DNA accumulation for PepLCLV and DNA B of ToLCNDV in all three plant species. Sequence analysis showed that predicted rep-binding iterons in PepLCLV (GGGGAC) was different with two nucleotides from that of ToLCNDV DNA B (GGTGTC). This indicated tolerance of two nucleotide differences in iterated elements for replication. Based on this study, it is proposed that PepLCLV has been recently mobilized into chillies upon its interaction with DNA B of ToLCNDV. This is the first experimental demonstration of infectivity for a bipartite begomovirus causing ChLCD in chillies from Pakistan and suggests that component capture may contribute to the emerging complexity of begomovirus diseases in the region. NIBGE |School of Biotechnology NIBGE Faisalabad [QAU Islamabad] iv The purpose of this study was to develop a broad-spectrum resistance against ChLCD complex based on the concept of pathogen-derived resistance. A hairpin RNAi construct (peAC1-AC2dsRNA/pFGC) based on overlapping region of highly conserved region of Rep and TrAP of PepLCLV was produced in a binary vector pFGC5941. In order to study silencing efficiency of peAC1-AC2dsRNA/pFGC, the construct was transiently challenged with PepLCLV along with DNA B ToLCNDV. Results showed that the RNAi construct was successful in blocking the viral infection as all tested plants were symptomless. Transgenic tobacco plants expressing this construct challenged with PepLCLV and DNA B of ToLCNDV by agroinoculation and with viruliferous whiteflies showed variable resistance ranging from 6.6% to 93.3%. Lines showing resistance more than 75% were ranked resistant/tolerant while lines showing resistance less than 50% were ranked susceptible. One line TA14 showing 93.3% was ranked as highly resistant/tolerant while the line TA 3.2 showing 6.6 % resistance/tolerance was ranked as highly suscepteible. These lines also exhibited significant resistance against ToLCNDV. The relatively conserved nature of Rep and TrAP and their ability to help in development of resistance against heterologous virus suggested that the technology may be useful to develop broad-spectrum resistance. Plants need broad spectrum resistance because they were often infected with multiple begomoviruses in the field. Some viral proteins interfere with different cell signalling pathways and induce symptoms in plants. For example expression of P6 protein of CaMV in Arabidopsis induced dwarfness in transgenic plants. It is reported that Arabidopsis plants with TIR3 gene mutated (tir3) are also dwarf. P6 transgenic (A7, B6) and tir3 Arabidopsis plants which were resistant to auxin and ethylene also showed resitance to 2,3,5-Triiodobenzoic Acid (TIBA) treatment. It indicates that P6 interacts with a pathway overlapped with TIR pathway. Symptoms in Arabidopsis expressing the P6 protein of CaMV probably comes by disturbance of auxin response factor 10 (ARF10), ARF16, and ARF17 also. Also P6-expressing transgenic Arabidopsis plants showed reduced accumulation of miR160 which is known to regulate ARF10, ARF16 and ARF17. A protocol was also developed for chilli plant regeneration using hypocotyl and cotyledon explants. The study was conducted to observe the effect of genotypes, culture conditions and growth regulators on plant regeneration of chili pepper (C. NIBGE |School of Biotechnology NIBGE Faisalabad [QAU Islamabad] v annuum) genotypes grown in Pakistan including Seedex Pepper (SP), Loungi, Tatapuri and Sanam. Of the evaluated genotypes, SP was found to be the most responsive for both hypocotyl and cotyledon explants. Hypocotyl and cotyledon explants were tested for transformation by A. tumefactions LBA4404 having the 35S GFP/pFGC construct and A. tumefaciens EHA105 with peAC1-AC2dsRNA/pFGC construct. Co-cultivation at different temperatures (22 and 25ºC), photoperiods (16h light 8h dark, and complete darkness) as well as co-cultivation time periods, were evaluated. GFP assays showed that putative transgenic calli had not been transformed and calli died after 40-60 days. The experiment was repeated ten times. The data presented in this thesis should help in devising novel control strategies against Begomoviruses. A combination of novel sources of resistance with natural sources of resistance may help to exploit the technology in the field conditions. However, because most pepper varieties are recalcitrant to genetic transformation, control of diseases caused by the ChLCD complex using this strategy awaits future progress.
