Bioremediation of colored textile wastewater loaded with dyes and metals is a matter of great concern due to the hazardous risks associated with their untreated discharge into water bodies. The present study was conducted to isolate and characterize dye decolorizing metal tolerant bacterial strains for treatment of textile wastewater. For this purpose, 220 metal tolerant bacteria isolated from textile wastewaters coming from different textile industries in Faisalabad were tested for their potential to decolorize four different reactive dyes viz. reactive red 120 (RR120), reactive black 5 (RB5), reactive orange 16 (RO16) and reactive yellow 2 (RY2). Based on the potential to decolorize RR120, RB5, RO16 and RY2 in the presence of a mixture of four heavy metals (Cr, Cd, Zn, Pb), four bacterial strains viz. ZM17, ZM130, ZM160 and ZM183 were selected for subsequent studies. These strain were identified following the amplification, sequencing and bioinformatics analyses of 22 their 16SrRNA, and were named as Raoultella sp. ZM17, Pseudomonas aeruginosa strain ZM130, Pseudomonas sp. ZM160 and Pseudomonas sp. ZM183. The selected bacterial strains were found to have varying levels of tolerance against the presence of different heavy metal ions in the media. Moreover, the decolorizing potential of these isolates was found to be decreased with an increase in the initial dye concentration. Furthermore, the interactive effects of four input variables including salt content, pH, C source concentration and level of multi-metal mixture on decolorization of RB5 by the selected bacterial strains were examined following response surface methodology (RSM) statistical modeling. All the four strains were also found capable of simultaneously removing Cr(VI) and all the reactive azo-dyes under study. However, the strain ZM130 was found to be the most efficient among these strains. Additionally, these strains also showed considerable potential for production of IAA and phosphate solubilization. The strains ZM17 and ZM183 were also found to harbor a dye decolorizing azoreductase Azr gene, whereas, a cadmium tolerant czcABC gene was found to be amplified from the DNA of the strains ZM130, ZM160 and ZM183. Enzymatic analyses of the strain ZM130 indicated that azo-reductase and tyrosinase activities were absent in this strain, however, this strain exhibited considerable activity of laccase and NADH-DCIP reductase activity during the decolorization of RR120. While testing the applicability of the strain ZM130 for treatment of synthetic wastewater, efficiency for color and COD removal of this strain was found to be favored and better in the anaerobic batch bioreactor as compared to aerobic batch bioreactor system. However, presence of yeast extract significantly increased the potential of ZM130 for color and COD removal with attached growth of microbial culture. The potential of the most efficient dye decolorizing metal tolerant bacterial strain Pseudomonas aeruginosa ZM130 for treatment of synthetic colored wastewater containing both RB5 (300 mg L-1) and hexavalent chromium (25 mg L-1) was also tested in vertical soil columns bio-augmented with this strain using sterilized and nonsterilized soil. It was interesting to note that the highest level of color, COD and Cr(VI) removal (˃90%) was achieved in the treatments where the soil columns were bio-augmented either with the sludge alone or the sludge along with ZM130 followed by the strain ZM130 along with addition of yeast extract. Based on the findings of the present study, it might be concluded that the selected bacterial strains, particularly Pseudomonas aeruginosa strain ZM130, harbor a varying but highly considerable potential for bioremediation of different xenobiotic compounds including dyes and heavy metals. Furthermore, the concurrent plant growth promoting ability offers a great promise for increased and sustained crop productivity even under stressed soil environment. Hence, this strain could be used to develop the bio-treatment systems (bioreactors) for addressing the problem of azodyes and metals in the wastewater.

Effect of storage on PHYSIO-CHEMICAL EVALUATION OF PEANUT YOGURT Effect of storage on peanut yogurt

ABSTRACT: Peanuts may be consumed in a variety of processed forms like roasted, raw and processed etc. And represent as a multimillion dollar crop worldwide with many potential dietary benefits as it contains high protein and health effective oils. Objective: The present investigation was planned to evaluate thephysio-chemical properties of peanut milk yogurt by the addition of different concentration of peanut milk (0 %, 10 %, 20 % and 30 %), skimmed milk liquid (60 %, 70 %, 80 %, and 90 %), skimmed milk powder (9 %) and sugar (1 %). Methods: The physico-chemical tests (pH, acidity, moisture, ash, fat, protein, syneresis, and viscosity) were examined after every 5 days of interval for a period of 15 days at 4 ºC. Results: The results of physico-chemical analysis revealed that pH, ash, fat, protein and viscosity decrease during storage period where as acidity, moisture and rate of syneresis increased during storage. Treatment T1 (10 % peanut milk) was comparatively best for manufacturing of peanut milk yogurt followed by T2 (20 % peanut milk + 70 % skimmed milk liquid + 9 % skimmed milk powder + 1 % sugar) while peanut milk yogurt from (30 % peanut milk + 60 % skimmed milk liquid + 9 % skimmed milk powder + 1 % sugar) had the lowest degree of firmness. Conclusions: It was noticed that correlation among fat, total solids and protein contents in peanut milk affect the extent of serum separation and pH of yogurt. Impact of Superpave Mix Design Method on Rutting Behaviour of Flexible Pavements

Effective communication is a key to national progress. Pakistan is located in a region where South-Asia converges with Central Asia and the Middle East. Blessed with extensive natural resources and rich agricultural land, it improves its economy particularly by exporting valuable items. In order to improve trade and economic activities and to materialize regional linkages with China, Afghanistan, Iran, Russia and other neighbouring Central Asian countries, the country is gearing up towards a large infrastructure network. Roads constitute a vital part of the infrastructure. In Pakistan most of the roads are constructed using flexible pavement concept, due to their comparatively low construction and maintenance cost. Pakistan has national highways with a length over 9555 Kilometers and motorways of 515 Kilometers. The drastic increase in traffic volume during the last few decades has resulted in premature pavement failures of almost the whole infrastructure of Pakistan. Premature rutting of flexible pavements is one of the major pavement distresses being faced by the country which is primarily due to uncontrolled axle load and high ambient temperatures. Rutting in asphaltic concrete depends on many factors, such as the composition of asphalt mixes, grading and quality of aggregates, type of binder, percentage of the bituminous binder, air void contents, degree of compaction, environmental conditions, load repetition, the substructure, and the bearing capacity of the subgrade. The objective of this research work was to compare the Superpave, Stone Mastic Asphalt (SMA) and Marshall methods of mix design of asphaltic concrete and to propose rut resisting asphalt mix suitable for local loading and environmental conditions. The mixes selected for the study were dense graded in case of Superpave and Marshall methods whereas gap graded for SMA. A comprehensive testing program was conducted on the samples prepared in the laboratory at the design asphalt contents and aggregate gradations. iiiPhysical properties of aggregates and asphalt were determined in the laboratory confirming to ASTM and AASHTO specifications. Mechanical Properties of Marshall, Superpave and Stone Mastic Asphalt (SMA) were evaluated by performing Indirect Tensile Modulus Test, Uniaxial Loading Strain Test (Creep Test), Dynamic Modulus Test and Wheel Tracking Test under prevailing load and environmental conditions of Pakistan in order to compare the performance of mixes. The study revealed that Superpave mixes performed better than Marshall and SMA mixes in terms of low induced permanent strains, high modulus of resilience, high dynamic modulus and better resistance against wheel rutting during wheel tracking test. Superpave technology can be adopted for the design of Hot Mix Asphalt (HMA) pavements in the country due to its superiority over the conventional mix design procedures. The guidelines for implementing Superpave mix design procedure in Pakistan have been proposed. In addition, a performance grading map has been proposed to be implemented in Pakistan by dividing it into seven zones according to the highest and lowest pavement temperatures.
