Antibiotics are used since 1940 to fight bacterial infections due to versatility and high efficacy against bacteria. Metallo-β-lactamases (MBLs) are zinc ion dependent enzymes that are responsible for the emergence and spread of β-lactam resistance. Clinically useful MBL inhibitors are not yet available. The increasing number of novel members of this family is threatening to global health care. Sequence identities and structural characteristics at active site has led to the inception of MBL subclasses B1, B2, B3 and recently B4. AIM-1 (Adelaide IMipenemase-1) is a novel and poorly characterized MBL that was found in Adelaide, Australia and found to be similar in many ways to numerous other enzymes that pose a threat to human health because of their ability to protect bacteria from penicillin like compounds. Being an exclusive member of B3 subclass AIM-1 is suggested as a potential candidate for broad spectrum MBL inhibitor. In present study, a series of mutant forms of AIM-1 was produced by site-specific mutagenesis. D155, D220 and T223 were selected as second shell metal coordinated residues in case of Zn1 whereas D120 was selected as primary ligand. The native and mutant enzymes were then expressed and purified so that the kinetic properties of the mutant enzymes could be compared with those of the native enzyme. Metal chelators were used to remove the metals from the native and mutant forms of the enzyme so that ITC (isothermal titration calorimetry) could be used to determine the metal binding constants. Kinetic data suggest that Asp120 and second shell ligands have showed clear effects and are important for enzymatic activity. In case of Zn1 D to N mutagenesis in second metal coordinates accommodate the smaller substrate molecules like Carbapenems and can effect metal binding whereas D to A mutation severely impaired catalytic rate. Two exothermic metal binding events were observed where one is stronger and other is weaker. Later, two putative MBLs were identified by protein database search, in silico characterized and compared with AIM-1. Information gained from this study may also be useful in the design of inhibitors that could be used as anti-bacterial agents.

جابر علی سید(۱۹۲۳ء۔۱۹۸۵ء) سیالکوٹ میں پیدا ہوئے۔ ۱۹۴۲ء میں ادیب فاضل کیا اور بنگلور چلے گئے۔ جہاں اپنے بھائی تراب علی کے ساتھ انگریزوں کو اردوپڑھاتے رہے۔ عالمی جنگ ختم ہونے کے بعد سیالکوٹ چلے آئے۔۱۹۴۷ء میں اورینٹل کالج سے فارسی میں ایم۔اے کیا۔ اس کالج میں آپ نے ڈاکٹر سید عبداﷲ اور صوفی تبسم جیسے اساتذہ سے کسبِ فیض کیا۔ ۱۹۵۳ء میں گورنمنٹ کالج جھنگ میں فارسی کے لیکچرار مقرر ہوئے۔(۸۲۵)

ENHANCING REHABILITATION PRACTICES THROUGH ROBUST REGULATION AND INTERDISCIPLINARY COLLABORATION IN PAKISTAN

Rehabilitation forms an important tire in well operating health care systems. Within the areas as mentioned on the World Health Organisation factsheet about rehabilitation the therapeutic professions such as physiotherapists, occupational therapists, speech and language therapists, prosthetists and orthotists and clinical psychologists play an important role. They do not only treat people’s conditions such as diseases, disorders, injuries or trauma, they furthermore promote good health and help to prevent diseases. This can significantly lead to better overall wellbeing of the population. To gain the best client outcome and support the health care system in an effective way, regulation is as important as interprofessional collaboration. Thus there is an intense need to highlight these two main aspects to ensure quality rehabilitation provided by therapeutic professionals.

An Analysis of the Effects of Fiscal Decentralization on Economic Growth: A Case for Pakistan

This study examines the effects of Fiscal Decentralization on Economic Growth in the context of Pakistan. The current structure of fiscal arrangements in Pakistan consists of three tiers, federal, provincial and local. Resources are distributed among these tiers through National Finance Commission (NFC) and Provincial Finance Commission (PFC) awards. Besides, local governments also receive random transfers including special/development grants, discretionary funds of parliamentarians and executives, etc. Empirical theorists postulate that Fiscal Decentralization may have positive effects on economic growth due to its efficiency advantages. However, it may have negative effects on economic growth and brings macroeconomic instability when it is not accompanied by enhancing the skill of local government apparatus and by better political accountability. Following the theoretical models of fiscal decentralization presented by Barro (1990) and Davoodi & Zou (1998), we have developed a model for this study in the context of Pakistan and within the framework of a simple endogenous growth model, Johansen’s co-integration technique is used to estimate the effects of fiscal decentralization on economic growth in Pakistan for the period from 1972 to 2010. The study uses expenditures and revenues as indicators of fiscal decentralization and a set of control variables in the analysis.However, due to paucity of data at local level, it is assumed that over the time, public spending and revenue generation is done by two tiers of government i.e federal and provincial. Empirical results show that fiscal decentralization has positive and significant effects on economic growth in the long run but there is weaker evidence for the short run in case of Pakistan during the period of analysis. In addition, fiscal decentralization is also positively associated with private investment and macroeconomic stability over the sample period of the study.
