The project “Chronic effects of metals on their accumulation, DNA damage and oxidative stress in the fish” was conducted in four phases i.e. (i) acute toxicity of metals to the fish (ii) chronic exposure based accumulation of metals in the fish (iii) DNA damage caused by chronic exposure of metals (iv) effects of chronic exposure of metals on oxidative stress in the fish. The acute toxicities (96-h LC50 and lethal concentration) of cadmium, chromium, copper, lead and metals mixture (Cd+Cr+Cu+Pb) were determined for 120 days old three species of fish viz. Catla catla, Labeo rohita and Cirrhina mrigala, separately (n=10), under controlled laboratory conditions. After determination of 96-h LC50 of selected metals and MM, all the three species of fish were exposed to 2/3rd, 1/3rd, 1/4th and 1/5th of their respective 96-h LC50 for 14, 28, 42, 56, 70 and 84 days, separately, and after each period of exposure the accumulation of metals in their organs and the extent of DNA damage in the peripheral erythrocytes and oxidative stress in terms of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) activities in the fish tissues were determined. The sensitivity of three species of fish, determined in terms of 96-h LC50, for all the individual metals and MM varied significantly, showing species-specific sensitivity towards each metals/MM. C. mrigala were significantly (p<0.05) less sensitive to all the individual metals and MM according to 96-h LC50. Exposure of metals mixture caused significantly higher toxicity to all the three species of fish, followed by Pb, Cu, Cd and Cr exposures. The accumulation of all metals in the fish exhibited significantly positive correlation with both dose and duration of exposure. Chronic exposure of metals caused their significantly higher accumulation in C. mrigala, followed by L. rohita and C. catla. However, the overall amassing of metals in the fish, during exposure of individual metal and MM followed the order: Cr > Cd > Cu > Pb. Fish gills, liver, kidney, muscle and brain exhibited significant variability for the accumulation of all the individual metals and MM. Fish liver and gills showed significantly higher ability to concentrate all metals while fish muscles showed significantly lowest ability to amass metals. The overall accumulation of metals in the organs of three species of fish followed the order: liver > gills > kidney > brain > muscles. The accumulation of all metals in the fish body showed significantly direct correlation with the concentration and duration of exposure. The chronic exposure of Cd, Cr, Cu, Pb and MM caused significantly variable damage to the DNA of peripheral blood erythrocytes of all the three fish species under investigation. However, C. mrigala exhibited significantly higher frequency of damaged nuclei, followed by L. rohita and C. catla. The genetic damage index (GDI) values for three species of fish, exposed to metals, varied significantly also. However, both C. mrigala and L. rohita showed significantly higher GDI values. The fish exposed to MM showed significantly higher mean GDI value while it was significantly lower due to Cd exposure. The overall nuclear damage in the fish, caused by individual metals and MM exposures, followed the order: MM > Cu > Pb > Cr > Cd. C. mrigala developed significantly longer tail length of comets while it was significantly lower in C. catla. There existed significant differences among individual metals and MM to induce comet tail lengths in the nuclei of fish erythrocytes. However, exposure of MM induced significantly longer tails, followed by Cu, Pb, Cr and Cd exposures. Therefore, both GDI and CTL of comets may be used to predict genotoxic potentials of various toxicants for the fish.

یہ خبر سن کر بڑا صدمہ ہوا کہ ۴؍ مارچ کو پروفیسر محمد یونس نگرامی کا انتقال ہوگیا، اناﷲ وانا الیہ راجعون۔ ادھر مہینوں سے ملاقات نہیں ہوئی تھی، لکھنؤ آنے جانے والوں سے ان کی علالت کی خبر ملتی تھی مگر یہ خیال نہیں تھا کہ وہ اتنی جلد رختِ سفر باندھ لیں گے۔
ان کی پیدائش لکھنؤ ضلع کے مردم خیز قصبہ نگرام میں ۱۹۴۱؁ء میں ہوئی تھی، ان کا خاندان علمی و دینی حیثیت سے ممتاز تھا، درس و تدریس، تالیف و تصنیف اور وعظ و ارشاد اس کا طرہ امتیاز تھا۔ ان کے والد مولانا محمد اویس نگرامی ندوی دارالعلوم ندوۃ العلماء لکھنؤ میں شیخ التفسیر تھے۔ وہ دارالمصنفین کے رفیق اور مولانا سید سلیمان ندویؒ کے محبوب تلامذہ میں تھے، ان کی صحبت میں مولانا کے تفسیر و قرآنیات کے ذوق کو بڑی جلا ملی، بعد میں وہ دارالمصنفین کی مجلس انتظامی کے رکن بھی ہوئے۔
پروفیسر محمد یونس نگرامی نے اپنے والد ماجد کے زیر سایہ دارالعلوم ندوۃ العلماء میں تعلیم پائی۔ پھر جامعہ اسلامیہ مدینہ منورہ گئے، لکھنؤ یونیورسٹی سے پی۔ایچ۔ڈی کی ڈگری لی، پہلے یونیورسٹی کے شعبہ عربی میں لکچرر اور اب کئی برس سے پروفیسر ہوگئے تھے۔
ان کا تحقیقی مقالہ ’’ہندوستان میں عربی زبان و ادب‘‘ کے موضوع پر تھا، ان کی دوسری کتب و رسائل کے نام یہ ہیں۔ تھوڑی دیر اہل حق کے ساتھ، خیالات، مثالی خواتین، نماز کیسے پڑھیں، تذکرہ مولانا محمد اویس نگرامی، تقدس حج، خلیجی جنگ وغیرہ۔

White Gold was invented in the early 19th century, then it was a mixture of platinum and palladium, but nowadays white gold is a mixture of nickel, platinum, palladium and magnesium, while sometimes it contains copper, zinc and silver. It turns white with color. First White Gold was introduced by Germany in 1912 for sale in the market and then by 1920 White Gold gained popularity as an alternative to platinum. Nowadays white gold is more popular, more favored and is more expensive than yellow gold. White gold is actually yellow gold, with addition of various metals it turns to white so it will apply all the rules that Islam has applied to gold and it is not permissible for a Muslim man to wear its ornaments. However, it is permissible for a woman to wear all kinds of jewelry and Zakat will be obligatory on the man and woman who have the white gold according to the quantity limit prescribed by the prophet (SAW).

Bioactivity Based Proteomics of Selected Medicinal Plants for Drug Discovery

Emergence of multidrug resistance, side effects and heavy cost of synthetic medicine make it a challenge to explore new drugs having no such problems. To overcome these problems, there is a need to explore natural resources like plants. Medicinal plants are most abundant resources and their bioactive properties and possibly undiscovered novel modes of actions can be a solution to control the multidrug resistance. This study is also a part of this big project in which plants are explored to find novel peptides/protein based drugs by mass spectrometric approaches. Eight medicinal plants were screened on the basis of their antibacterial activities. Eruca sativa and Linum usitatissimum showed highest antibacterial potential and selected for proteomic studies. Before moving to proteomics, these two selected plant species were evaluated further for other biological activities viz antifungal, antioxidant, thrombolytic and hemolytic. Significant results proved that the biological potential was due to the presence of bioactive peptides/proteins, as the activities were almost abolished after proteinase K treatment. By bottom-up proteomics, 22 differentially expressed proteins from E. sativa were identified in this study having a good range of biological functions including antimicrobial, antiaphid, antigenic and cardio-protective. Eighteen constitutively expressed bioactive peptides (belong to 7 different classes of AMPs) were identified from L. usitatissimum by top-down proteomics. Literature reveals that these peptides have a good potential against life threatening diseases including antimicrobial, anti-HIV, anti-angiogenic, anticancer, uterotonic and insecticidal potentials. These peptides/proteins may have potential to be used in drug development after further structural and functional characterization.
