HCV infection is a worldwide common health problem having a high affinity for long persistent infections which may develop to considerable changes in liver like hepatocellular carcinoma, cirrhosis and chronic hepatitis. HCV is inherently unstable, giving rise to multiple genotypes and more than 50 subtypes. Mutations that occur during viral replications result in its substantial heterogeneity and overtime in one entity multiple dozen mutant strains may be found. Due to geographical distribution of several different genotypes of Hepatitis C Virus in the world, many of them giving out multiple routes of infection. Constant response to treatment of HCV genotype is a burly forecaster. Assessment of rating and phase is used in evaluating histological outcome in clinical trials or as variables in statistical analyses of progression or therapeutic response. Beginning of amalgamation treatment with ribavirin and interferon has deliberately enhanced clinical result. The achievement of these treatments may be measured in conditions of biochemical comeback and virohistological response. Keeping the above facts in observation the current research is planned to find out the genotyping of HCV RNA causing persistent hepatitis in interior Sindh and to investigate its association with mode of transmission, histological staging and grading, threat and response to therapy. This multi centric study was conducted at Research Medical Centre of LUMHS Jamshoro, Pathology Department of PUMHS Nawabshah and Biotechnology Department of Karachi University, during August 2006 to December 2009. Blood samples of 344 (239 male and 105 female) patients were collected from medical wards of PUMHC Hospital, LUMHS Hospital, CMC Hospital Larkana, and Muhammadi Medical College Hospital Mirpurkhas to cover the population of the whole the interior Sindh. The suspected patients were informed about the study and who also signed a consent form. ELISA for the presence of HCV antibodies was performed and positive patients were submitted to a laboratorial protocol. The clinical and epidemiological data were recorded on a proforma and various biochemical tests were performed. HCV RNA was detected by RT PCR (Quantitative and Qualitative methods), the first viral load was low in 295 (85.96%) cases, medium in 29 (8.43%) cases, high in 15 (4.36%) and very high in 5 (1.45%) cases. The genotying was performed and the results of genotyping revealed that HCV 3a was the major genotype in 242(70.34%) cases, followed by genotype 3b in 19(5.52%) cases, genotype 1a in 10(2.61%) cases, genotype 1b in 5(1.45%) cases, genotype 2 in 4(1.16%) cases, genotype 5 in 3(0.87%) cases and mixed genotype in 9(2.61%) cases. However no other genotype could be identified in 52(15.11%) cases and hence were considered as “untypable”. Determining the risk factor, majority of the patients 135(39.24%) had parental history of multiple use of needles/syringes followed by minor/major surgery in 64(18.60%) cases and dental procedures, transfusion of blood and blood products in 50(14.53%) cases, needle accidents in 34(9.88%) cases, history of nail clippers, tooth brushes shaving razors, and piercing instruments in 35 (10.17%) patients, 10(2.9%) cases had promiscuous sex act history, I/V drug users in 8(2.32%), and tattoo marks in 3(0.78%) cases. The commonest route of transmission in genotype 1a and 1b (60%) was major/minor surgery, each followed by multiple use of needles/syringes; needle accidents and IV drug use. While the commonest route of transmission in genotype 3a, 3b, 2, 5, mixed and untypable was multiple use of needles and syringes followed from the past blood products, transfusion of blood, surgery, history of shaving of razors etc, I/V drug use promiscuous sexual practice (act) and tattoo marks. Histopathological analysis indicates 38(11.04%) cases in grade A1, 151(43.89%) in A2, 117(34.01%) in A3 and 38(11.04%) cases in A4 and no case in A0. Fibrosis seen in stage F2 and F1 in 121(35.17%) and 102(29.65%), followed by F3, F4 and F0. In correlation with genotypes most of the cases of 1a and 1b fall into A3, A4,F3 and F4, while most of cases of genotype 3a, 3b, 2, 5 fall in A1, A2,A3,F1 and F2. The response to combined 6-12 months (depending upon genotype) therapy, 276(80.23%) cases showed positive response, while 68(19.76%) showed no response which include 1a, 1b, 3a, mixed and untypable genotypes mostly with high and very high viral load cases. The sustained response to combined therapy showed that 276(80.23%) had positive response after 6-12 months of combined therapy, while 214 (62.20%) cases had sustained response after 6 months of stopping the combined therapy. Out of 276 cases of positive response with combined antiviral therapy, 272 (98.55%) cases showed low level of initial viral load, while out of 68 unresponsive 48 (66.1%) showed medium, high and very high level initial viral load. The current study verifies the results of various international researches; this is the first ever study conducted which covers the data of the whole interior Sindh. The information gained out of the current study will help improving our understanding of HCV which will be beneficial for clinicians in treating these patients with precise therapy. Furthermore, investigating the association of genotype with histological staging and grading, threat and response to treatment will also show its impact on progression of persistent hepatitis C and curing stratigies. گذشتہ مہینے (۱۳ مارچ) کواجڑنے اور باربار بسنے والی دلّی کی ایک اور فخر روزگار شخصیت مولانا حفیظ الرحمن واصف کی شکل میں اس دنیا سے اٹھ گئی اور دلّی کی بساط علم و دین اورادب وشعر پرچھایا ہوا اندھیرا کچھ اور گہرا ہوگیا، ان کی وفات پر محدود صحافتی اورعلمی حلقوں میں اضطراب اور ہلچل کی کمزورسی کیفیت نظر آئی جو مولانا حفیظ الرحمن واصف جیسی جلیل القدر شخصیت کے ماتم کے لیے نہ صرف ناکافی بلکہ کہناچاہیے کہ ان کی رفعت شان سے حددرجہ کم تر تھی۔ وہ ان ٹمٹماتی ہوئی شمعوں میں سے ایک شمع تھے جوآزادی سے پہلے اورآزادی کے بعد کی دہلی کی تمدنی تبدیلیوں اورلسانی اورسماجی تلاطم کی نوعیت اور کیفیتوں کی عکاس تھی اوروہ خود آزادی سے پہلے اور آزادی کے بعد کے درمیان نہ صرف حدِ فاصل کی حیثیت رکھتے تھے بلکہ ان کاشمارسماجی انقلاب حال کے مشہور ماتم گساروں اور میرؔ،سوداؔ، غالبؔ ،حالیؔ اورداغؔ جیسے نوحہ خوانوں میں ہوتاتھا۔

The Problems Faced by a Translator

Translation is a separate genre and should be treated as one with its specie set of issues, nuances and problem. The Islamic scholars face a peculiar set of problems as their root-medium is Arabic and it is from Arabic to the other languages--- that conversion words a translators' main job are hard to find. The following article is an effort to highlight the set of problems and issues faced by Muslim non-Arab translators while dealing with the medium of Arabic language.

Efficacy of Entomopathogenic Fungi and Their Combinations With Some Selective Insecticides Against Bemisia Tabaci Genn. and Tetranychus Urticae Koch on Cotton

The cotton whitefly, Bemisia tabaci Genn. and Two-Spotted Spider Mites (TSSM), Tetranychus urticae Koch are emerging as important sucking pests of cotton crop. The indiscriminate use of pesticides against insect pests have resulted in the development of resistance among these pests as well as environmental and health hazards. The use of microbial control agents is an important approach to combat this menace. The present study was therefore, carried out to determine the efficacy of some strains of entomopathogenic fungi viz., Metarhizium anisopliae, Isaria fumosorosea, Lecanicillium muscarium and Beaveria bassiana against all immature stages of whitefly and eggs and adult females of TSSM in the Department of Entomology at University of Agriculture, Faisalabad, Pakistan and Department of Pure and Applied Ecology, Swansea University, Swansea U.K. for the bioassays on whitefly. The potential of various entomopathogenic fungi was evaluated on 2 nd instar nymphs of whitefly and four of them were found promising. The detailed studies were carried out on other immature stages of whitefly with the promising strains. The dose- mortality response was observed by using serial dilutions, 1×10 5 to 1×10 8 conidia mL -1 against most susceptible stage using detached leaf immersion method for whitefly and spraying the detached leaf for bioassay with TSSM. All tested fungal isolates caused mortality in the tested stages of both insects under consideration except eggs of whitefly for which very low susceptibility was observed. The L. muscarium (V17) isolate was highly virulent among the tested ones for whitefly nymphs while Pf (n32) isolate was highly virulent against both the stages of TSSM. The mortality of each stage was recorded daily up to 8 days for both the insects under study. The % mortality significantly differed based on fungal isolates, stage of the insect and conidial concentrations. The % mortality increased with increasing the dose of conidial concentration of each fungal isolate in each insect. For whitefly, the most virulent isolate, (V17) has LC 50 value of 1.9×10 5 conidia mL -1 and it killed about 89.58 % of the treated second instar nymphs 8 days post treatment while its LT 50 value at higher concentration is 4.03 days. For TSSM, the most virulent isolate was Pf (n32) with lowest LC 50 value of 9.1 × 10 4 conidia mL -1 on 8 th day and LT 50 value of 4.58 days at 1×10 8 conidia mL -1 . It killed about 82.16 % of the inoculated females 8 days post treatment. The xresults indicated that L. muscarium and I. fumosorosea strains have potential as microbial control agent against cotton whitefly and mites, respectively, in Pakistan. The studies were also carried out to determine the LC 50 values and also efficacy of some commercial insecticides and acaricides against whitefly and TSSM in the laboratory. Four different insecticides were evaluated in the lab against 2 nd instar nymphs of whitefly on cotton. All the tested insecticides caused significant mortality of whitefly from 3-5 days post treatment. However, the most effective insecticides for whitefly, up to 5 days were Mospilon ® and Confidor ® with very low LC 50 values while others were also equally effective but with high LC 50 values. Similarly, the chlorfenapyr 36 SC, pyridaben 15 EC and fenpyroximate 5 SC proved to be the best miticides among the tested ones for the control of TSSM based on their LC 50 values. The nine commercial pesticides and their effects on germination of conidia and mycelial growth of highly efficacious fungal strains viz., I. fumosorosea and L. muscarium were investigated in vitro. There was a significant variation in conidial germination as well as mycelial growth by all tested pesticides depending upon the dose of pesticide and type of fungus. Among the pesticides tested azocyclotin was proved to be highly toxic to germination of spores as well as mycelial growth followed by pyridaben, acetamiprid and propergite while buprofezin was the least toxic. Isaria was proved to be more sensitive at field recommended doses than Lecanicillium strain to all pesticides. Generally, among all pesticides used the insecticides were more compatible to fungi than acaricides although these also inhibited germination of spores and fungal growth to some extent. The combined application of pesticides with selective fungi proved to be synergistic giving more control than used alone. The combined effects were tested under greenhouse conditions on potted plants. Buprofezin, acetamiprid, imidacloprid and diafenthuron were used against 2 nd instar nymphs of whitefly while chlorfenapyr, pyridaben and fenpyroximate were used against TSSM. Only buprofezin and imidacloprid insecticides showed synergistic effects and others additive while no one showed antagonistic results. Chlorfenapyr combined with fungus yielded highest mortality among acaricides tested. However, further investigations under field conditions may reveal more comprehensive results.
