Cytokine Interleukin-2 (IL-2) has a prevalent role in the growth, activation, and differentiation o f T-cells. To suppress immune responses associated with organ transplant rejection and other autoimmune diseases, it is important to disrupt the interaction of IL-2 with its receptor system. IL-2 is now emerging as a target in the discovery of novel therapeutics for addressing the problems related to immune system. The main goal of this study was to establish an effective in silico protocol for identification of IL-2 inhibitors. It describes a pharmacophore based virtual screening combined with docking study as a rational strategy for identification of novel IL-2 inhibitors. Structure based pharmacophore model was developed using the crystal structure of IL-2/IL-2Rα (PDB ID: 1Z92) complex. The predictive pharmacophore model consisted of three features, two hydrophobic and one cationic feature with three excluded volumes. The pharmacophore was validated using a training set of thirty known IL-2 inhibitors. Pharmacophore model as a 3D search query was searched against ZINC and MOE database, in order to retrieve new chemical scaffolds that may be potent IL-2 inhibitors. The hits retrieved from this search were filtered based on their RMSD values and pharmacophoric features. Hits that were retained were used in a molecular docking study to find the binding mode and molecular interactions with crucial residues at the active site of the target. Pharmacophore based molecular docking was carried out on virtually screened compounds using 1Z92 as target by MOE software that led to the identification of 15 hits belonging to diverse classes of heterocycles. These hits were further optimized and a library of forty six compounds including 5-6 membered azaheterocycles namely dihydropyrimidines, heteroazepines, pyrazoles and benzimidazoles besides some compounds such as chalcones and Schiff bases, was designed and synthesized. All newly synthesized compounds were characterized by their MS and NMR spectral analysis. IL-2 inhibition studies on the members of the synthesized library led to the identification of novel IL-2 inhibitors with IC 50 values ranging from <2- >50μg/ml using cyclosporine as a standard drug. This entire set of experiments in both dry and wet labs led to a successful designing and synthesis of a variety of compounds as novel scaffolds that may be developed into interesting immunomodulators.

Youth Perception on Climate Change and Its Impact on Health (Islamabad)

Climate change is a global issue characterized by a shift in regular weather patterns over decades or longer, such as rising temperatures, more precipitation, or decreased humidity. Pakistan is experiencing the consequences of climate change, with increased glacier melt, droughts, and warmer winters and early summers. Pakistan does not make a significant contribution to global warming or climate change. It is, nevertheless, the seventh most severely affected country. In the last century, the average temperature of Pakistan increased by 0.6°C. Pakistan’s government has already initiated the Ten Billion Trees Tsunami Project, one of the world’s most ambitious endeavors to extend and repair its forests. The objective of this study was to investigate the perceptions of youth of Islamabad on climate change and its impact on human health. The mixed methodology (qualitative and quantitative) is used to get better results in apprehensive research work. Our research found that a large percentage of people in the capital city believe climate change is happening and is caused by global warming and deforestation. The youth are aware of climate change and its effects on humans, but they require further education on the subject and guidance on how to mitigate climate change. The government should do something at the national or university level to raise awareness about climate change.

Digital Right Protection of Relational Databases

Design and development of knowledge-preserving and robust water- marking techniques for digital ownership rights protection of relational databases is an active area of research. In this dissertation, various dis- ciplines have been used to design and develop watermarking techniques which ensure knowledge-preserving and robustness characteristics of wa- termarking techniques by bringing tolerable distortions in the original data. Different parameters are used to define data distortions in terms of information loss as a result of watermarking. The intelligent min- ing techniques and statistical measures have been utilized to define and measure information loss after watermark embedding in the relational databases. The data owner usually defines the usability constraints to control this information loss. These usability constraints in turn iden- tify the available bandwidth for watermark embedding. The watermark decoding accuracy of a watermarking algorithm generally depends on this bandwidth; larger the bandwidth the better the decoding accuracy (watermark robustness) and vice versa. However, this dissertation pro- poses a model to make the watermark decoding accuracy independent of this bandwidth and hence the usability constraints; as a result, max- imum decoding accuracy can be achieved even with very tight usability constraints and minimum data distortions. Such mechanism also helps to preserve the knowledge in the databases to a maximum level; as a consequence, the classification results for such databases are also pre- served after watermark embedding. In the pilot study, empirical study and formal modeling have been used to prove the knowledge-preserving and robustness characteristics of the proposed watermarking techniques for digital right protection.
