Effects of Different Preparations of Nigella Sativa Ns on Glucose and Lipid Metabolism in Type Ii Diabetic Patients

The basic aim of this placebo control study was to investigate the effects of three different preparations of Nigella sativa especially on glucose and lipid metabolism in type 2 diabetes mellitus (DM2) patients (group-A); however normal individuals (group- B) was also studied. Both groups were treated with N. sativa Seed Powder (NsP), N. sativa Aqueous Extract (NsE) and N. sativa Oil (NsO), in addition to a placebo in phases 1, 2 and 3 respectively. Each phase was of 80 days (40 days of treatment with N. sativa preparation followed by 40 days of placebo administration). Levels of 0 day were taken as base line (concurrent control) in each phase. Fasting blood levels of glucose, insulin, total cholesterol (TC), LDL & HDL cholesterols, triglycerides (TG), total leukocyte & platelet counts, hepatic profile and blood urea were determined in subjects of both groups on 0, 40 and 80 th day of each phase of the study. In phase-1 a highly significant fall in fasting blood glucose, TC, LDL cholesterol and TG while an increase in insulin and HDL was observed in group-A after treatment with NsP as compared to concurrent control. These levels significantly reversed at the end of placebo except the HDL cholesterol. Almost similar results were obtained in subjects of group-B. In phase-2 although a decrease in glucose and increase in insulin levels was observed in group-A after treatment with NsE; which reversed after the placebo yet the changes were not significant. However a significant fall was observed in TC and TG levels of diabetics after NsE treatment; which reversed after the placebo significantly in TG but insignificantly in TC. In group B, results regarding glucose and insulin levels were similar to that of group-A of this phase. Concerning lipid profile, a significant fall in TG and insignificant decrease in TC and LDL cholesterol was observed after NsE treatment. The changes reversed after the placebo, significantly in TG and LDL cholesterol but insignificantly in TC. In phase-3, a significant fall in fasting blood glucose and a rise in insulin levels was observed in group-A after treatment with NsO2 as compared to concurrent control levels. A significant decrease in LDL & increase in HDL was observed while TC remained statistically unchanged; however a significant increase was also recorded in TG after NsO treatment. LDL and TG levels reversed significantly, while no significant change was observed in HDL and TC levels after the placebo. Subjects of group-B behaved similar to group-A, as for glucose and insulin are concerned. Regarding lipids, a decrease in LDL cholesterol (significant) and TC (insignificant) while increase in HDL cholesterol and TG (insignificant) was observed in group-B after the use of NsO. LDL and TC increased significantly after the placebo while no significant change was recorded in HDL cholesterol and TG. Total leukocyte and platelet counts remained statistically unchanged in both groups A and B, when treated with NsP, NsE, NsO or administered with placeboes in phases 1, 2 and 3 respectively as compared to their base line levels Similarly no unpleasant effect of any of the N. sativa preparation was observed regarding hepatic profile, blood urea and the subjective feelings of the diabetic and normal individuals. In the light of foregoing facts it can safely be concluded that all preparations of N. sativa had some or more antidiabetic effect. However keeping in view the overall performance, NsP was found more effective. Further large scale human studies with different dose schedules of NsP are recommended.

Obesity and Healthy Eating Habits

Obesity is a growing problem, most prevalent in the developed countries, especially US. Children and adolescents are gaining weight at a fast pace as compared to their parents. Although, it is very complicated to exactly know the cause of obesity, its risk factors, its prevention in different populations, yet, it has been observed that changes in life style and particularly in eating habits has contributed to an increased obesity incidence globally. These habits include eating junk food, synthetic and bakery products, increased hoteling, increased consumption of meat and unstaturated fats. If we compare the urban and rural regions at a global scale, we may come to know that Obesity and gastrointestinal problems are more prevalent in urban areas. It also indicates that these disorders are mostly attributed to bad eating habits and wrong choices of food which lack nutritional value but cause various disorders. Atleast 2 decades ago, when there was less digitalization and technological advancements, if people consumed high cholesterol diet or meat, they could digest it as they were physically very active. But in these days, there is less physical activity and our digestive systems cannot digest such food. Moreover, junk food may contribute to obesity. No doubt genetics may also has some role which cannot be denied but the modifiable risk factors can be controlled, monitored and may prevent such disorders. However, global investigations in this regard are direly needed to know the dietary habits and patterns worldwide, their effects in different populations, so that policies may be devised and implemented. Parents are also not aware of a proper healthy pattern that meets the requirements of their children. Knowledge, Attitude and Practice (KAP) Surveys and then awareness campaigns may play a positive role in this regard. Teaching healthy dietary patterns for different age groups at school level may also serve the purpose.

Fabrication and Study of Photovoltaic Devices Using Macrocyclic Organic Semiconductors

During the last decade, different structures of photovoltaic (PV) cells fabricated from organic semiconductors have drawn tremendous economic and scientific interest due to their high optical absorption efficiency, low fabrication cost, lightweight, high mechanical flexibility and continuous growth of their power conversion efficiencies. Bulk heterojunction organic solar cells can be fabricated by simple processing techniques, such as, screen printing, spin casting, etc., and, therefore, are potential candidates for the mass production of flexible and cost-effective devices. In this dissertation, based on the soluble macrocyclic organic semiconductors 5,10,15,20- tetraphenyl-21H,23H-porphine zinc (ZnTPP) and copper (II) tetrakis (4-cumylphenoxy) phthalocyanine (Tc-CuPc) bulk heterjunction structures of ITO/PEDOT:PSS/ZnTPP:PCBM/Al and ITO/PEDOT:PSS/Tc-CuPc:PCBM/Al were fabricated employing spin casting and vacuum thermal evaporation techniques. The effect of donor to acceptor (D:A) mass ratio was investigated on photovoltaic properties of the ZnTPP:PCBM BHJ solar cell and the optimum D:A ratio was identified. Effect of the thickness and surface morphology of the active layer on the photovoltaic properties of this porphyrin-fullerne BHJ was also studied and the optimum active layer thickness was identified. Bulk and hybrid-bilayer heterojunctions of copper (II) tetrakis (4- cumylphenoxy) phthalocyanine (Tc-CuPc) and vanadyl 2,9,16,23-tetraphenoxy- 29H,31H-phthalocyanine (VOPcPhO) heterojunctions were also fabricated. Temperature dependent electrical properties of these devices and optical performance of the Tc- CuPc:PCBM bulk heterojunction was also investigated. Macrocyclic semiconductors e.g. metallo-phthalocyanines (MPcs) and metallo- porphyrins (MPPs), are restricted to dry processing techniques due to their insolubility in common organic solvents. Thus MPcs and MPPs are used in typical thermally evaporated donor-acceptor bi-layered solar cells. The performance of bi-layer solar cells is low due to small exciton diffusion length; only the excitons generated within 10 nm from the D/A interface are expected to contribute to photocurrent. This issue is resolved in BHJ architecture, in which the D-A interfacial area is distributed throughout the volume (bulk) viiiof the active layer. In other words the D-A interface is brought near to the exciton generation site. Soluble versions of MPcs and MPPs can be helpful to study their opto- electronic characteristics in BHJ solar cell architecture. In BHJ architecture there is a lot of flexibility and ease for rapid research e.g. changing solvent, varying concentration, using different D to A mass ratios, changing active layer thicknesses by spin coating at different speeds, incorporating different exciton-blocking layers etc. A lot of variations can be studied in a very short time and with less cost. However, as more and more research groups start studying soluble macrocyclic organic semiconductors in BHJ solar cells, further efficiency improvements and availability of new soluble macrocyclic materials are worthwhile. Since the microstructure of bulk heterojunction layer is strongly dependent on the donor to acceptor (D:A) mass ratio, an optimum D:A ratio improves device performance by improving the charge separation, transport and collection process. As one aspect of this research work, porphyrin-fullerne BHJ solar cells with ITO/PEDOT:PSS/ZnTPP:PCBM/Al structure were fabricated with different D:A mass ratios and their optical parameters were measured both under simulated solar spectrum AM 1.5G and monochromatic illuminations. The active layer thicknesses were extracted from their respective optical reflection and transmission measurements using average absorption coefficient. Scanning electron microscope (SEM) image of the device cross section was studied to verify layer thicknesses and examine the quality of layers. Effect of thermal annealing on performance of some devices was also studied. Incident photon- to-current conversion efficiency (IPCE) spectra at different D:A ratios was determined. On basis of the measured optical parameters such as fill factor FF, open circuit voltage V OC , short circuit current density J SC , and power conversion efficiency η , the optimum D:A ratio was identified. Maximum IPCE value of 21% was obtained for D:A ratio of 1:9 which lead to PCE of 0.21% which was 36 times better than a previously reported value in which C 60 was used as acceptor. Also incorporating PCBM as the acceptor instead of C 60 improved the open circuit voltage (V OC ) for all the D:A ratios. The reason for high V OC is greater difference between the ZnTPP HOMO and PCBM LUMO . Furthermore the 1:9 devices showed consistency in optical parameters when reproduced.
