Plant response to the altered environmental conditions usually includes initiations of several defense mechanisms. One of these mechanisms is the heat shock proteins (HSPs) production, a universal response. Chloroplast small heat shock proteins (Cp-sHSPs) are known to protect Photosystem II under stress conditions in plants. Cp-sHSPs production is positively correlated with the plant thermotolerance. While the protective role of HSPs has been greatly recognized against several physiological stresses such as heat, metals, cold and different developmental stages individually or in different combinations. Recent progress in this field has revealed unique aspects of regulations of these genes under stress. Cis-regulatory elements present in promoter regions with the combinations of several factors dictate the gene expression levels and specific patterns. Not much is known about the regulation and mechanism of action of Cp-sHSP genes in plant protection. We analyzed four novel promoters of Chenopodium album (two US ecotypes) Cp-sHSPs for the presence of putative cis-regulatory motifs (Shakeel et al., 2011). Interestingly we found more than one cis-regulatory elements in the promoter regions of these Ca-sHSPs and based on this unique promoter architecture, we proposed a differential regulation of a single Ca-sHSP gene under variety of different abiotic stresses including heat, metal, cold, drought and salt stresses individually. To analyze the Ca-sHSP gene expression patterns under different abiotic stress conditions, we treated C. album (Pakistani ecotype) plants at different abiotic conditions and initially studied the effect of above mentioned stresses on physiology of the plants. On the basis of physiological data, we decided the lower and upper limits of stress conditions for further analyses of Ca-sHSPs. Stress induced transcripts of Ca-sHSP genes were sequenced individually and analyzed; our data showed that one novel Cp-sHSP family member, CaHSP26.13p is regulated differently under different abiotic stresses. This observation was based on 100% amino acid sequence similarities among these transcripts. Expression of Chloroplast Small Heat Shock Proteins of C. album under Different Abiotic Stresses Relative abundance of transcript and protein levels was determined by real-time PCR analysis and immunoblotting of C. album leaf samples treated with and without stress conditions. Antibodies specific to methionine rich region were used for Cp-sHSPs detection by immunoblotting. Interestingly we detected 1-2 bands of precursor and processed Cp-sHSP proteins (~26 and 21KDa) depending upon the stress conditions, while only precursor protein (~26KDa) with differential expression was detected in all types of treatments suggesting the role of single Ca-sHSP in plant protection under heat, metal, cold, drought and salt stress. Due to the absence of correlation between transcript and protein levels in most of these cases, we speculate some role of post-transcriptional regulation in plant protection by this mechanism. This study demonstrates multiple roles of single C. album Cp-sHSP in variety of environmental conditions by differential regulation and can be used to develop stress tolerant plant species to face the challenge of next decade. اعظم گڑھ کے متعدد علماء کو بین الاقوامی شہرت نصیب ہوئی خود مبارکپور کے جو اعظم گڑھ کا مشہور قصبہ اور علم و تعلیم اور صنعت و حرفت کا بڑا مرکز ہے، مولانا عبدالرحمن مبارکپوری صاحب تحفۃ الاحوذی اور مولانا عبیداﷲ رحمانی شارح مشکوٰۃ المصابیح کا آوازہ شہرت عالم اسلام میں بلند ہے، انہی لوگوں کی صف میں مولانا قاضی اطہر مبارکپوری نے بھی اپنی جگہ بنالی تھی، مگر افسوس کہ دست اجل نے انہیں ہم سے چھین لیا، یہ بڑا علمی سانحہ ہے اور قاضی صاحب کا خلا پر ہونا آسان نہیں۔
ان کی ولادت ۱۹۱۶؁ء میں ہوئی، ان کے نانا مولانا احمد حسین رسولپوری عربی زبان و ادب کے ماہر اور صاحب تصانیف کثیرہ تھے، قاضی صاحب نے ان کا عربی دیوان مرتب کر کے شائع کیا تھا، انہوں نے عرصہ دراز تک ڈھاکہ میں مسند درس کو رونق بخشی۔ انہی سے قاضی صاحب نے عربی کی ابتدائی کتابیں پڑھیں تھیں پھر مبارکپور کے مشہور مدرسہ جامعہ احیاء العلوم میں درسیات کی تکمیل کی اور جامعہ قاسمیہ مراد آباد میں مولانا فخرالدین، مولانا سید محمد میاں اور مولانا سید اسماعیل سنبھلی سے صحاح ستہ کا درس لیا۔

According to Islamic Criminal Law, Apostasy is a crime, severely punishable in this world and hereafter. Some people are of the view that worldly punishment is not mentioned in Quran. Rather the rule has been given that a man is free to choose or give up any religion; no one would be compelled in this regard. This is essential to contemplate this critical matter; What is the accurate interpretation of the verses of the Quran in this regard? How the worldly punishment of Apostasy is derived from Quran? What is the meaning of "There is no coercion in Islam"? Either Islam prohibits every form of coercion or it has also been used in some commands of Islam?  In this article, a critical analysis of these doubtful questions regarding the punishment of Apostasy is presented in the context of the Holy Quran.

Exploring Teachers Knowledge and Skills to Do Formative Assessment in Chemistry Teaching at Secondary Level in a Public School of District Skardu, Gilgit-Baltistan

Assessment for learning has been given prime importance in K-12 education across the countries. This study aimed to explore the assessment practices and perceptions of two grade IX Chemistry teachers in a Government secondary school Gilgit-Baltistan, Pakistan. The study adopted qualitative case study design, where data were generated through semi-structured interviews, classroom observations, and document analysis. The findings of this study highlight that the teachers perceive that effectiveness of assessment is determined by its potential to promote further learning. Such assessment practices not only improve student’ learning but also provide opportunities to teachers for reflection and improvement of their instruction. The participant teachers have knowledge about number of formative assessment techniques to gauge students’ learning during the class. Among them, questioning and feedback were the two key elements of participant teachers’ assessment practices. Teachers argued that extensive reason based questioning for assessment purposes provided a venue to students to share their science conceptions in class and get feedback on them by fellow students and teachers to check whether their own conceptions are in line with established scientific ideas. The study also identified that the teachers were very concerned about monitoring students’ learning. However, teachers’ assessment practices are much influenced by the local board examinations which are mainly based on regurgitation and time bound paper-pencil tests. Because of expectations from parents and school management, teachers were obliged to train students to get high marks in the board examination. Therefore, it is recommended that the teachers’ assessment during the course of study should be given some weightage in the cumulative board examination results. Teachers should also be given professional development extensive repertoire of assessment techniques including use of ICT. There should be consultative dialogue with parents to make them understand the importance of students’ creative work which may not reflect through their marks and grades.
