Circadian rhythms are endogenous, self-sustained oscillations of multiple biological processes with approximately 24-hr rhythmicity. Circadian genes and their protein products constitute the molecular components of the circadian oscillator that form positive/negative feedback loops and generate circadian rhythms. The circadian regulation extends beyond clock genes to involve various clock-controlled genes (CCGs) that include various cell cycle genes. Aberrant expression of circadian clock genes could have important consequences on the transactivation of downstream targets that control the cell cycle and on the ability of cells to undergo apoptosis. This may lead to genomic instability and accelerated cellular proliferation potentially promoting carcinogenesis. The current study was carried out to gain further insights into the roles of circadian genes and their downstream targets (cell cycle genes) in chronic lymphocytic leukemia (CLL). We analyzed peripheral blood from 37 CLL patients and equal number of their age- and sex-matched healthy controls for the expression of the four circadian clock and three cell cycle genes. The expression levels of BMAL1, PER1, PER2,MYC, CYCLIN D1 and WEE1 were significantly impaired in CLL cases compared with those in healthy individuals (P < 0.001). BMAL1, PER1, PER2 and WEE1 were found down regulated whereas MYC and CYCLIN D1 were found upregulated. This implies that the deregulated expression of circadian clock genes through their influence on downstream clock-controlled cell cycle genes can play a role in the manifestation of CLL. Moreover, when expression levels of abovementioned genes were compared between shift workers and non-shift workers within the CLL group, the expression levels were more aberrant in shiftworkers compared to non-shift workers. This indicates that circadian dysregulation in terms of shift work may also be a contributing factor in the etiology of CLL. In the current study, serum melatonin levels were also determined in 37 CLL cases and their healthy controls. Serum melatonin levels were found significantly low (P<0.05) in CLL subjects as compared to healthy controls. Furthermore, melatonin levels were found still lower in shift workers as compared to non-shift workers within CLL group. Our results suggest that down regulation of BMAL1, PER1 and PER2 is related to upregulation of Cyclin D1, MYC and down regulation of WEE1 in CLL. Thus, aberrant expression of clock genes can lead to abnormal expression of downstream cell-cycle genes and play a role in the manifestation of CLL. Moreover, low melatonin levels in CLL patients may play a part in xivderegulation of circadian clock gene expression and shiftwork serves as a further contributing factor to an already perturbed circadian clock genes’ expression and low melatonin levels in CLL.

قرآن مجید فصاحت و بلاغت کےاعتبار سے ایک مثالی کتاب ہے۔علم معانی ہو یا علمِ بیان یا علم بلاغت کے ماہرین نے اس کی لسانی وادبی خصوصیات کو نگارشات کا موضوع بنایا ہے۔ فصاحت و بلا غت کی انہی خوبیوں کی بنا پر قرآن مجید کو کلام مبین بھی کہا گیا ہے۔ قرآن مجید میں جہاں ایجاز و اطناب اور ربط و مساوات کے اسالیب نظر آتے ہیں، وہیں تقدیم و تاخیر اور حذف کے اسلوب بھی نمایاں ہیں۔ قرآن مجید کے مختلف و منفرد اسالیب میں سے ایک اہم اسلوب ِ استفہام ہے۔
استفہام عربی زبان کا لفظ ہے جس کے معنی سوال کرنے، جاننے،فہم حاصل کرنے اور استفسار کرنے کے معنوں میں استعمال ہوتا ہے۔ ادب میں استفہام کسی حقیقت سے مخاطب کو آگاہ کرنے ، مخاطب کو غورو فکر کی دعوت دینا، اور اپنی بات کا ثبات کرنا وغیرہ کے معنوں میں مستعمل ہے۔چنانچہ قرآن مجید میں بھی استفہام کا اسلوب بکثرت استعمال کیا گیا ہے۔

IMPLEMENTASI KEBIJAKAN SISTEM ZONASI PADA PENERIMAAN PESERTA DIDIK BARU (PPDB) 2019/2020 DI KOTA YOGYAKARTA

The acceptance of new students (PPDB) using the zoning system was first implemented in Yogyakarta in 2018/2019. This zoning system aims to equalize education and improve the quality of education. This research was conducted to find out how the zoning system policy in Yogyakarta city under the auspices of the Yogyakarta City Education Office. This research uses qualitative approach with case study method. The results of the study showed that the zoning system in Yogyakarta has changed from the previous year. In 2019/2020 the zoning system provides a 10% chance of superior seeds, 30% of school zones, 10% of poor learners, 40% of quality pathways, 5% of out-of-school zoning pathways, and 5% of special pathways such as prospective learners whose parents moved from other regions to Yogyakarta. With this zoning system, the whole community has the opportunity to get a good quality education.

Effect of Natural Growth Promoters on the Nutritive Quality and Antioxidant Attributes of Leafy Green Vegetables

Epidemiological evidences encourage the suitability of intake of fresh leafy green vegetables to combat against a list of neurodegenerative disorders including diabetes, breast cancers, cardiovascular and other diseases. To meet such targets, the field of agriculture has taken a mold towards the betterment of nutritional status of these vegetables via number of plant growth regulating substances. These economic strategies are implemented to nullify the use of pesticides and other environmental threatening agents. In current study, the application of different plant growth promoters (Moringa leaf extract, 6-Benzyl amino purine, Humic acid, Biofertilizer etc.) was appraised on some leafy green vegetables. Moringa leaf extract, 6-Benzyl amino purine and Humic acid, alone and in conjunction with each other (mixture) were applied in foliar mode while Humic acid (6 h, 9 h, 12 h duration) and Biofertilizer (slurry form) were applied as seed priming treatments. The optimized results of pot study were selected on the basis of antioxidant activity on which field experiment was raised later on. Field experiment included various parameters i.e. soil chemistry analysis, growth parameters, biochemical, proximate and antioxidant attributes. Although all treatments of plant growth regulators improved the efficacy of plant against control samples but Moringa leaf extract proved the potent plant growth regulator amongst all regarding biochemical, proximate, growth parameters and as an antioxidant action. Among other foliar treatments, 6-Benzyl amino purine and mixture treatments yielded comparable results and Humic acid showed the least performance. Among seed priming treatments, Biofertilizer and Humic acid (9 h) treatments showed better performance than rest of the treatments i.e. Humic acid 9 h and 12 h. Urea presence in soil showed improved results than in absence in all cases except in case of antioxidants. Overall study concludes that natural plant growth promoters can be a better and economical substitute of fertilizers in view of environmental deterioration. Moringa leaf extract can be preferred as an effective plant growth promoter due to its inherited composition of vitamins (A, B, C, E etc.), minerals (Na, Mn, Mg, K, P, Ca, S, Zn, Fe, Cu etc.), cytokinins, humic acid and other suitable bioactives.
