Formulation Development and in-vivo Performance of Pharmaceutical Cocrystals The use of soluble cocrystal for delivering drugs with low solubility, although a potentially effective approach, often suffer from the problem of rapid disproportionation during dissolution, which negates the solubility advantages offered by the cocrystal. This necessitates their robust stabilization in order for successful use in a tablet dosage form. The cocrystal between carbamezepine and succinic acid (CBZ-SUC) exhibits a higher aqueous solubility than its dihydrate, which is the stable form in water. Using this model system, I demonstrate an efficient and material-sparing tablet formulation screening approach enabled by intrinsic dissolution rate measurements. Three tablet formulations capable of stabilizing the cocrystals both under accelerated condition of 40oC and 75% RH and during dissolution were developed using three different polymers, Soluplus® (F1), Kollidon VA/64 (F2) and Hydroxypropyl methyl cellulose Acetate sucinate (F3). When compared to a marketed product, Epitol® 200 mg tablets (F0), drug release after 60 min from formulations F1 (~82%), F2 (~95%) and F3 (~95%) was all higher than that from Epitol® (79%) in a modified simulated intestinal fluid. Studies in albino rabbits showed correspondingly better bioavailability of F1 – F3 than Epitol® tablets. All the three experimental formulations gave statistically significant improvements in AUC0-72 of CBZ than Epitol tablets by applying ANOVA followed by Tukey’s post hoc test (*P˂0.05 for F0-F1, ***P˂0.05 F0-F2, ***P˂0.05 F0-F3). Among the three formulations, F2 tablet formation was the best based on both in-vitro and in-vivo evaluations. Towards the second formulation approach, I first used common polymers to study the supersaturation of carbamezepine-succinic acid (CBZ-SUC) cocrystal at different conditions. In-situ Raman spectroscopy was used to monitor the solid phase during dissolution studies. The solid phase at the end of each experiment was characterized by FTIR and powder X-Ray diffractometry. In-vivo study was performed on selected xi suspension formulations. In absence of polymers, no dissolution advantage was attained by cocrystals due to rapid crystallization of CBZ dihydrate. At room temperature Polyvinyl pyrrolidone (PVP) at concentration of 2% w/v did not stabilize the supersaturated solution, whereas polymer concentration of 0.025% w/v hydroxypropyl methyl cellulose acetate succinate (HPMCAS) stabilized the cocrystal and enhanced the solubility of CBZ in buffer solution at pH 6.8 by 3.2folds to that of stable dihydrate form. A formulation of CBZ-SUC cocrystal containing HPMCAS as a crystallization inhibitor and soluplus® as solubilizer, superior in-vitro dissolution performance was achieved compare to pure CBZ having similar composition at 37±1Ċ. Pharmacokinetic studies in rabbits showed that one of the formulations F7-X (1% w/v cocrystal, 1% w/v HPMCAS and 2% w/v soluplus®), caused around 6folds enhancement in AUC0-72 (***P˂ 0.05), and also led to a much higher Cmax of 4.73 μg/mL than 1.07 μg/mL Cmax of ‘neat’ cocrystal given orally. Also, the F7-X outperformed a reference formulation of CBZ by {1.37folds AUC0-72 of cocrystal formulation (***P˂ 0.05) and Cmax of 3.9 μg/mL} with similar composition to F7-X.

مولانا احسان اﷲ خاں تاجورؔ ۱۸۹۳ء میں نجیب آبادضلع بجنور میں پیدا ہوئے، روہیلہ افغانوں کے خاندان سے تعلق رکھتے تھے۔ابتدائی تعلیم وطن میں ہوئی پھر دارالعلوم دیوبند میں داخل ہوکر اسلامی علوم وفنون کی تکمیل کی۔ شعروشاعری اورلکھنے کاذوق فطری تھا چنانچہ زمانہ طالب علمی میں بھی دارالعلوم دیوبند کے بلندپایہ علمی اوردینی رسالوں ’’القاسم‘‘ دوراوّل اور ’’الرشید‘‘میں ان کے مقالات نکلتے تھے۔ یہاں سے فارغ ہوکر وہ لاہور پہنچے اورسرعبدالقادر مرحوم ایسے مربی اورمشفق کے فیض صحبت وتوجہ نے ان کوایسا چمکایا کہ وہ جلدہی نہ صرف لاہور بلکہ شمالی ہندوستان کی علمی اورادبی محفلوں کی رونق وزینت بن گئے۔ وہ اردو کے بلند پایہ شاعر اور زبان کے ماہر و نقاد اور نامور ادیب کی حیثیت سے پنجاب کے اساتذۂ فن کی صف میں ایک نمایاں جگہ کے مالک ہوگئے۔ سینکڑوں نوجوان ان کے فیض صحبت وتعلیم سے اردو کے نامور ادیب وشاعر ہوئے۔ وہ زندہ دلان پنجاب کے ادبی اکھاڑہ میں ایک پہلوان کی حیثیت سے رہتے تھے۔

Last revelation namely al- Quran has addressed the human beings in an eloquent way using all types of expressions and diction. The divine method of articulation for holy commandments is miraculous and opts all appealing techniques of communications that also includes use of homographs and metaphors. The word that are spelled the same but have different meaning are called Homographs. The reciter andreader of the Quran faces some difficulty in deciding the meaning of a Homographs used in the Quran that leads to difference of opinions. In the books of Quranic Sciences this term is called Mushtarak al –Lafzi. The article has been aimed to elaborate what is Mushtarak al –Lafzi and what are the impact of vagueness originated from these words of the Quran on Quranic exegesis. Some examples have been produced fromthe books of Quranic Studies regarding its influence on exegetical literature.

Financing and Managing Poverty Reduction in Rural Pakistan a Case of Dg Khan and Rajanpur Districts

In an attempt to reduce poverty in Pakistan, financial resources are provided by microfinance institutions to poor and vulnerable people to engage in income generating activities on soft terms and conditions. Zakat institution and BISP provide free cash to needy and poor as living allowances. ZI, PBM and NGOs provide financing for human capital development through education and training to manage poverty reduction. These institutions facilitate the poor people directly to manage poverty reduction on sustainable basis. The research presented in this study, hence, aims to explore and evaluate the financial dimensions of managing poverty reduction in rural Pakistan through a micro level study to evaluate the outcome and effectiveness of poverty reduction programmes in Pakistan by focusing on the impact of such programmes in DG Khan and Rajanpur Districts. For this purpose, primary data is collected through a questionnaire survey to measure the perceptions of the households, in the form beneficiaries and non-beneficiaries, on the outcome and efficiency of the poverty reduction programmes in DG Khan and Rajanpur. The data is analyzed through non-parametric (Mean U Whitney tests and Kruskal Wallis) and parametric inferential statistics techniques, such as logit model, to draw the result for research questions. The findings from the non-parametric test have shown that institution, employment, marital status, working female members, working male members, assets like land, livestock, business assets, savings and loan are significant and ranked at 1 for income related questions. Training, education, gender, age, child dependency and district variables are also significant and causing for poverty but ranking at second number. The results from Logit model show that beneficiaries and households are statistically significant and positively correlated with probability of being poor. It also concluded that education, institution, gender, age, employment, working male member, and working female member as variables are statistically significant and negatively correlated with probability of being poor. Additionally efficiency of institutions is tested. Zakat institution is found to be contributing towards working male member and change in income. Working male members are found to be statistically significant and negatively correlated with probability of being poor. However BISP is not contributing in determinant for managing the poverty reduction while PBM is contributing in working male member, which is significant and negatively correlated with probability of being poor. As regards MFI is concerned it is found that it is contributing in education and change in income, which are negatively correlated with probability of being poor. The study concludes that financial capital and human capital development are essential elements for financing and managing the poverty reduction in rural Pakistan. The success of which depends on the coordination of different poverty reduction programmes.
