RB1, CCND1 and CDK4 are major cell cycle check point players related to head and neck cancer (HNC). Polymorphisms in these genes have frequently been reported in literature and are known to follow diverse patterns in relation to different populations. The current study was designed to screen these genes in head and neck cancer patients and control samples at DNA, RNA and protein level. Blood and tissue samples of pathologically confirmed head and neck cases were collected from different hospitals. Polymerase chain reaction (PCR) and single-strand confirmation polymorphism (SSCP) were used for germline screening, followed by sequence analysis. In second step Real time-PCR was performed to study expression of these genes at mRNA level. Immunohistochemistry was performed to evaluate the protein expression of these molecules in cancer tissues as well as controls. Epigenetic analysis was performed using methylation specific PCR. In the last part of the study, in vitro studies were carried out to characterize cancer cells with selected RB1 mutations and their response to drug resistance (doxorubicin). In first part of this study, PCR-SSCP and DNA sequencing were used to analyze the coding regions of RB1, CCND1 and CDK4 in 730 individuals (380 head and neck patients and 350 controls). Sequence analysis of coding regions of RB1 gene results in 18 mutations (one silent, 10 missense, 3 frame shift, 2 nonsense mutations and 2 splice site substitutions). Among these mutations, one silent synonymous mutation was observed in exon 12 (g70282A>G), 5 missense mutations observed in exon 14 (g76474C>T, g76475G>C, g76476A>G, g76467G>C g76468T>C and 2 missesnse mutations in exon 16 (g77041A>T, g77043A>G). One frameshift mutations in exon 12, was a result of deletion of five nucleotide GATGA (g70285_70289delGATGA). 2 frameshift mutations were found in exon 21, one novel frameshift mutation was due to the insertion of nucleotide G (g160601_160602insG) while the other was due to insertion of nucleotide A [(g160623_160624insA) (CI952130)]. In exon 14 and exon 24, 2 nonsense mutations Lys462stop (novel) and Ser834stop (CM952105) were also observed, respectively. Germline analysis of CCND1 coding revealed total 4 mutations, one missense, one frameshift, one silent and one in 3''UTR. One novel missense (g3578C>A) and one novel xframeshift mutation (g3383delA) was observed in exon 3. One silent variation as substitution of G>A (G870A [rs9344]) was observed in exon 4. In addition C>A transition (rs7177) was observed in untranslated region (3''UTR) of CCND1. CDK4 germline mutation analysis showed 2 missense mutations (g5051G>C and g5095G>C) in exon 2 and 2 missense mutations in exon 5 (g5906C>A and g5907C>G). One novel frameshift mutation as a result of three nucleotide deletion (g7321_23delTGA) was observed in exon 7. Additionally 2 novel frameshift mutations as a result of insertion of nucleotide G (g7121_7122insG) and deletion of nucleotide G (g7143delG) were also observed in 3''UTR. mRNA expression of Rb pathway genes (RB1, CCND1 and CDK4) was studied in 72 head and neck tumors samples and adjacent un-involved tissues (control), using real-time PCR. A statistically significant (p<0.001) down-regulation of RB1 was observed in tumor samples compared to control samples. CCND1 (p<0.004) and CDK4 (p<0.02) showed over expression in tumor samples versus control samples. Immunohistochemical analysis of RB1, CCND1 and CDK4 suggested 67% tumors exhibited down-regulated expression of RB1, 62% of tumors showed up-regulated CCND1 expression and 54% of tumors showed up-regulated expression of CDK4. Epigenetic analysis was performed to check methylation status of RB1 promoter region, through methylation specific PCR (MSP). Methylation analysis of RB1 promoter region after sequence analysis revealed that 3% of CpG island was methylated in patients as compared to controls (un-involved tissues). In vitro characterization of selected RB1 mutations was carried out using cancer cell lines, revealed that RB1 mutations in important functional domains result in abnormally reduced protein stability causing defects in RB1 functioning which may play a role in tumor proliferation. Results indicate that loss of RB1 function makes the condition selectively advantageous in tumor aggression which highlights the importance of RB1 in tumor protection. Results from this study also suggest that cells expressing mutant RB1Arg455Ser-V5 and RB1Ile835Asn-V5 may get a survival advantage against doxorubicin induced cytotoxicity, which may enhance drug resistance.

IMPLEMENTASI STRATEGI HAFALAN AL-QUR'AN TERHADAP HASIL BELAJAR SISWA DI MADRASAH ALIYAH AL-HUDA KOTA GORONTALO

  Abstrak Artikel ini bertujuan untuk mengungkapkan Implementasi Hafalan Al-Qur’an terhadap Hasil Belajar Siswa di Madrasah Aliyah Al-Huda Kota Gorontalo bahwasanya mengikuti program tahfiz peserta didik senantiasa terlatih untuk mempermantap bacaan Al-Qur’an mengetahui penerapan ilmu tajwid yang baik dan benar. Strategi implementasi hafalan Al-Qur’an bagi peserta didik ialah untuk menuntun siswa agar mudah dalam mengahafal Al-Qur’an, peserta didik yang ikut serta dalam program tahfiz rata-rata memiliki kemampuan yang baik dalam membaca Al-Qur’an yang baik dan benar berdasarkan kaidah tajwid, serta memiliki hafalan ayat-ayat Al-Qur’an yang nantinya menjadi bekal dasar untuk memperdalam ilmu pengetahuan agama. Implikasi penelitian ini adalah guru tahfiz di Madrasah Aliyah Al-Huda Kota Gorontalo terus meningkatkan sumber daya dalam strategi hafalan Al-Qur’an untuk menunjang proses kegiatan pembelajaran khususnya mata pelajaran yang membidangi Al-Qur’an seperti Al-Qur’an Hadist, Ilmu Al-Qur’an Tajwid dan Tahfiz. Kepada semua komponen yang ada di Madrasah Aliyah Al-Huda Kota Gorontalo yakni pihak yayasan, komite, orang tua dan kepala sekolah, diharapkan dapat bekerjasama untuk kelancaran program pendidikan terutama pada program tahfiz Al-Qur’an. Shell Side Direct Expansion Evaporation of Ammonia on Plain Tube Bundle With Inlet Quality and Exit Superheat Effects

Shell and tube heat exchangers are widely used for various industrial refrigeration applications. There is no data available in open literature on the direct expansion evaporation characteristics of refrigerants on the shell side of tube bundle. With the current Ozone Depletion and Global Warming issues it is critical to develop systems with low charge refrigerants especially with natural refrigerants such as ammonia which has zero Ozone Depletion Potential (ODP) and zero Global Warming Potential (GWP). A comprehensive literature survey has been presented to summarize previous single phase correlations for estimating heat transfer coefficient inside circular tubes and studies performed with ammonia boiling outside single tube and bundles and the correlations thereof developed. Considerable amount of work has been performed and correlations developed for estimating heat transfer and pressure drop in smooth circular tubes for single phase flow. There are limited studies available on pool boiling and spray evaporation of ammonia outside single tube and bundle. As of now, there are no studies available in the open literature on the subject of direct expansion evaporation of refrigerants. The basic purpose of the present study is to quantify the effects of various parameters such as saturation temperature, heat flux, inlet quality and degree of exit superheat on the heat transfer performance for ammonia boiling on the shell side of tube bundle under direct expansion mode, and to develop empirical correlations. Overall performance of the heat exchanger and performance of individual passes are also studied. The design and details of the single phase and two phase experimental setup, instrumentations, data acquisition system and test heat exchanger are described. Single phase experiments were performed to develop single phase correlations for specific geometry and flow conditions for estimating heat transfer coefficient inside tube bundle using simple Wilson plot and modified Wilson plot method. Prandtl number was varied from 10 to 20, with Reynolds number ranging from 3000 to 15000. The correlations developed are compared vii with existing single phase correlations and were found to be in good agreement with the previous studies. Two phase experiments were performed at steady state conditions with ammonia boiling on the shell side of heat exchanger, while water/glycol solution flowing in the tubes. Saturation temperature was varied from -1.7 to -20 , inlet quality was varied from 0 to 0.3, and heat flux was varied between 5 and 45 , with exit degree of superheat ranging from 2 to 10 . Heat transfer coefficient was observed to increase with saturation temperature and heat flux and decrease with exit superheat (quality) because of the diminishing effect of boiling and dominance of single-phase sensible regime. The effects of inlet quality on the average bundle performance were found to be negligible. Heat transfer coefficient slightly decreased or remained unchanged with inlet quality. The quality effects were found to be more dominant at higher saturation temperature than at lower saturation temperature. Bundle effect was observed between the passes which was a function of saturation temperature, heat flux and exit degree superheat. The present data suggests that bundle effect is less significant at low heat flux, with lower degree of exit superheat. The variations in heat transfer coefficients of individual passes are usually smaller under these circumstances. At higher exit degree superheat and heat flux the upper tubes experienced greater dry out which resulted in a sharp drop in their performance. The trend was similar across the saturation temperature range. The effects of exit degree of superheat became more dominant on the bundle performance over heat flux when ammonia was subjected to vapor quality at the inlet of the evaporator. With increase in the inlet quality the performance of lower three passes converge. Two phase pressure drop under direct expansion mode was found to be sufficiently small. The effects of saturation temperature and inlet quality on pressure drop were negligible. Pressure drop marginally increased with decrease in the degree of exit superheat and with increase in the heat flux.
