Lactuca sativa (lettuce) in addition to being an important vegetable has been traditionally used for therapeutic purpose due to its valuable secondary metabolites. Rol genes of Agrobacterium are known to increase the synthesis of these versatile phytochemicals. This study was designed to enhance antioxidant and antidiabetic potential of lettuce by transformation using two constructs: harboring rol ABC genes and rol C gene separately. Transgene integration and expression was confirmed through PCR and semi-quantitative RT-PCR, respectively. Various in-vitro, in-vivo and cell line based assays were performed for comparison of transformed, untransformed and field grown plants of lettuce. Results showed 60% or 100% increase in total phenolic content for plants containing rol ABC genes or rol C gene, respectively compared to untransformed control plants. For total flavonoid content, 60% increase was noticed in both types of transformed plants. In contrast to control, total antioxidant capacity of plants transformed with rol ABC genes and rol C gene increased up till 110% and 70%, respectively. Total reducing power assay showed up till 125% higher activity in both rol ABC and rol C transformed plants compared to control. Results of lipid peroxidation, DPPH and DNA damage assays showed 50%, 80% and 60% increase of antioxidant activity for rol ABC and rol C plants, respectively, compared to untransformed plants. The extracts of both genes showed similar but significant enhancement behavior in hot plate analgesic and carrageenan induced hind paw edema test in rats. Up to 2 fold increase in activity was detected with 81% and 73% enhancement for the rol C and rol ABC genes respectively. These results suggest the dual inhibition nature and strong positive correlation between analgesic and anti-inflammatory activity. The viiitransformants of rol C gene exhibited prominent antidepressant activity with 190% increase as compared to untransformed plants. As for as the anticoagulant activity is concerned, an 80% increase in activity was found for the rol C extracts only. No enhancement in antidepressant and anti-coagulant activity was detected in the rol ABC gene extracts. Additionally, current findings show that seed extracts have the highest antidepressant activity. In-vitro bioactivity testing showed that rol ABC and rol C significantly improved the ability of L. sativa for its antidiabetic potential. In α-glucosidase enzyme inhibition assay, the transfomants of rol C showed 64% increase while rol ABC transfomants showed 100% increase in activity. In the comparative study of transformed and untransformed plants, the transfomants of rol C showed 84% increase while rol ABC transfomants showed 85% increase in their ability to inhibit DPP-4 enzyme. In case of GLP-1 secretion, the transfomants of rol C showed the greatest increase in GLP-1 level with 559% increase, on the other hand rol ABC transfomants showed 405% increase compared to the untransformed control plants. In the untargeted metabolomics study (LC-QTOF-MS), out of 5333 metabolites profiled and quantified, the levels of 3637 and 1792 significantly differed in rol ABC and rol C, respectively compared to untransformed plants. We identified 76 lettuce metabolites (including sesquiterpene lactones (12), non-phenolic (24), and phenolic (40) compounds) some of which changed by several folds. For example, the secondary metabolite ferulic acid methyl ester increased up to 115 fold. In conclusion, we propose that rol gene transformation significantly alters the metabolome of L. sativa and enhances its phytochemical, antioxidant, analgesic, anti-inflammatory and antidiabetic properties.

مولانا سیماب اکبر آبادی کے فرزند اور رسالہ ’’شاعر‘‘ کے مدیر، اعجاز صدیقی پر ۹؍ فروری ۱۹۷۸؁ء کو دل کا دورہ پڑا اور وہ اسی روز اپنے مالک حقیقی سے جاملے۔
ایک عرصہ سے مختلف امراض کی وجہ سے اعجاز صاحب کی صحت خراب ہوچکی تھی، کئی بار اسپتال میں داخل کئے گئے، اچھے اچھے ڈاکٹروں نے بڑی توجہ اور شفقت سے علاج کیا، مگر بقول شاعر:
الٹی ہوگئیں سب تدبیریں کچھ نہ دوا نے کام کیا
دیکھا ، اس بیماری دل نے آخر کام تمام کیا
اعجاز صدیقی فروری ۱۹۵۱؁ء میں اپنے وطن آگرہ سے بمبئی آئے اور یہیں انھوں نے مستقل سکونت اختیار کرلی، ان کا رسالہ ’’شاعر‘‘ (ماہنامہ) جو پہلے آگرے سے نکلتا تھا، اسی سال سے بمبئی سے شائع ہونے لگا، وہ اس کے معیار کو بلند رکھنے میں انتھک، کوشش کرتے تھے، اردو سے پر خلوص محبت اور اپنے قارئین کو صاف ستھرا ادب پیش کرنے کی کوشش، ان کی زندگی کے دو ایسے نمایاں پہلو تھے کہ جن کی وجہ سے اردو کے اچھے اور اہم لکھنے والوں اور شعراء کا انھیں غیر معمولی تعاون حاصل ہوتا رہا، جس کے سہارے وہ ’’شاعر‘‘ کے بڑے ضخیم خصوصی نمبر نکال سکے، ان میں کرشن چند نمبر، ناولٹ نمبر، افسانہ اور ڈرامہ نمبر اور آخری میں ہم عصر اردو ادب نمبر ہماری زبان و ادب میں قابل قدر اضافہ ہیں، حقیقت یہ ہے کہ مسلسل علالت گرتی ہوئی صحت اور محدود مسائل کے ساتھ ایسے ضخیم اور اچھے نمبر شائع کرنا، بڑی جرأت کا کام تھا، بلاشبہ اعجاز صاحب غیر معمولی قوت ارادی کے حامل تھے۔

سید محمد محسن اور ان کا منظوم ترجمۂ قرآن: ایک تنقیدی مطالعہ

Sayyid Muḥammad Moḥsin was Lucknow based poet. The biographers, though, seem unaware of his life and poetic profile. He did interpretative translation of the Holy Qurʼān in Urdu in Mathnavī form with the title of ‘Manẓūm Urdu Tarjama’. This translation was published from Lucknow in 1986. The translation in poetic form asks for technicalities which this work lacked perhaps. This article surfaces errors which were identified in areas of poetic exposition, concept formation and stylistic coherence in the translation work.

Novel Hybrid Models Based on Hesitant Fuzzy Sets

Hesitant fuzzy set theory is an impressive tool, to express the vagueness and ambiguity derive from experts, to analyze the characteristics of objects, and to determine the membership functions for each criterion domain. Due to the unpredictability of real world problems, and the limitation of human’s observations, it is very challenging to apply only a particular or specific uncertainty method to deal with complex problems. One can approach to more impressive and effective models to handle decision-making problems, by relating or associating the advantages of several other distinct theories of uncertainty. Hesitant fuzzy sets and soft sets are considered more efficient to deal the uncertain conditions. By combining these theories with several other useful models, novel hybrid models can be developed for decision-making problems. The aim of this research is to construct novel hybrid models based on hesitant fuzzy sets, because the use of hesitant fuzzy computations makes the decision-makers assessments more feasible and significant in decision-making. The proposed hybrid models are named as, hesitant N-soft sets, hesitant fuzzy N-soft sets, interval-valued hesitant fuzzy N-soft sets, m−polar hesitant fuzzy sets and hesitant m−polar fuzzy sets. These models are obtained by the hybridization of hesitant fuzzy sets with N-soft sets and m−polar fuzzy sets, which are more flexible and expressive for modeling and processing of vague information under hesitancy. These models provide us information about the occurrence of ratings or grades and enable us to tackle multi-polar information with hesitancy. Various properties of hybrid models are investigated with different methods of their construction. Intrinsic operations are highlighted and explored with several new concepts including weak, top weak and bottom weak complements. The concept of decision-making methods named as choice value, L-choice value, TOPSIS and ELECTRE-I, is explored by applying them on proposed models. Potential applications related to real life situations are presented, that illustrate the procedure of the proposed decision-making methods. Efficient algorithms are developed to solve the decision-making problems based on hybrid models.
