Influenza viruses, in seasonal and pandemic forms, cause the most severe respiratory infections in humans. The currently licensed, egg-based vaccines fail to meet the global vaccine demand during a pandemic. Moreover, there is a constant need to reformulate these vaccines on annual basis according to the circulating viral strains. In order to develop efficient prophylactic measures, alternate vaccine strategies and production platforms need to be exploited. Subunit vaccines, like recombinant hemagglutinin, offer an alternative over conventional vaccines. Current study was designed to clone and express soluble influenza hemagglutinin (HA) of H9 and/or H7, which could be used as a vaccine candidate. To achieve high level of secretory expression and stabilized native trimer structure, HA was assembled with sequence for the tissue plasminogen activator signal peptide (tPA-SP) and foldon domain of bacteriophage T4 fibritin (FT) respectively by employing splicing by overlap extension amplification. The amplified product was cloned in pCMVR-8kb having human cytomegalovirus (CMV) immediate-early enhancer and promoter for transient expression of HA in 293F cells. SDS PAGE analysis confirmed a band of ≃ 70 kDa corresponding to H9HA and H7HA which was further confirmed by Western blot analysis and purified by Ni-NTA agarose column. The purified recombinant protein was found biologically active. This expression and purification process can be scaled up easily and recombinant protein can be produced rapidly which can then be used for further studies on virus-host interactions, viral pathogenesis and development of vaccines.The failure of currently available antiviral agents, due to high mutation rate of the influenza virus, calls for employing rigorous strategies to develop safe and potent inhibitory strategies against the virus. Nanoparticles have been studied extensively as potential antimicrobials, their applications recently extended to development of antivirals. In the present study, silver nanoparticles were successfully developed by green synthetic approach using bark extract of Cinnamomum cassia (Cinnamon) and leaves extract of Thymus vulgaris (Thyme). The synthesized nanoparticles were characterized using UV-Vis absorption spectroscopy, scanning electron microscopy (SEM) and fourier transform infrared spectroscopy (FTIR). Plant extracts (Cinnamon and Thyme) and their corresponding nanoparticles were tested against avian influenza virus subtype H7N3 in Vero cells and the viability of cells was determined by tetrazolium dye (MTT) assay. The silver nanoparticles derived from plant extracts enhanced the antiviral activity and were found to be effective in both treatments, when incubated with the virus prior to infection and introduced to cells after infection. Moreover, the safety profile of the extract and the nanoparticles showed that they were non-toxic to Vero cells even at a concentration of 500 μg/ml. The biosynthesized nanoparticles may, hence, be a promising approach to provide treatment against influenza virus infections. Further research on characterization and mechanism of action of nanoparticles is required to develop better antiviral therapeutics for highly mutating viruses.

۔منکرین آخرت و رسالت سے تخاطب اور استفہامی اسلوب

 ایمان بالآخرة یا عقیدہ آخرت ارکانِ ایمان میں سے ایک رکن ہے ، قرآن مجید میں منکرین آخرت و رسالت کو اس اہم حقیقیت کیلئے دلائل دیتے ہوئے اللہ تعالیٰ نے استفہامی اسلوب کو اختیار کیا ہے،ذیل میں عقیدہ آخرت کے بارے میں آیات استفہام پیش کی جا رہی ہیں:

۱-سورة المآئدہ میں ارشاد باری تعالیٰ ہے:

"يَوْمَ يَجْمَعُ ٱللَّهُ ٱلرُّسُلَ فَيَقُولُ مَاذَآ أُجَبْتُمْ قَالُواْ لاَ عِلْمَ لَنَآ إِنَّكَ أَنتَ عَلاَّمُ ٱلْغُيُوبِ"۔[[1]]

"جس دن اللہ جمع کرے گا سب پیغمبروں کو پھر کہے گا تم کو کیا جواب ملا تھا؟ وہ کہیں گے ہم کو خبر نہیں تو ہی چھپی باتوں کو جاننے والا ہے"۔

 ابن جریر طبریؒ اس آیت کی تفسیر سے متعلق رقمطراز ہیں:

“'وہ دن کیسا ہولناک ہوگا جس روز اللہ تعالیٰ تمام پیغمبروں کو مع ان کی امتوں کے جمع کریں گے اس دن اول سے آخر تک پیدا ہونے والے سب انسان اس میدان میں حاضر ہونگے اور سب سے انکے عمر بھر کے اعمال کا حساب لیا جائے گا اللہ تعالیٰ فرمائیں گے کہ اے نبیوں جب تم نے اپنی قوم کو دین حق کی طرف بلایا تھا تو انہوں نے کیا جواب دیا تھا؟اس سوال کے مخاطب اگرچہ انبیاء کرام ہونگے مگر سنانا امتوں کو ھوگا انبیاء کرام عرض کریں گے کہ اے اللہ! بظاہر تو ہم کو جو معلوم ہیں وہ بیان کر دیں گے مگر انکے دل میں جو کچھ ہے اسکی ہمیں خبر نہیں اس کو آپ ہی جانتے ہیں کیونکہ بیشک آپ پوشیدہ باتوں کے پورے جاننے والے ہیں ہم لاعلم ہیں اور انبیاء کرام صرف اپنی...

EFFECTIVENESS OF COMMUNITY SCHOOL NETWORKING FOR CHILDREN WITH AUTISM: PERSPECTIVES OF TEACHERS AND SCHOOL ADMINISTRATORS

Objective: In order to provide equal educational opportunities, community school networking is an emerging trend to facilitate inclusion of children with mild-moderate Autism. This quantitative research aims to investigate the effectiveness of community networking for children with Autism from Pakistani lower socio-economic stratum of society. Study Design: Qualitative Research Design Study Settings and Participants: Six mild-moderate autistic children were enrolled in three mainstreams schools and a liaison between these schools, and a rehabilitation center located in the same area was created to facilitate inclusion. The researchers interviewed six teachers from mainstream schools, three school administrators and one administrator of rehabilitation regarding the effectiveness of community school networking for children with Autism after eight months of this collaboration. Data Collection Tool: Data were obtained through semi-structured interviews Results: All participating administrators and teachers underscored the changes in social and behavioral patterns of autistic children which included an imitation of positive behaviors from peers, acceptance, and awareness as strengths of community school networking model. However, major challenges faced were unacceptability from parents of normal children, learning differences, curriculum modifications, time constraints and dependency on the resource teacher. The administrators and teachers recommended that creating awareness programs for parents of normal children, curriculum modifications and in-house psychologists can further facilitate inclusion of children with disabilities. Conclusion: It was concluded that community school networking model can assist inclusive education and encourage engagement for all children, including those who are autistic.

Feminism and Beyond: A Post-Structuralist Reflection on the Image and Identity of Pakistani Women

This qualitative-cum-descriptive piece of research aims at highlighting the popular conventions of (re)presentation with which women of Pakistani society have been portrayed and given voice to in the mainstream indigenous feminist discourse. Since fact and fiction both constitute the making of such conventions, therefore, the study focuses on the fictional and theoretical aspects of Pakistani women's identity and image in the contemporary feminist debate within Pakistan and seeks to see what brand of feminism is born out of it. The literary and imaginative conventions are critiqued side by side with the theoretical assumptions as espoused by indigenous feminism. What informs such analysis is a constant sense of inadequacy in terms of giving voice and representations to Pakistani women in matters of their identity and status. The insight drawn out of such analysis is the need to revise the perspectives and patterns with which to analyze and understand the position and image of Pakistani women as well as to incorporate and include multiple and diverse perspectives in order to appreciate and understand the complex yet lived realities of women in their peculiar socio-cultural milieu. Given the diversity and multiplicity of views and voices, the principal operating assumption of my work is to suggest patterns (of behavior and thought) which could "unify" and "include" rather than "alienate" and "exclude". My work attempts to see and seek the identity and image of Pakistani women to be reformulated and remolded by encompassing a wide range of women with their unique and shared perceptions about their own identity in particular and other women in general.
