The effective control and eradication of brucellosis in developing countries can be achieved by quick and accurate diagnosis and effective vaccination. Protective efficacy of existing vaccines as well as diagnostic accuracies of various serological tests have not been thoroughly studied and verified in buffaloes, the prime dairy animal of South Asia. In the Present studies Brucella abortus strain 19 (S19) and strain RB51 (SRB51) vaccines were tested for inducing humoral and cell mediated immune responses in riverine buffaloes of various age groups. The protective efficacy of these vaccines against abortions was also evaluated by challenge protection studies. Protein antigen of Brucella abortus vaccinal strains involved in protection was identified by western blot analysis. Moreover an indirect enzyme linked immunosorbent assay (I-ELISA) for detection of anti-brucella antibodies in buffaloes was developed. The diagnostic sensitivity and specificity of developed test was compared with commercial ELISA test. Agreement between the two tests for negative sera was 100% and for positive sera it was 78%. In the vaccination study using S19 the peak antibody titres (P<0.05) were observed 30 days post vaccination using I-ELISA and Rose Bengal plate test (RBPT). The mean ELISA titre persisted for 3 to 4 months in the vaccinated buffaloes but waned quickly in calves than adults and heifers. The agglutinating antibodies detected by RBPT declined below the analytical threshold earlier than ELISA. Vaccinated and non vaccinated control animals were challenged subcutaneously with 5× 109 cfu of virulent Brucella abortus during 1st gestation after vaccination. Significantly lower (P<0.1) abortions were observed in vaccinated animals. In SRB51 vaccinates significant increase (P<0.05) in the humoral immune responses were detected in all the three age groups by an I-ELISA using acetone killed SRB51 antigen 30 days post vaccination. However, all SRB51 vaccinates were detected negative by RBPT. The vaccinated and control animals, when challenged with locally isolated virulent biovar 1 between 6-7 months of gestation, revealed non-significant (P>0.1) differences between the probabilities of abortion occurrence. However, the percentage of abortion occurrence in vaccinated animals (44%) was lower than the non-vaccinated buffaloes (78%). Lymphocyte proliferation assays detected moderately high and consistent proliferative responses in all of the S19 vaccinated adults, heifers and calves after 6 weeks of immunization. The lymphocyte proliferative responses of SRB51 vaccinate were low and inconsistent and only 77% of the immunized animals exhibited positive proliferative responses. Western immunoblot analysis using antisera raised against all three B. abortus strains in buffaloes indicated a near similar pattern of immune-reactive OMPs in S19 and field strain. Two OMPs of molecular weight 37-38 and 19 kDa were immuno-reactive in all strains in buffaloes. Two distinct proteins of molecular weights of 190.5 and 151.3 kDa were identified in field strain but not in both of the vaccinal strains of B. abortus.

کیونکہ ام سلمہؓ کے میکہ والے مزاحم ہوئے اور انہوں نے اسے روک لیا ،ام سلمہ ؓ اپنے گھر واپس آئیں تو ان کے شوہر ابو سلمہ ؓ کے گھر والوں نے اس سے بچہ چھین لیا ، جس کا نام سلمہ تھا ۔وہ اپنے بچہ سے بچھڑ گئیں ۔ وہ روزانہ گھر سے نکل پڑتی اور ابطع میں بیٹھ کر رویا کرتی تھی ۔ سات آٹھ دن بعد ابطع سے ان کے خاندان کا ایک شخص نکلا ۔ اس نے ام سلمہ کوروتے دیکھا تو اس کا دل جذبہ ترحم سے بھر آیا ۔ گھر آکر لوگوں کو جمع کر کے کہا کہ اس پر ظلم کیوں کرتے ہو ؟ اسے جانے دو : اور ساتھ ہی اسے بچہ دے دو ۔ لوگوں نے بات مان لی اور بچہ ام سلمہ ؓ کے سپرد کر دیا اور مدینہ جانے کی بھی اجازت دے دی ۔ وہ تنہا سفر کر رہی تھی ۔ تنعیم کے مقام پر کلید بردار کعبہ عثمان بن طلحہ جو ابھی مشرف بہ اسلام نہیں ہوئے تھے ، انہوں نے سیدہ سلمہ ؓ سے پوچھا ، کہاں کا ارادہ ہے ؟ انہوں نے کہا ۔مدینہ ۔ انہوں نے پوچھا : کوئی ساتھ ہے ؟ آپ نے فرمایا ’’ خدا اور یہ بچہ ‘‘ ۔انہوں نے اونٹ پر سوار کر لیا اور خود مہار پکڑ لی ۔ جب قباء کی آبادی پر نظر پڑی تو عثمان نے کہا ’’ اب تم اپنے شوہر کے پاس چلی جائو، وہ یہیں قیام پذیر ہیں ‘‘ ۔ سیدہ سلمہ ؓ قباء کو اور عثمان مکہ کو روانہ...

الاصابہ فی تمییز الصحابہ میں نماز و روزے سے متعلقہ روایاتِ اَحکام کا تحقیقی جائزہ Research Review of the Traditions of Rulings on Prayer and Fasting in Al-Aṣābah Fī Tamīz al-Ṣaḥābah

The book “Al-Aṣābah Fī Tamīz al-Ṣaḥābah” is a famous book of Ibn Ḥajar ‘Asqalānī on the life and contribution of the Companions. This book is quoted as a reference to decide the status of companions. It would not be wrong to call it the encyclopedia of the companions of the Holy Prophetﷺ. Apart from its status as a reference book regarding the companions this book encompasses the Seerah of Holy Prophet in a detailed and well composed manner. The distinction of the book is not hidden from the experts of science and research. In the book and other similar books, the life of the companions was discussed and the biography of the Prophet ﷺwas also described, but another aspect that remained unexplored in these books is the virtues and commandments mentioned in it. There are also narrations about virtues. In this paper the researcher has evaluated the Hadiths related to prayer and fasting that are narrated in Al-Aṣābah Fī Tamīz al-Ṣaḥābah. The evaluation comprises of authenticity of narrators and text, narrators’ mutual association or disassociation and the final decree about the authenticity of a hadith in the light of all available evidences.

Combining Ability Effects of Yield and Salinity Tolerance Related Traits in Brassica Napus L.

Among the numerous abiotic stresses restricting the crop production, salinity stress is the most significant one. Estimation of genetic variability and development of suitable selection criterion are fundamentals for development of salinity tolerant types of crop plants. Sixty accessions of Brassica napus were evaluated for salinity tolerance using hydroponics technique. Six sensitive and ten tolerant lines were selected and crosses were attempted in line × tester fashion. The resultant F1 crosses and their parents were evaluated at seedling stage using hydroponic culture technique and at maturity stage in a pot experiment. Data were recorded on various seedling (root length, shoot length, fresh root weight, fresh shoot weight, dry root weight, dry shoot weight, proline content, Na+/K+ ratio, osmotic potential) and morphological traits (days to 50% flowering, days to 50% maturity, plant height, number of primary branches, number of silique per plant, seeds per silique, oil content, thousand seed weight, yield per plant). Recorded data were analyzed to estimate the genetic variability, general and specific combining ability effects, heterosis, gene action and association among traits. Results showed that genetic variability existed in present breeding material that may be used in development of high yielding salinity tolerant Brassica napus. Combining ability analysis revealed that lines ZMR-2 and R-3 and testers Faisal and Shirlaee for seedling traits and lines R-3 and 23627 and tester Faisal for morphological traits were best general combiners for most of the traits under normal and salinity stress conditions. Crosses R-3×Laclone, RBJ-8007×Laclone and ZMR-1×ZMR-3 were best specific combiners for most of the traits under all treatments. Cross RBJ-8007×Laclone also had significant and positive mid parent, better parent and commercial heterosis for most of the traits under control and salinity stress treatments. Correlation and path analysis revealed that number of silique per plant, oil contents, days to 50% flowering and days to 50% maturity may be used as selection criteria for development of salinity tolerant types with higher yield as they showed significant association with yield per plant under salt stress conditions.
