Organisms have naturally build defense mechanisms against infections and diseases caused by pathogens. This system known as immune system comprises of innate immunity and adaptive immunity. Innate immunity is non specific and produces a quick response to the invading pathogens. Macrophages play central role in innate immunity, also called natural immunity. The other part of immunity is rather specific and it takes some time to show its response, this type of immunity is called adaptive immunity. Adaptive immunity can be either T cell mediated or B cell mediated in response to intra cellular or extra cellular pathogens respectively. When it is T cell mediated it will lead to the secretion of cytokines including interferon along with other substances, which help to fight intra cellular pathogens especially viruses by activating natural killer cells and macrophages. B cell mediated immune system leads to production of antibodies in response to extra cellular pathogens in blood or other body fluids. The whole immune system including innate immunity and adaptive immunity mediated by both T cell and B cell are not separate from each other but rather linked with each other and activating each other against foreign invaders. For example macrophages activate T cells, T cells activate macrophages, B cells and other T cells. Purpose of present study is to know about the adaptive immune system of two farm fishes, rainbow trout and common carp which are main source of protein in Pakistan. This study will provide a guide line to improve the immune system of farm fishes so that they can be resistant to diseases. This will ultimately benefit the human beings as they will get more Omega 3 rich protein, which is good for the over all health of human beings. In the present work both cellular and humoral aspects of adaptive immune system mediated by T and B lymphocytes were studied. Two teleost fishes rainbow trout Oncorhynchus mykiss (Scottish farm fish) and common carp Cyprinus carpio (Pakistani farm fish) were taken as experimental animals. Immune genes which regulate interferon production as a cellular immune response to viruses or other intracellular pathogens, werestudied in rainbow trout Oncorhynchus mykiss, B cell mediated immune system leading to production of antibodies was studied in common carp Cyprinus carpio. It was found that production of interferon is regulated by immune genes known as interferon regulatory factor (IRF) genes. These IRF genes are 10 in number and are named as IRF1, IRF2, IRF3, IRF4, IRF5, IRF6, IRF7, IRF8, IRF9& IRF10 and altogether form IRF family. IRF genes have been sequenced so far in many species including human beings. In rainbow trout IRF1, 2, 3&7 have been sequenced till to date. In present project work was done on remaining members of the IRF family. During the project two full length genes IRF 4& IRF8 were sequenced along with partial sequences of IRF 5& IRF10. It was found that certain toxicants like Poly I: C trigger IRF genes to produce positive immune response by secreting cytokines like interferon. Poly I: C (Polyinosinic: polycytidylic acid) is a synthetic double-stranded RNA that is used experimentally to model viral infections in vivo. IRF4 gene was expressed in rainbow trout spleen cells treated with Poly I: C for 4 hours. IRF 8 gene was expressed in spleen and head kidney cells. After multiple alignments of genes it was found that the gene structure of IRF4&8 genes found in rainbow trout had close structural resemblance with IRF4&8 genes found in other fish like zebra fish or even in mammals like human being and mouse. IRF4 &IRF8 play an important role in maturation of B and T lymphocytes. The other aspect of adaptive immune system, B cell mediated or humoral immunity was studied in common carp Cyprinus carpio. It was found that humoral antibodies are detected in serum of fish after secondary immunization with particulate antigen SRBC (sheep red blood cells). Proliferative response in lymphoid tissues especially in spleen and kidney was high while there was less response in liver. HGG localization in these tissues was studied by using fluorescent labeled antibodies against HGG. Farm fish in Pakistan are built in agricultural lands and so these fish are in contact with the pesticides used on crops through water run off to these fish farms. It was found during experimental work that toxicants like pesticides affect the immune system of common carp by suppressing the production of antibodies. Histological studies were also conducted which showed that besides affecting immune system of fish, these pesticides also have harmful affects on lymphoid tissues of fish and also on immune system of fish.

مولانا شبلی نعمانی ؒ کی ایک ہی جسمانی یادگار باقی رہ گئی تھی وہ بھی مٹ گئی، یعنی ان کے اکلوتے صاحبزادہ حامد نعمانی صاحب نے ۶۲؍ برس کی عمر میں ۲؍ ربیع الاول ۱۳۶۱؁ھ مطابق ۲۰؍ مارچ ۱۹۴۲؁ء کی شب کو جونپور میں دفعتہ وفات پائی، وہ کئی برس سے مرض قلب میں گرفتار تھے، علاجوں کے سہارے سے چلتے پھرتے تھے، مگر اندر سے کھوکھلے ہوچکے تھے، ۱۹؍ مارچ کو وہ ایک ضرورت سے جونپور گئے تھے، شام کو پہنچے، اپنا کام کیا، رات کو ۳ بجے کے قریب درد دل کا دورہ ہوا،ان کے میزبان دوست ان کے کراہنے کی آواز سن کر ان کے پاس آئے، مرحوم نے کہا کہ مجھے ذرا سہارا دے کر بٹھا دو، انہوں نے اپنے سینے کے سہارے سے بٹھا دیا، اسی کے ساتھ مرحوم نے ان کو السلام علیکم کہا، اور آخری سانس لے کر نامعلوم سفر کی منزل پر روانہ ہوگئے، انا ﷲ و انا الیہ راجعون، ۲۰ کی صبح کو لاش کار سے اعظم گڑھ آئی، اور شبلی منزل میں باپ کے پہلو میں بیٹے کو ہمیشہ کے لیے سلا دیا گیا۔

ANALISIS KESULITAN MEMBACA PADA SISWA KELAS V SDI WAIROTANG

Tujuan dari penelitian ini adalah untuk mendeskripsikan kesulitan membaca yang di alami siswa kelas V SDI Wairotang dan untuk mengetahui faktor-faktor yang menghambat siswa dalam  kesulitan membaca. Penelitian ini menggunakan pendekatan kualitatif dengan jenis metode penelitian deskriptif kualitatif. Sumber data dalam penelitian ini adalah guru dan siswa SDI Wairotang yang berkesulitan membaca. Teknik pengumpulan data menggunakan teknik wawancara dan dokumentasi. Teknik analisis data menggunakan Miles and Huberman yaitu mereduksi data, menyajikan data dan penarikan kesimpulan. Hasil penelitian yang menunjukkan kesulitan yang dialami siswa adalah membaca belum lancar, mengeja, dan penggunaan  tanda baca kurang tepat dan faktor-faktor yang menghambat siswa dalam membaca yakni kurangnya minat belajar membaca serta kurangnya dukungan keluarga dalam belajar membaca. Berdasarkan hasil penelitian yang dilakukan maka dapat disimpulan bahwa kesulitan-kesulitan siswa dalam membaca dan faktor penghambat dalam membaca di kelas V SDI Wairotang yaitu: belum  lancar  membaca, mengeja dan penggunaan tanda baca yang kurang tepat serta kurangnya minat belajar membaca dan kurangnya dukungan keluarga dalam belajar membaca.  

Biochemical Changes in Protein and Lipid Fractions of Ultra-High Temperature Treated Milk and Dairy Drinks at Different Stages of Processing and Storage

This study is comprised of five experiments, in the first experiment chemical changes taking place in lipid fraction of pasteurized milk was studied. Raw milk from the same source was at once cooled down to chilling temperature and in second instance it was put on the lab shelf for two hours at ambient temperature and then chilled to same temperature, followed by pasteurization and chilled storage. In the second experiment, samples of UHT milk were tested for chemical characteristics and lipid oxidation. In third experiment, impact of UHT treatment (142oC for 2 sec) and 90 days ambient storage (30-35oC) on protein and amino acid profile of UHT milk and sedimentation was investigated. In fourth experiment, samples of UHT Treated products were stored for 90 days. Transition in antioxidant capacity, vitamin A, E, fatty acid profile was studied. Transition in protein profile, Maillard reaction products and lipid oxidation of flavored UHT treated milk was investigated. In fifth experiment, Samples of mango flavored UHT milk 200 ml pack (475 Nos.) were purchased from the market and stored at ambient temperature for the duration of 90 days. Proteolysis, hydroxyl methyl furfural furosine, Nε-carboxymethyl lysine, FAP, FFA, PV and sensory characteristics were studied at 0, 45 and 90 days of storage. It was observed that chemical characteristics and storage stability of raw milk in the second instance was different from the milk cooled at once. Therefore, it is recommended that milk collection system in the developing countries should be improved to avoid any delay in between milking and chilling of raw milk. The results of the second experiment disclosed that fatty acid and triglyceride profile were not affected till 60 days with a steady increase in lipase activity. Strong correlations between peroxide value and induction period were established. For the anticipation of oxidative stability of UHT milk, induction period can be used. After UHT treatment, β-lactoglobulin, β-lactalbumin, immunoglobulin and serum albumin decreased by 12.1%, 15.4%, 10.6% and 10.2% with no effect on casein fractions. Sedimentation was comprised of 14.9% αs1-casein, 3.42% αs2-casein, 9.88% β-casein, 2.27% κ-casein, 4.36% β-Lactoglobulin, 1.88% β-lactalbumin, 1.84% immunoglobulin and 0.61% Serum Albumin. Out of 80% casein and 20% whey, 38.5% and 8.7% became the part of sedimentation. Tyrosine content of flavored UHT milk increased during the storage. Tyrosine content of flavored UHT milk at 0, 45 and 90 days of storage were 3.5, 6.9 and 15.2 µg tyrosine/ml. HMF content of UHT milk at 0, 45 and 90 days of storage were 1.56, 4.18 and 7.61 (µmol/L). Furosine content of flavored UHT milk at 0, 45 and 90 days of storage intervals were 278, 392 and 561 mg/100g protein.
