Host-parasitoid interactions in insects offer a great opportunity to identify new genes and molecules responsible for the alteration of the host physiology. To circumvent the host immune response, female of the parasitic wasp injects a blend of secretions containing mainly ovarian proteins, calyx fluid, polydnaviruses and venom in their host insects. Crude venom extract containing a wide range of bioactive proteinaceous compounds inhibits the motility of larval hemocytes and prevents the formation of cell aggregates upon artificial envenomation in vitro. In the present research work, a host parasitoid relationship of ectoparasitic wasp, Bracon hebetor (Say) (Hymenoptera, Braconidae) and host insect, greater wax moth, Galleria mellonella (Lepidoptera, Pyralidae) was studied for comprehensive analysis of the venom glands Transcriptome of the wasp B. hebetor through different Bioinformatics tools for fishing out immunosuppressive genes which reveals the occurance of a number of novel genes with immune suppressive acitivities in the venom blend of the wasp suggesting their toxic/ insecticidal potential. Out of them, Venom acid phosphatase, Tryptase -2 and CTD nuclear envelope phosphatase were selected for more detailed studies on the basis of high sequence homology/identity. The selected contigs were partial sequences of the immunosuppressive genes and their 5’ ends were isolated by Rapid Amplification of cDNA Ends repair kit (RACER kit, Invitrogen) and then full-length genes were isolated by III RT module of Gene Racer kit. Isolated genes were cloned in pGEMTeasy vector in DH5-Alpha competent cells. Screening of blue white colonies of E. coli and colony PCR were also performed for the confirmation of inserts along with the plasmid extraction of the desired colony by Plasmid DNA mini prep. Cloned genes were sequenced, analysed and characterized by different bioinformatics tools i.e., Expasy translate tool, Clustal Omega and SignalP etc. Deduced amino acid sequences of immune suppressive genes were multialighned with other parasitoid species in the database which revealed significant homologies with other braconid species. Functional bioassays of the crude venom and recombinant proteins were performed through artificial microinjection techniques in which different concentrations of the crude venom and different combinations of the recombinant proteins were injected into the last instar larvae of the host and mortality data was recorded. The effects of parasitization by the wasp on cellular immune response of the host larvae were also investigated by following an approach of hemocytes count methodology. It may be concluded that B. hebetor venom and its recombinant proteins have significant impact on survival of the host larvae by inducing biochemical and physological alterations in the parasitized host insects. Analysis of the venom glands Transcriptome of the B. hebetor has shown the natural occurance of a number of novel venom genes with potential insecticidal activity involved significantly in the suppression of host immune response in the venom blend of the wasp which may be exploited further for indepth studies through integrated approaches of functional genomics and proteomics for the development of innovative plant protection strategies for sustainable control of diverse range of insect pests of Pyralidae of agriculture crops. Further, this preliminary research work has a significant contribution towards sustainable programmes for food security and food safety by controlling the insects through natural means and reducing the toxic residues of the synthetic chemicals in food chain supply system

تاریخِ ادبیاتِ سیالکوٹ ایک مطالعہ

          دریائے چناب جوچن اور آب کا مرکب ہے ۔نیز چندرا اور بھاگا دو دریاؤں کے ملنے سے وجود میں آیا ،ایک طرف تو (چن اور آب )حسن و خوب صورتی کا استعارا ہے تو دوسری طرف(چندرا بھاگا) معنوی لحاظ سے چاند کی قسمت لیے ہوئے ہے۔ اسی لئے اسے دریائے حسن و عشق کے نام سے بھی تعبیر کیا جاتا ہے اور سب جانتے ہیں کہ حسن اور عشق لازم و ملزوم ہیں ۔ یعنی جہاں حسن ہوتا ہے وہاں عشق ہوتا ہے ۔حسن میں جمالیات اور عشق میں جمالیاتی حظ اور ذوق ہوتا ہے ۔یہ حظ اور ذوق تخلیق کا باعث بنتے ہیں ۔شاعری اور ادب چوں کہ تخلیقی عمل ہے ،لہٰذا اس سارے سلسلے کو ایک دوسرے سے جدا نہیں کیا جاسکتا۔ جہاں دریائے چناب کا تعلق ہے تو اسے دانشورانِ عصر نے بہت سوچ سمجھ کر دریائے حسن و عشق کا نام دیا ہے ۔کیوں کہ اس کے کناروں پر آباد بستیوں میں بسنے والوں کو خالقِ کائنات نے حسنِ وافر عنایت فرمایا ہے جس نے عشق کی بنیاد رکھی۔یہی حسنِ وافراور عشق ادب کی تخلیق کا باعث ٹھہرا ہے ۔ سیالکوٹ بھی دریائے چناب کے کنارے ایک ایسی ہی بستی ہے جہاں دیگر شعبہ ہائے زندگی کے ساتھ ساتھ عکسِ زندگی اور نقدِ زندگی کی بھی فراوانی ہے۔اس دھرتی پر زندگی کا عکس پیش کرنے اور زندگی کو اپنی تنقیدی صلاحیتوں سے نکھارنے والی ایسی ایسی شخصیات نے جنم لیا جو قومی اور بین الاقوامی سطح پر اس دھرتی کی پہچان بنیں ۔ان معتبر شخصیات میں سے بہت سا ذکر آپ کو ’’تاریخِ ادبیاتِ سیالکوٹ‘‘ میں پڑھنے کو ملے گا لیکن تحقیق میں چوں کہ حرفِ آخر نہیں ہوتا اس لئے اس تحقیق میں بھی کئی نام ایسے رہ گئے ہیں جن کا ادبی مقام کسی سے ڈھکا چھپا نہیں۔اِس...

Biodiversity and Molecular Characterization of Spiders Fauna in Khyber Pakhtunkhwa Pakistan

Biodiversity is the source of energy and goals to maintain ecosystem for future. Present study of “Biodiversity and molecular characterization of Spider fauna of Khyber Pakhtunkhwa” was carried out in districts (Mardan, Swabi, Malakand, Swat, Dir Lower, Dir Upper, Peshawar, Charsadda, Kohat, Hangu, Karak, Lakki Marwat, Dera Ismail Khan, Mansehra, Battagram and Bajur Agency (F. A. T. A) from 2013 to 2016 in Khyber Pakhtunkhwa Province of Pakistan. All the species were collected during different times of seasons but mostly from March to November of above mention years. The specimens of spiders were collected through hand picking, beat sheet, pitfall trafs and sweep nets in different ecological niches. Preserved specimen were observed under stereomicroscope with supply of light, While pictures were taken by Leca MZ 16 microscope with power light and diagram ofmales and females genitalia were drawn with compound microscope with tube camera (Lecucida). Also, Molecular work was carried out, to extract DNA from four legs of spider species of subtribe Aelurillina (Salticidae) by DNA extraction kits. Four genes (28SrDNA, Wingless, 16S-ND1, COI) were amplified and sequenced by using primers. This study admits nine families of spiders (Araneae) contain twenty Six Genera and distributed in thirty seven species. The species are Aelureillus sp.nov, Aelureillus logunov, Bianor albobimaculatius, Bianor Pujabiicus, Carrohotus sannii, Castianeira sp.nov., Clubiona sp.nov., Crossopriza maculipes, Epocilla sp.nov., EvarchaArabeca, Hasarius adansonii, Heteropoida afghan, Hippasa paritita, Langona calcarata, Langona sp.nov., Lycosa sp.nov., Myrmarachni bicuirvata, Olios sanguinifrons, Oxyopies javana, Pardosa birmanica, Pardosa chaihraka, Pardosa flavisteirna, Peuceitia Arabica, Phelgra bresnieri, Plexippus Kondarensis, Plexippus paykulli, Plexippoides flavescens, Pseudicius admirandus, menemerus nigli (female first time described), steinaelurillus sp.nov., Thanatus fornicatius, Thyene Imperialis,Xysticus preuidocristatus, Yaginumaella sp.nov.,Zelotes spinuilosus. These Species are distributed in genera Bianor, Carrohotus, Castianeira, Chinattus, Clubiona, Crossopriza,Epocilla, Evarcha, Hasarius, Heteropoda, Hippasa, Olieos, Oxyopies,Lycosa,Myrmarachni, Pardosa, Peuceitia, Phlegra, Plexippous, Plexippoides,Pseudicius, Rhene, Thyene,Steinaelurillus, Xysticus, Yaginumaella, Zelotes and consitituted by the Families Corinnidae Clubionidae, Lycosidae Oxyopidae, Philodromidae, Pholcidae, Salticidae, Sparassidae, Thomisidae,. Present study determined 10 new to science species from genera Aelureillus, Castianeira, Chinattus, Clubiona, Epocilla, Evarcha, menemerus Female, Langona, Lycosa, Rhene, Steinaelurillus, Yaginumaella, some of them have been submitted for publication. The more diverse family is Salticidae with 14 genera and 23 species. The aim of the present study was to know the current status of unexplored region for spiders biodiversity with molecular phylogenetic studies to show the present relations of species with their ancestors. We recommend future work on Taxonomy, Biodiversity and Molecular Phylogenetic study of araneae and related arachnids of Pakistan.
