Textile dyes have always been considered in the context of recalcitrant xenobiotics compounds in water and soil ecology. Present research work was focused on the bioremediation potential of Agaricus species to decolorize and to detoxify the different synthetic dyes and dye containing effluents. The purification, characterization, kinetics & thermodynamic studies of ligninolytic enzymes produced by the fungi during dye decolorization process, were also performed. Complete (100%) removal of NOVASOL Direct Black and 83.34 % Reactive Black by Agaricus bitorqus A66 & Agaricus bisporus A21 were attained by optimizing optimal conditions like media composition, pH, temperature, inoculum size, carbon and nitrogen supplements, metal ions, redox mediators and dye concentration. In the entire processes of optimization of decolorization lignin peroxidase (LiP) was the major enzyme with minor activities of MnP and laccase when the decolorization of Novasol black was carried out by Agaricus bitorqus A66 and manganese peroxidase (MnP) was the main enzyme with minor LiP and laccase activities in case of reactive black treated by Agaricus bisporus A21. As highly efficient decolorization (100%) of synthetic dye was attained by these Agaricus species in a reasonably short period of time under optimum conditions, these were therefore, applied to check out their potential application in the decolorization of different practical/real textile industry effluent. It was observed that complete (100%) and 95.66 % decolorization of Navy blue colored effluent was achieved by Agaricus bitorqus and Agaricus bisporus A21 , respectively under optimum conditions. To access the water quality parameters, the treated and untreated effluent samples were analyzed to determine COD, BOD, TSS and TDS. The fungal treated effluents were less polluted as compared to original industrial effluents. The enzymes (LiP & MnP) produced under optimum decolorization conditions were purified to 1.54 & 6.9 fold with % yield of 6.74 and 8.48, respectively by ammonium sulfate precipitation, followed by three purification steps including, Dialysis, gel-filtration and anion- exchange chromatography. MnP and LiP secreted by respective fungi displayed optimum activities pH 7.0 & 6.0 and at 45 &400C, respectively. The Km values for veratryl alcohol and MnSO4 were 16.67 mM, 3.33 mM for MnP and LiP, having Vmax values of 179.17 and 231.00 mM, respectively. Thermodynamic and activation parameters provide a detailed mechanism for many chemical and biological reactions. The kinetic and thermodynamic characterization revealed Gibbs free energy ΔG* values for denaturation of MnP and LiP as 272.12 & 272.6 KJmol-1, enthalpy of thermal unfolding ΔH* of transition state was 39.91& 72.83 KJmol-1 and entropy of thermal unfolding (ΔS*) was -707.94 &, -609 Jmol-1K-1 at 50oC along with activation energy for inactivation of Ea 42.6, 75.6 KJmol-1, respectively.

علم اسباب ورود الحدیث: ایک تحقیقی جائزہ

Asbab-e-worood-e-Hadith means the context of background of a Haith. It is impossible to understand the original myth of hadith without knowing its background and context. This Article presents the concept of asbab-e-worood-e-Hadith. The discussion has been premeditated to explore the meanings and importance of asbab-e-worood. In this regard, keeping in view the nature of topic, the guidance has been sought from the Ahadith of Holy Prophet (PBUH). Asbab-e-worood in its nature having similarities with asbab-e-nazool-e-Quran. This article also describes the relationship between asbab-e-worood-e-hadith and asbab-e-nozool-e-Quran. In this regard some examples have been discussed also. This article also describes how asbab-e-worood is important to specify the meanings of text and explain the textual ambiguities. Key Words: Asbab-e-worood-e-Hadith Asbab-e-Nozool-e-Quran Sabab-e-worood qasasia 4.         Sabab-e-worood sawalia

Have Developing Countries Been Seeking to Minimize Welfare Cost of Taxation? Evidence from Barro’S Tax Smoothing Hypothesis

The developing economies are characterized by severe fiscal deficit, sky-rocketing public debt and unstable economic growth. To finance fiscal activities governments’ resources are limited. The deficits can be corrected through fiscal adjustment and regulations in the shape of government spending cuts, tax increase and/ or debt creation. One of the basic decisions the government has to make is to share out the burden of fiscal adjustment between spending, borrowing and taxing with a view to satisfying the dictates of efficiency and equity, including inter-temporal equity. This fine balancing of policy instruments to achieve well-known fiscal objectives involves, among other things, an evaluation of the level of taxes and spending to decide whether to adjust them at the economically realistic levels. Tax increases may be less problematic than reducing expenditure if the current level of tax revenue is comparatively low based. However, in the former case, considerations of the tax smoothing acquire significance. Another important issue, which this study tackles, is the problem of causality between taxes and spending. Particularly, in order to decide which variable should be given temporal priority, it should be known whether changes in spending lead, follow, occur simultaneously, or are independent of the changes in tax rates. The present study finds that the fiscal stances of Sri Lanka, India and Pakistan are not significantly different from other developing countries, so that our analysis of these countries can be safely generalized to other developing countries. It also aims to check whether these developing countries have in effect adopted a tax smoothing policy to overcome the fiscal deficit and what forms such policy has taken. The empirical analysis presented here reveals that Pakistan and Sri Lanka have tried to minimize the welfare cost of taxation but these have not been policies fully consistent with the best practice tax smoothing. On the contrary, India has not sought to smooth its tax rates to minimize the welfare cost at all. Moreover, fiscal policies in Pakistan, Sri Lanka and India have been consistent with the fiscal synchronization, the spend-and-tax, and the institutional independence hypotheses respectively. The present study makes quite a few non-trivial recommendations, which may or may not accord with so-called common sense approaches to such problems. For instances it shows at length that to minimize the welfare cost of taxation the governments should finance their permanent expenditure by increasing the tax rate while transitory shocks to the expenditures or output should be financed by creating public debt. Such debt should, however, be contingent and retired in good days. In the same vein, it recommends that, a countercyclical (debt falls in booms and rises in recessions) policy might also be adopted. On the other, a pro-cyclical policy may lead to volatility in tax rates and increase the welfare cost of taxation. It is asserted that if developing countries fiscal policies are reformed along the lines suggested in this study it would lead to major over-hauling of the fiscal stances of the developing countries----those which would lead to efficient and equitable policies based on robust theoretical and empirical foundations.
