Tularemia, caused by Francisella tularensis (F. tularensis), is a zoonotic disease transmitted through contact with infected animals and contaminated environment. The disease has been reported from many countries of the world but no study has been done in Pakistan. In the current project, a total of 2280 soil samples representing 456 villages of eight districts of Punjab province were collected from way-points having human-animal interaction, processed for genomic extraction and tested through real time PCR for presence or absence of F. tularensis. Association of risk factors was determined from data such as gender and age of animals, plough method, irrigation system, fertilizer type used, availability of veterinary services, level of farmer education, physical and chemical composition of the soil. Moreover serum samples (n=707) collected randomly from goat (n = 200), sheep (n = 175), cattle (n = 179), and buffalo (n = 153) were analyzed for antibodies against F. tularensis by using enzyme-linked immunosorbent assay. Seventy four soil samples (3.24 percent) were found positive for F. tularensis. Phylogenetic analysis showed 100 percent similarity index with F. tularensis sub specie holarctica reported from other regions like USA, Sweden, Spain, Turkey and Germany. Presence of F. tularensis in soil showed negative association with increase in number of human density (0.7159; 0.3834- 0.2054). Prevalence of anti- Ft ELISA antibodies were significantly higher (p<0.05) in large ruminants (cattle and buffalo) as compared to small ruminants (goat and sheep). Age and gender- wise analyses showed non-significant differences (p>0.05) between small and large ruminants. Whereas, rain-irrigation system (2.96: 1.35- 6.48), lack of veterinary services (4.77:1.26-18.03) and use of organic fertilizer (5.3: 11.38- 20.39) have positive association with prevalence of anti- Ft ELISA antibodies in the serum. Sero-prevalence of F. tularensis in the animals has significant viiassociation with quantity of clay in soil (p<0.05). A conventional PCR based test has also been optimized for detection of F. tularensis using tul4 gene specific primers. Specificity of primer showed Ft detection in soil DNA in the presence of other cross-reactive organism. Sensitivity was determined in two fold dilutions with detection limit of up to 320 pg/μL. Utilizing pET28a vector, a construct was prepared containing transformed tul4 gene (450bp) showing 100 percent sequence homology to query gene sequence. For manufacturing diagnostic assays especially in developing countries where availability of BSL-3 facilities and positive control reagents is an issue, provision of tul4 gene based constructs in vector can act as positive control and safe to use.

جنت کے معنی باغ بھی ہیں ، بہشت کے لیے بھی جنت کا لفظ بولا جاتا ہے۔ جنت کا تصور جب ذہن کے در یچوں پر دستک دیتا ہے تو قلب و اذہان میں اس صحت افزا تخیل کے باعث نئے نئے شگوفے کھلنے شروع ہو جاتے ہیں۔ اور اس وقت جسم و جان میں پیدا ہونے والی تازگی و طراوت روح تک سرایت کر جاتی ہے۔ جس کو جس سے جتنا عشق ہوگا وہ اس کو اپنی جنت اور اپنی بہشت قرار دے گا۔ کسی کی جنت اس کا گھر ہوگا ،کسی کی جنت اس کا در ہوگا،کسی کی جنت اولا د ہو گی ،کسی کی جنت امّ اولا د ہوگی، کسی کی جنت اس کا مکان ہوگا ،کسی کی جنت اس کا سلطان ہوگا۔ سب کی جنت ان کے اپنے اپنے ذوق کے مطابق ہے لیکن میری جنت میرا پاکستان ہے کیونکہ یہ ہے تو سب جنتیں ہیں۔ اگر یہ نہیں ہے تو پھر جنت بھی جہنم کا عذاب ہے کیونکہ اسی کے دم قدم سے حقیقی جنت کی بہاریں ہیں۔

مانگی ہوئی جنت سے دوزخ کا عذاب اچھا
وطن اس مقدس سرزمین کا نام ہے جس کی آغوش میں انسان جنم لیتا ہے جس کی ہوائیں اسے پروان چڑھاتی ہیں جس کی فضاؤں میں اس کی نشوونما ہوتی ہے جس کی مٹی سے اس کا خمیر اٹھتا ہے۔ اور اسی کے ذرّے ذرّے سے انسان کی عقیدت وابستہ ہوتی ہے۔ اس کی فضاؤں میں محبت کی خنکی ہوتی ہے، اس کے کھیتوں میں آنکھوں کا نور ہوتا ہے، اس کے گلستانوں میں چاہت کی چاشنی ہوتی ہے، اس کے ویرانوں میں یگانگت کی اپنائیت ہوتی ہے۔ اس کی ہر چیز جنت کا نمونہ پیش کرتی ہے۔

Career Decision Making: Career Episodes of Four Postgraduate Students in a Private University in Karachi, Pakistan

There are three components to making a career decision: knowledge about self, knowledge about success and what it takes to be successful in a certain occupation, and finally knowledge oflogical reasoning and knowing how it can be used to determine if a match exists between the two (Parson,1909). The aim of the research was to investigate the way postgraduate students of a private university in Karachi, Pakistan make their career decisions, as well as to highlight the factors that influence them and explorethe career advice they seek for making their career choices. The qualitative data was collected through a selected sample of postgraduate regular students (N=4) studying in a private university of the city of Karachi. Semi-structured open-ended interviews were conducted with students during university hours. The findings of the study revealed that all the students make their career decisions in different ways. Mostly the decisionsare made with the influence of male members within families particularly with the influence of the ‘father’ due to which students areunable to pursue the careers they dream of. The other factors that influence the career decisions of postgraduate students are: conditional family support; contextual factors such as lack of career guidance and counselling, lack of knowledge about labour market and emerging career trends. Therefore their main focus remains on finding a job that enables them to get high remuneration rather than focusing towards a strong career. Furthermore, it was also found that thestudents’ career decisions change overtime at various stages of their lives. This indicates a low satisfaction and confidence level of students regarding their future careers. Moreover, difference in ways of making career decisions is found among male and female research participants. Males gain more independence in making career decisions as they start earning and grow professionally, while females remain dependentuponfamily instructions throughout their lives pertaining to their career decisions.
