Polyphenol oxidases (PPOs) are the nuclear encoded, copper binding enzymes and are ubiquitous in nature. These enzymes are responsible for oxidation of ortho-and para- diphenols to quinones, which are highly reactive. Oxidized phenolic compounds in plant tissues due to PPO activity results is the formation of undesirable browning leading to unwanted color changes in food items during storage and processing which is one of the main causes of quality loss. However, upon damage or injury to plant, phenolic compounds oxidize to polymeric structure in order to protect against insects or micro- organisms. PPO is the source of quinones that limits the accessibility of alkylating dietary proteins to insects and ultimately starvation may happen. PPOs are of significant interest because of their high capacity to oxidize aromatic compounds. They are also suitable for some biotechnological applications in food, medicine, paper and textile industry. PPOs have a wide range of antioxidant activities and supplements with diet play a significant role in health care. Keeping in view the importance of PPOs, a study was carried out to characterize rice PPO promoter (OsPPOP) region at molecular level. The OsPPO promoter region was amplified, cloned and sequenced. Bioinformatics tools based comparative analysis of OsPPO promoter sequence and six other PPO promoter sequences available in Genbank revealed that PPO promoter regions possess common, novel and distinct cis-regulatory elements. The identified cis-regulatory elements have roles in environmental stresses (biotic and abiotic), activation of pathogen related genes and secondary metabolite path ways. Moreover, light responsive elements, pathogen inducible elements and copper binding elements are also present in the promoter region of PPO genes. Maps of cis- regulatory elements were framed manually in order to know the number, density and location of these elements available both on positive and negative strands of PPO promoters analyzed. Further, blast revealed that small regions of 1020 bp OsPPO promoter region have homology at chromosome 2 with the stretch of regions encoding ethylene gene, 3-isopropylmalate dehydratase small subunit and transposons (superfamily of elements Rim2/Hipa) and conserved sequences of CACTA-like family. xxiThen an expression construct was designed for the functional analysis of OsPPO promoter region in transgenic tobacco plant. The expression construct was transformed into tobacco via leaf disc method. Expression was observed in transgenic tobacco by GUS staining. The microscopic analysis revealed that the PPO promoter region is tissue specific and the GUS expression was observed in leaves; leaf veins and leaf petiole, stem; epidermis, vascular bundles, pith, collenchyma, sclerenchyma, xylem and phloem and roots; xylem and phloem. It is being first time reported that OsPPO promoter region is a strong wound inducible promoter. Further, Real-Time PCR study revealed that higher level of OsPPO promoter expression is at lower temperature and with the increase in temperature the expression level gradually decreases. The observed properties of OsPPO promoter can be used for plant transformation related studies as the OsPPO promoter region is plant origin promoter with wound inducible properties and it can also be expressed at higher level in low temperature conditions. Thus, the findings elucidate that OsPPO promoter is an important promoter which can be exploited for crop improvement and plant transformation studies.

مرحوم خلیق ٹونکی فن خطاطی میں اعلیٰ شہرت کے مالک تھے اس کے علاوہ مرحوم میں بڑی خوبیاں تھیں، صوم وصلٰوۃ کے پابند تھے، منکسر المزاج تھے، اخلاق کریمانہ کے مالک تھے۔ اور مفکرمِلّت حضرت مفتی عتیق الرحمن عثمانی ؒ کے شیدائی اور معتقد تھے۔ حضرت مفتی صاحبؒ نے ان کے فن کونکھارنے میں بڑا تعاو ن کیا۔ ادارہ ندوۃ المصنفین اوراس کے رسالے ’’برہان‘‘ ہی سے انہوں نے فنِ کتابت کی ابتدا ء کی۔ قبلہ مفتی صاحبؒ نے ادارہ ندوۃ المصنفین اوراس کے رسالے ’’برہان‘‘ کے لیے کتابت وطباعت میں ہمیشہ ہی اعلیٰ معیار کو برقرار رکھنے کی بھرپور کوشش کی۔اسی لیے انہوں نے ہندوستان کے اعلیٰ فنکاروں کوکھوج کھوج کرچُن چُن کرادارہ میں جمع کیا۔جناب آل حسنؒ، جناب محمد یوسفؒ، جناب عبدالقیوم ؒ اور خلیق ٹونکی کوبحیثیت کاتب کے ادارہ ندوۃ المصنفین میں رکھا۔ اﷲ تعالیٰ کے فضل سے خلیق تونکی کتابت کی دنیا میں شہرت کی بلندی پر پہنچے، سرکاری ایوارڈ بھی انھیں حاصل ہوئے، ملک اور بیرونِ ملک میں ان کے ہزاروں شاگرد موجود ہیں۔ادارہ ندوۃ المصنفین کی کئی کتابیں ان ہی کی کتابت کی ہوئی ہیں۔ ادارہ کی مشہور کتابیں’ مصباح اللغات‘ اور’ قصص القرآن‘ جناب خلیق ٹونکی ہی کی کتابت کی ہوئی ہیں۔
انھیں ہمیشہ ہی اس بات کا اعتراف رہاکہ ان کی کامیابی وشہرت میں بفضلِ خدا حضرت مفتی عتیق الرحمن عثمانیؒ اوران کے ادارہ ندوۃ المصنفین کابڑا ہاتھ رہاہے۔اسی لیے وہ ہمیشہ ہی ادارہ ندوۃ المصنفین اوراس کے ڈائرکٹر راقم الحروف عمیدالرحمن عثمانی سے زندگی کے آخری لمحے تک والہانہ لگاؤ وعقیدت رکھتے تھے۔

IMPLEMENTASI TEORI BEHAVIORISME TERHADAP PEMBIASAAN MEMBACA ASMAUL HUSNA

The theory of learning behaviorism observe the change of one's behavior as a result of past experience. This theory emphasizes that the behavior is a result of the interaction between the stimulus and the response. This theory does not recognize the presence of innate intelligence or derivatives, but rather recognizes that intelligence is influenced by environmental factors. This theory can be applied in learning that requires practice, habituation and repetition. Therefore, to introduce and understand about Asmaul Husna, the theory of behaviorism can be applied in the process of habituation. The recitation of Asmaul Husna is very much contained in its role. Among them to introduce and hang students about Asmaul Husna, so students will feel closer to God if they often apply Asmaul Husna readings and will always feel that everything we do is supervised by God. Like the example of one of the traits contained in Asmaul Husna.  Based on the discussion, the problem raised is how to implement behaviorism theory to the habituation of Asmaul Husna reading.

A History of Pakistan-China Relations 1973-2013 Reappraisal

The study investigates the China-Pakistan relations in historical context applying into the theoretical framework of power transition theory in general and hard and soft power theory specifically and its implications on foreign policy of Pakistan. In fact, since the inception of Pakistan, Pakistan had always been in search of security because of its geo-strategic situation and external pressures. Further, the study addresses that how the international scenario and outward dynamics affected foreign policy of Pakistan. For example, during the Cold War era and USSR war in Afghanistan because of bipolar dimensions across the World, Pakistan had to tilt towards Western allies to protect state sovereignty and maintain the integrity of its citizens. One thing that proved very true that Pakistan always desired to join those allies who always committed to preserve state from external enemies. The study spotlights that how the changing global episodes since the event of 9/11 altered Pakistan’s foreign policy choices. Meanwhile, Pakistan decided to strengthen its ties with rising power of China. Despite Pakistan’s constant contribution in the war on terrorism, it had been considered as a state of ‘safe Haven’ for terrorists and did not able to attain the confidence of its Western allies. By the times, Pakistan’s overseas strategy slanted towards the Chinese foreign policy goals of influencing the World with the image of a soft power. Likewise, the social and cultural relations enabled to gain the psychological confidence of Pakistan and its people by enhancing people to people relations. The dissertation draws extensively on the declassified official records to prove the theoretical and empirical contribution of this study that how far bilateral and political relations turned the shape of economic to geo-economic and taking the U-turn from geo-economic to geo-strategic ties between both states in the 21st Century.
