Hereditary neuromuscular disorders are a clinically and genetically heterogeneous group of genetic conditions that affect about 1 in 1000 individuals worldwide. These diseases affect the muscles and their direct nervous system control. The conditions are characterized by progressive muscle degeneration and weakness and constitute a great disease burden. Genetic defects in the proteins that maintain the motor demand and neural inputs lead to neuromuscular disorders. This study was aimed to explore the genetic cause of four neuromuscular disorders identified in five Pakistani families using whole exome and Sanger sequencing. Genomic DNA was extracted from peripheral blood of the recruited families. Whole exome paired-end sequencing was performed by generating 51 Mb Sure Select V4 libraries. DNA shearing, hybridization using RNA-based library baits, target capture and bridged amplification were subsequently carried out. The imaging and extension was achieved in automated cycles by mounting the clusters-bearing flow cell onto the Illumina HiSeq 2000/2500 sequencer. The data was analysed using standard bioinformatic pipeline. Raw read sequences were proceeded for recalibration of the base quality and removal of duplicates. Genome Analysis Toolkit (GATK3.v4) was used to call variant quality score recalibration and short insertions and deletions. Picard-tools- 1.118 was used to mark the duplicates. The variants with minor allele frequency value less than 0.01 were considered rare pathogenic variants. Deleterious effects of the variants on the structure and function of the protein were predicted through various bioinformatics tools. The variants present in the healthy unrelated individuals were excluded. The genotype of candidate variants was confirmed in the family members through Sanger sequencing. The proband of the family A [lab ID: NP-08] affected with autosomal dominant familial hypokalemic periodic paralysis (hypoKPP) was subjected to whole exome sequencing. A heterozygous missense variant (c.919A>G; p.Met307Val) was found in KCNJ2. Sanger sequencing verified the variant segregation in the family with disease phenotype with incomplete penetrance. The bioinformatics tools ranked the p.Met307Val change in KCNJ2 as deleterious. The variant was conserved in the 100 vertebrate species and is the likely cause of the hypoKPP in the Pakistani family. This investigation expands the underlying genetic etiology of familial hypoKPP. The proband of the family B [lab ID: NP-05] affected with autosomal recessive Charcot- Marie-Tooth disease type II was subjected to whole exome sequencing. A homozygous missense variant (c.1591C>A; p.Pro531Thr) in IGHMBP2 was shortlisted. Sanger sequencing revealed full segregation of the variant with the disease phenotype in the family. The parents of the affected individuals were heterozygous for the position. In silico analysis of the c.1591C>A substitution predicted deleterious effect on the protein structure and function. The variant was conserved in the vertebrate species. We conclude that IGHMBP2 c.1591C>A variant is the likely cause of the CMT2 disease in the family. Whole exome sequencing of the proband of the family C [lab ID: NP-07] affected with autosomal recessive Charcot-Marie-Tooth disease type IV was performed. No variant was found in the genes previously linked to neuromuscular disorders. Following disease model and tissue and organ specific expression, three rare candidate variants were shortlisted including compound heterozygous variant (c.874_875insGA, p.Lys292fs; c.871_872delCG, p.Arg291fs) in HADHA, homozygous missense mutation (c.2062C>T; p.Arg688Cys) in SLC6A6 and homozygous missense variant (c.63917G>A; p.Arg21306His) in TTN. However, none of the variants cosegregated with the disease phenotype in the family. These results support the evidence of further genetic heterogeneity in CMT disease. We believe that a hitherto unidentified genetic or epigenetic factor is the cause of the disease in the family. The families D [lab ID: NP-12] and E [lab IDs: NP-13] affected with autosomal recessive infantile-onset Pompe disease (IOPD) were subjected to Sanger sequencing. Short oligonucleotide sequences were used to screen GAA in both the families. A rare novel homozygous missense c.2561G>A variant was found segregating in the family D. The parents were carriers for this variant. While a rare heterozygous variant (c.2773 A>C) in GAA was identified in the family E. The genotype of the mother was heterozygous for the variant but no GAA variant identified in the father. The other unidentified variant in the family may be present in the promoter or regulatory sequence. The variants were not present in our in-house database of the local unrelated healthy population. Different bioinformatic tools predicted the identified variants to have deleterious effects on GAA protein structure and function. The variants are also conserved in the vertebrate species. We suggest that these GAA variants are the likely cause of IOPD in these families. In conclusion, the study highlights the clinical significance of whole exome sequencing in the molecular diagnosis of heterogeneous hereditary neuromuscular diseases. The data should also help in the prenatal diagnosis and improved genetic counselling of the families.

                جدیداُردو نظم کا وہ پودا جسے آزادؔ اور حالیؔ نے لگایا تھا اور اس کی آبیاری میں اکبرالٰہ آبادی، اسماعیل میرٹھی اور علامہ اقبال جیسے شعرا کا ہاتھ ہے تو اسے پروان چڑھانے میں میراجی اور ن۔م۔راشد نے بھی اپنا حصہ ڈالا اور اس کی تراش خراش کر کے اس کو ایک مضبوط تناوردرخت بنانے میں اہم کردار ادا کیا۔ ۱۹۶۰ء کی دہائی کے شعرا نے آزادنظم کو اپنے اظہار کا سب سے اہم اور مقبول ذریعہ تصوّرکیا اور جان بوجھ کر پابند نظمیں لکھنے سے گریز کیا۔ ان شعراکی کوشش سے پہلی بار اُردو شاعری میں وسیع پیمانے پر آزادنظم لکھی گئی۔ اس دور کے اکثر شعرا نے تو صرف نظم کے میدان میں طبع آزمائی کی مگر کچھ ایسے تھے جنھوں نے غزل اور نظم دونوں میدانوں میں اپنے فن کے جوہر دکھائے۔

Konsep Ekopark Dan Ekowisata Di Ruang Terbuka Hijau Kawasan Pusat Pemerintahan Pasaman Barat

Kabupaten Pasaman Barat berada di Proponsi Sumatera Barat, ibu kota-nya adalah di Simpang Ampek. Potensi kawasan Simpang Ampek ini merupakan kawasan strategis sebagai pusat pemerintahan, pusat pertumbuhan ekonomi dan sosial budaya masyarakat Pasaman Barat, jika pertumbuhan semua sektor ini tidak berdasarkan Rencana Tata Bangunan dan Lingkungan yang sudah di rencanakan maka akan berdampak negatif di segala bidang. Di Kawasan Pemerintahan ini terdapat Peruntukan lahan Sebagai Ruang Terbuka Hijau berada pada zona II di kawasan Pemerintahan Pasaman Barat Padang Tujuh. Perencanaan Ruang Terbuka Hijau untuk publik seharusnya mengacu kepada kelestarian Lingkungan alam. Tujuan dari penelitian ini adalah untuk menemukan konsep rancangan ekopark dan ekowisata untuk menunjang kelestarian alam. Metode yang digunakan adalah dengan metode deskriptif kualitatif yang menganalisa studi lapangan maupun literatur, dengan cara mengidentifikasikan masalah-masalah yang ada di lokasi penelitian atau site dan sekitar site sehingga mendapatkan hasil dari pemecahan masalah, untuk menemukan sebuah Konsep Rancangan Ekopark dan Ekowisata pada Peruntukan lahan Ruang Terbuka Hijau.

Watershed Modelling of Malir River Basin to Evaluate the Surface Water Potential in Karachi: A Gis Perspective

Water is the basic necessity for life and equally important for every human being. However, water scarcity is a very serious problem in the world that is becoming a global challenge for the decision-makers and planners despite recent technological advancement. Phenomenal population growth with haphazard urban development and climate change are the main reasons behind growing water crisis that is mostly affecting metropolitan cities around the globe. Karachi, a city of more than 20 million people, obtains water from Indus River for its domestic use. Its local water resources are insufficient in quantity as well as quality. Water scarcity for metropolitan is getting worse day by day due to exponential population growth, insufficient water supply from Indus River, scarce but erratic rainfall and the overall absence of sustainable water management. The watershed potential is unexploited rather depleting due to massive impermeable infrastructure development for settlements and uncontrolled sand excavation activity in river beds. These are increasing surface runoff and reducing groundwater as a result. In case of heavy rain, a large amount of runoff water drained into the Arabian Sea due to lack of water storage arrangements. This research aims to explore the watershed potential in Karachi region for identifying local resources and their harnessing. The study evaluates the surface water potential for rainwater harvesting (RWH) and to find the best practices for optimum water resource management in nearby villages and newly planned urban housing schemes located in the potential watersheds. Advanced techniques of Geographic Information System (GIS) and Remote Sensing disciplines are integrated with rainwater runoff model to discover the potential of watershed areas in the region. Soil Conservation Service (SCS) Curve Number (CN) method is adopted to evaluate rainfall runoff potential, as this technique is widely used and most reliable as compared to other methods used for such studies. Spatial Multi Criteria Evaluation (SMCE) technique is integrated with Analytic Hierarchy Process (AHP) method in GIS environment for rainwater harvesting sites (check dam, percolation tank, and farm pond) and dam site selection Thirty years rainfall and runoff data are used for watershed modelling. In the study, watershed characteristics are delineated over high scales from stream network to sub-watersheds and watershed boundaries along with area maps of RWH sites (11 check dams, 11 percolation tanks and 19 farm ponds) and dam sites (3 dams) to understand their potential in a geospatial context. It is estimated that annually about 4,22,280 m3 water is available only in Malir River. If this water is harnessed in a realistically feasible proportion, it may be a sustainable solution for overcoming the water crises. The RWH and dam sites suitability maps provide a clear picture of the existing potential and its spatial extent; while projected dam profiles and their characteristics give an in-depth overview of the suggested sites that can be used for future planning and estimations. Furthermore, this study is very effective for creating awareness among the stakeholders at the local and regional scale. It is recommended to implement the RWH techniques for all stakeholders including government authorities who should improve their knowledge and take sustainable initiatives with full ownership.
