Molecular Genetic Elucidation of Inherited Retinal Diseases Inherited retinal dystrophies are characterized by gradual loss of vision due to underlying degeneration of light sensitive photoreceptors or the surrounding retinal pigment epithelium. Clinical subtype of RD called Retinitis pigmentosa involves progressive degeneration of Rod photoreceptor cells earlier in life than the cones cells, and represents one of the most common forms. As rod cells are sensitive to dim light the patients first experience night vision problems but might eventually end up with complete blindness as the degeneration prevails. Of the inherited forms of RP the autosomal recessive inheritance pattern is most common and accounts for more than 50% of cases. RP is genetically heterogeneous, where arRP is at the extreme as mutations in about 40 genes are known to cause same phenotype. Despite large number of genes associated with arRP these genes only explain about 60% of the cases and hence the further research is warranted to find out the missing pieces. In three families, missense mutations were identified in TULP1, which include p.(Thr380Ala), p.(Arg482Gln), and p.(Lys489Arg). In three arRP families protein- truncating mutations were identified in ABCA4 p.(Gln2220*), AIPL1 p.(Trp278*) and CERKL p.(Arg283*). In three families novel missense mutations were identified in different genes, which include CNGA1 (p.(Gly433Asp), EYS (p.(Asp2767Tyr) and PDE6A (p.(Arg544Trp). A novel splice site mutation (c.2493-2A>G; p.(?), was identified in CNGB1 in a family with arRP. In five other unrelated arRP families, previously reported mutations were identified, which include two families with a missense mutation p.(Glu150Lys) in RHO, two families with mutations p.(Arg44Gln) and p.(Ser121Leufs*6) in RPE65, and one family with a mutation p.(Thr745Met) in CRB1. In an X-linked RP family the causative mutation p.(Glu809Glyfs*25) was identified in RPGR. A splice site mutation c.488-1G>A; p.(?) in IQCB1 was identified in a family with syndromic RP. Two families with fundus albipunctatus were identified to have a frameshift p.(Val305Hisfs*29) and a missense p.(Met253Arg) mutation in RDH5 gene. xiiIn an arRP family Exome next generation sequencing (NGS) helped to identify a plausible pathogenic variant c.3269G>A; p.(Arg1090Gln) in SNRNP200, a gene previously found to be mutated in autosomal dominant RP (adRP). We hypothesize that the mutation identified in the Pakistani arRP family represents a hypomorphic variant but further experiments are warranted to prove the causality of the mutation. An overlapping homozygous region was identified that was shared between all the affected individuals of two arRP families. The region encompasses CLRN1, a gene previously found to be mutated in Usher syndrome type III. Two novel missense mutations p.(Pro31Leu) and p.(Leu154Trp) were identified, which were proven to be pathogenic. Hence a novel genotype-phenotype correlation was established for CLRN1. Taking together, sequence variants were identified in 32 of 41 (78%) families of our Pakistani RD cohort, which very likely explains the retinal phenotypes. In addition, we were able to identify nine potential novel arRP loci, which are likely to harbor novel retinal disease gene.

Empowering Women through Media in Algerian Non-Governmental Organizations

تعد وسائل الإعلام أداة تستخدمها العديد من المنظمات غير الحكومية في جميع أنحاء العالم. تهدف هذه الدراسة إلى التحقيق في الاستراتيجية التي تستخدمها المنظمات النسائية غير الحكومية في الجزائر لتعزيز ومناصرة المشاكل التي تؤثر على حقوق المرأة وتمكينها. يتم تحقيق ذلك من خلال النظر في كيفية تعبئة المنظمات غير الحكومية لمواردها واستخدام تقنيات محددة لدعم برامج تمكين المرأة. وتنتمي هذه الدراسة إلى البحوث الوصفية التي تختار طريقة مختلطة تجمع بين المقاربات النوعية والكمية. بتعبير أدق، استخدمت استبيانًا منظمًا وزع على 200 مستجيب و16 مقابلة شبه منظمة موجهة إلى مديري المنظمات غير الحكومية من 14 منظمة غير حكومية مختلفة في الجزائر العاصمة. تكشف النتائج أن المنظمات غير الحكومية تختار أدوات وسائل الإعلام اعتمادًا على صلتها بالجماهير والممارسات المستهدفة. في حين أن القنوات الإعلامية التقليدية لم تكن تستخدم بشكل متكرر من قبل المنظمات غير الحكومية وكانت فعالة من حيث الوصول إلى أهدافها الدعائية، كان لوسائل التواصل الاجتماعي تأثير أكثر عمقًا على الناس. ومع ذلك، استخدمته بعض المنظمات غير الحكومية للتأثير على الجمهور من خلال تعزيز النشاط وزيادة الوعي بقضايا المرأة. النظر في ظهور المرأة في أماكن جديدة، مثل وسائل التواصل الاجتماعي، والتحقيق في وظيفة المساحات التي تم تشكيلها حديثًا في عملية التمكين يجب استكشافها وتقييمها بشكل أكبر لمساعدة المنظمات غير الحكومية على القيام بعمل أكثر فعالية في هذا المجال.

Dynamical Evolution of Collapsing Stellar Systems in F R Gravity

This thesis is devoted to study the dynamical instability of some relativistic collapsing self-gravitating structures with both Newtonian and post-Newtonian approximations in metric f(R) gravity. In this setting, we consider evolution of spherical, cylin- drical and restricted axial stellar systems ¯lled with expansion and expansion-free adiabatic/non-adiabatic matter con¯gurations. We analyze the role of adiabatic index (sti®ness parameter) in the instability constraints of these self-gravitating structures. We construct dynamical equation using contracted Bianchi identities of the e®ective dark sources as well as usual matter distribution. The perturbation approach is ap- plied on physical variables and then formulate modi¯ed versions of collapse equation which leads to instability constraints at both N and pN regimes. We ¯rst consider spherically and cylindrically symmetric spacetimes ¯lled with charged and uncharged expansion-free anisotropic matter distributions. It is found that the adiabatic index does not have any role in the expansion-free evolution within N and pN approximations. Rather, this range is governed by pressure anisotropy, ra- dial pro¯le of system energy density, f(R) model and electric charge (for charged distribution). We also explore the role of heat radiations in non-viscous charged spherical and cylindrical systems as well as shearing viscous uncharged axial relativis- tic interior. It is found that heat radiations try to decrease stability of the evolving systems, while viscosity tends to increase system stability. The electromagnetic ¯eld decreases instability regions for charged spherical systems while its opposite e®ects have been observed for cylindrical collapsing self-gravitating systems. We conclude that f(R) dark energy sources coming up from the well-known f(R) models a®ect xi xii the whole dynamical behavior of collapsing systems due to its repulsive nature. Finally, we study the factors involved in the energy density irregularities of rela- tivistic planar °uid distribution in the presence of Palatini f(R) corrections. For this purpose, we develop a link between the Weyl scalar and structural properties of the system by a couple of di®erential equations. We also investigate the e®ects of Palatini f(R) terms in the formulation of structure scalars obtained by orthogonal splitting of the Riemann tensor in general relativity. We then identify the parameters which produce energy density irregularities in expansion and expansion-free dissipative as well as non-dissipative matter distributions. It is found that particular combinations of the matter variables lead to irregularities in an initially homogeneous °uid dis- tribution. We conclude that Palatini f(R) extra corrections tend to decrease the inhomogeneity, thereby imparting stability to the self-gravitating system.
