Salinity is a major abiotic stress affecting plant growth and productivity worldwide. The salinity-induced reduction in growth and development of plants is associated with ionic/osmotic effects, nutritional imbalance or oxidative stress. Chillies are sensitive to salinity and its growth reduction is reported in developing countries. Different germination and pot trials were conducted to identify morphological, chemical and bio-chemical attributes that can be used for characterization of salinity tolerance in chilli genotypes. In first experiment, 10 Chilli genotypes were screened out for salinity tolerance. One salt sensitive and one salt tolerant genotypes were selected on the basis of morphological and chemical attributes (germination percentage, shoot length, root length, plant fresh weight, plant dry weight and leaf sodium and potassium contents). Then, effects of different salinity levels were assessed on morpho-physiological, chemical, biochemical and enzymatic attributes of salt tolerant and salt sensitive chilli genotypes. With the increase in salt stress, the antioxidant enzymatic activity was increased in both genotypes but higher enzymatic activity was noted at 5.0 and 7.0 dS m-1 salt stress in salt tolerant genotype Plahi. In the third experiment, both genotypes were tested for phospholipid signaling under salt stress and both showed increasing pattern of PIP2 and PIP responses and again maximum signaling response was observed for the salt tolerant genotype Plahi. There was an optimization experiment to find out the best proline dose that could alleviate the salinity induced affects in chilli plants. With the application of different levels of foliar proline, all growth attributes increased but maximum increase was recorded at 0.8 mM proline level. Then foliar spray of the best proline dose 0.8 mM was applied to both genotypes under salt stress of 5 dS m-1. There was significant increase in compatible solutes (proline and glycinebetaine), enzymatic activity, photosynthetic rate and stomatal conductance by proline application under salt stress. There was significant decrease in toxic ions (Na+ and Cl-) and increase in K+ concentration while maximum decrease in Na+ and Cl- contents was observed in salt tolerant genotype Plahi relative to the salt sensitive genotype A-120. Hence, this study implies that proline application is a useful tool to combat salinity stress in chilli genotypes

Intelligence system is considered to be one of the important tools used by military and civil secret agencies to defend and strengthen a nation. Intelligence system is thought to be one of the oldest studies of known history. Intelligence system consists of correct and accurate information, gathered after great struggle and facing difficulties. This department if related to both peace and war. Intelligence is a basis of formulating all military strategies and plans. The importance of intelligence system both in day to day life and as a nation cannot be overemphasized. This article recounts the intelligence systems and management of the resources of secret services of the companions of the Holy Prophet (SAW) and, thereafter, the Muslims rulers. Furthermore, the principles derived from the era of the companions of the Holy Prophet (SAW) regarding intelligence system have also been discussed in this chapter. The guiding principles that are still valid even today includes: (a) Training of Personnel Since espionage helps to strengthen the roots of a state and protect it from its enemies, therefore it requires a team of well trained professionals with latest technology and trends. Islam emphasized on two aspects of early warning, one is professional and the other is ethical.(b) Counter Espionage. An Islamic state must have an effective network of espionage to keep an eye on all the activities of the enemy. This is known as counter espionage. (c) Reconnaissance. This aims at the fore knowledge of the intentions of the enemy so that one can have a better planning in case of an attack. (d) Verification of Information. Information from an agent should be verified from other sources. An operative may feed false information due to lack of experience and competency and that may create an embarrassing situation. (e) Security of Information. Don’t share your secret, try to protect them. If national secrets are compromised they may cause an extensive damage to national interest. (f) Interrogation of POW. Whenever enemy spies or soldiers are arrested in a war they should be interrogated for extraction of information. They may be subjected to mental stress. (g) Fore Warning of the Enemy. This requires the launching of own agents in the enemy ranks for knowledge of their future plan likes attacks. (h)Treatment of Spies. If anyone is found to be guilty of spying for enemy, he may be penalized with death punishment.

Molecular Characterization of Thermostable Cellulase from Thermotoga Naphthophila.

Current study deals with the production, purification and characterization of recombinant thermostable cellulase from Thermotoga naphthophila. PCR using the genomic DNA of T. naphthophila as template resulted in amplification of 1 kb cellulase gene. The amplified cellulase gene was cloned in pTZ57R/T and sub-cloned in pET28a. The expression of recombinant cellulase was analyzed using BL21 CodonPlus (DE3) cells as expression host. The expression studies resulted in the production of recombinant enzyme as soluble protein. The recombinant protein was purified by affinity column chromatography. The characterization studies of purified protein demonstrated the optimal enzyme activity at 90 °C and pH 4.8. The presence of cobalt enhanced the cellulase activity and 2.5 mM cobalt was recorded the optimal concentration for the maximal cellulase activity. SDSPAGE analysis confirmed the molecular weight of recombinant protein as 39 kDa. The protein was found thermostable which retained more than 70% residual activity with an incubation of 1.67 h at 90 °C in the presence of cobalt. Presence of ionic and non-ionic detergents showed an inhibitory effect on the enzyme activity. Kinetic studies of recombinant protein demonstrated the km and Vmax values of 0.22 mg/mL and 2500 µmoles/min respectively using carboxymethyl cellulose as substrate. The deinking potential of recombinant cellulase to remove ink from the paper makes this enzyme a suitable candidate for its use in paper Industry. We are reporting a new member of M42 Family of aminopeptidases. The stability of this recombinant cellulase at wide range of temperature, pH, its high level activity and its paper deinking potential makes it a suitable candidate for its use in paper industry.
