Insulin secretory dysfunction is major pathophysiology of diabetes which is aggravated by β-cell apoptosis. Metformin and sulfonylureas, two major oral hypoglycemic agents used for the treatment of diabetes, enhance insulin sensitivity and stimulate insulin secretion, respectively. Though some dipeptidyl peptidase-IV inhibitors are reported for β-cell protective activity; however, results are not conclusive yet. Therefore, taking advantage of our in house pure compounds library an attempt was made to identify compounds having β-cell protective as well as insulin secretory activity, and study their mechanism(s) at molecular and cellular levels. In order to evaluate the compounds for this dual activity, we used pancreatic β-cell line MIN6 cells to develop H2O2 mediated apoptosis and glucose stimulated insulin secretory assay systems. Out of 34 tested compounds, seven (7) compounds showed strong β-cell apoptosis inhibitory activity. Amongst them genestein (GS), quercetin (QCT), and cinnamic acid (CA) were evaluatedthrough triple channel immunostaining for mitochondria-actin-nuclei which revealed restoration of mitochondrial membrane potential, preservation of cytoarchitecture of cells and reduced nuclear condensation in MIN6 cells. Oxidative stress mediated activation of cleaved caspase 3 was undetectable in MIN6 cells after treatment with these compounds, further confirming the inhibition of mitochondria mediated apoptosis. Moreover, real time PCR study of mRNA expression showed that QCT and GS both downregulated the expression of apoptotic gene Casp9, and increased expression of both Ins1 and Ins2 genes. GS, QCT and CA also stimulated insulin secretion from MIN6 cells/ mice isolated islets. We found CA significantly inhibited nuclear condensation, decreased TUNEL positivity of pancreatic β-cells and preserved islets cytoarchitecture in Wistar rats. The effect of nicotinamide-cinnamic acid (NA-CA) was also studied in vivo, and we found that NA-CA significantly decreases β-cell apoptosis and induces insulin secretion than these agents alone. Immunohistochemical analysis of NA-CA pre-treated rat pancreas revealed decreased cleaved casp3 levels and increased phosphorylation of ERK½ in β-cells. Moreover, real time PCR anlaysis of NA-CA showed decrease in expression of Casp3 and Casp9 mRNA in MIN6 cells. This suggests that dual effect of NA-CA seems to be mediated via ERK½ signaling pathway and through modulating the mRNA expressions of apoptotic proteins. Intriguingly, we found orobol, tambulin and hispidulin as novel insulin secretagogues in the current study. TM enhanced insulin secretion in a dose dependent manner only at stimulatory glucose concentration in isolated mice islets. Pharmacological inhibition of protein kinase A and calcium channels significantly decreased insulin secretion induced by TM. This suggests that TM exerts an exclusive insulin secretory effect by modulating Ca2+ channels and PKA pathway. From the current study, some compounds with potent insulin secretory activity were identified. Interestingly few lead compounds having dual activity were also discovered. Taken together, these compounds may serve as lead compounds to be further studied for their anti-diabetic activity.

۲۱ جولائی سن ۱۹۶۰ کے مقدمہ رشیدہ بیگم بنام شہاب الدین میں مغربی پاکستان ہائی کورٹ کے ایک رکن فاضل جج محمد شفیع صاحب نے مذکورہ مقدمہ کے فیصلے میں تعدد ازواج کے مسئلے پر سورہ النساء کی آیت نمبر ۳’’ وَاِنْ خِفْتُمْ اِلَّاّ ۔۔۔۔ ثَلَاثَ وَرُبَاعًا ‘‘ کے تحت اجتہاد کیا اگرچہ حضانت اور سرقہ کے بارے میں بھی اجتہاد کیا لیکن ہمارا مضمون تعدد ازواج کے بارے میں ہے لہذا اسی کو زیر بحث لاتے ہیں ۔اور مولانا مودودی کے جوابات ان اجتہادی غلطیوں کے بارے میں پیش کرتے ہیں ۔
پہلی غلطی: تعجب ہے کہ فاضل جج کو اپنے ان دونوں فقروں میں تضاد کیوں نہ محسوس ہوا ۔ پہلے فقرے میں جو اصولی بات انہوں نے خود فرمائی اس کی رو سے زیر بحث آیت کا کوئی لفظ زائد از ضرورت یا بے معنی نہیں ہے ۔ اب دیکھئے ! آیت کے الفاظ صاف بتا رہے ہیں کہ اس کے مخاطب افراد مسلمین ہیں ۔ ان سے کہا جا رہا ہے کہ ’’ اگر تمہیں اندیشہ ہو کہ یتیموں کے معاملے میں تم انصاف نہ کر سکوگے تو جو عورتیں تمہیں پسند آئیں ان سے نکاح کر لو دو دو سے ، تین تین سے اور چار چار سے ، لیکن اگر تمہیں اندیشہ ہو کہ اگر نہ کر سکوگے تو ایک ہی سہی ۔۔۔‘‘ ظاہر ہے کہ عورتوں کو پسند کرنا ، ان سے نکاح کر نا اور اپنی بیویوں سے عدل کرنا یا نہ کرنا افراد کا کام ہے نہ کہ پوری قوم یا سوسائٹی کا۔ لہٰذا باقی تمام فقرے بھی جو بصیغہ جمع مخاطب ارشاد ہوئے ہیں ، ان کا خطاب بھی لا محالہ افراد ہی سے ماننا پڑے گا ۔ اسی طرح یہ پوری آیت اول سے لے کر آخر تک دراصل افراد کو ان کی انفرادی...

The Effect of Physical Activity on the Menstrual Cycle at Martial Art Putri at the University of Muhammadiyah Makassar

Menstruation is a factor that reflects a woman's functional potential which can be influenced by a number of variables including age, family history, socioeconomic status, education, physical activity, personality, exercise, weight, height, stress, infection, percentage of fat distribution and hormonal. Physical exercise or sports affect the menstrual cycle of the participants of  martial art Muhammadiyah. This study aims to determine the effect of physical activity on the menstrual cycle, namely female combatants in the martial art Muhammadiyah arts education. According to research conducted in a number of countries, including other developing countries, it is said that menstrual disorders are quite a problem faced by Hillard women and found that female students show more frequent problems with irregular menstruation.

Investigation of Nonlinear Dispersive and Dissipative Structures in Dense Plasmas

Quantum effects have been investigated in highly dense non relativistic, relativistic and ultra-relativistic plasmas. Various issues have been brought under consideration right from the Alfven solitary structure in non-relativistic, relativistic and ultra-relativistic electron-ion degenerate quantum plasmas. The full set of nonlinear QHD equations for a relativistic degenerate electron-ion plasma is used to investigate circularly polarized Alfven waves via the Zakharov equations. The solitary structures in non-relativistic and ultra-relativistic degenerate quantum plasmas are discussed by applying the Sagdeev potential technique. Also, we have studied obliquely propagating electromagnetic excitations in dissipative in the presence of relativistic and quantum effects. In this regard, relativistic quantum magnetohydrodynamic model and reductive perturbation technique is incorporated to study the linear and nonlinear magnetoacoustic modes in non-relativistic and ultra-relativistic electron-ion plasma. Also the effects of electron concentration, obliqueness and the magnetic field strength on the shock structures are investigated to both fast and slow modes.
