The focus of current research was to evaluate pharmacological ability of Acacia hydaspica methanol extract (AHM) and its derived fractions. Qualitative screening of methanol extract demonstrated the occurrence of terpenoids, anthraquinones, coumarins, cardiac glycosides, saponins, flavonoids, tannins, phlobatannins and alkaloids. GCMS chemical fingerprinting of AHM reveals the presence of 30 different chemical constituents belonging to diverse classes, owing enormous biological activities. HPLC analysis of AHM and various fractions reveals the presence of four well known standards; catechin, gallic acid, rutin and caffeic acid in AHM in varying concentrations. Catechin and Gallic acid were detected in both AHE and AHB while myrecitin was detected only in AHE. Mineral analysis indicated the existence of various macro and micromineral elements in A. hydaspica. AHB, AHE and AHM demonstrated high level of total flavonoid and phenolic content and a noteworthy correlation with the EC50 values was determined for the quenching of DPPH radical, hydroxyl radical, hydrogen peroxide radical, ABTS radical, iron chelating, β-Carotene bleaching inhibition and anti-lipid peroxidation. Significant anti-hemolytic, antimicrobial, cytotoxic actions were demonstrated by AHM and its AHE and AHB fractions. AHM and its fraction AHE reveal marked antipyretic, anti-inflammatory, analgesic and antidepressant potential. The in vivo studies indicated that AHE fraction of A. hydaspica possessed potent chemoprotective ability against cisplatin and doxorubicin induced toxicity in rats. Treatment of rats with AHE markedly improved the serum biomarkers of organ toxicity and tissue antioxidant status by significantly ameliorating the oxidative tissues markers enzymes levels near to control. Histopathological studies of different organs verify the biochemical observations. A. hydaspica was subjected to bioassaydirected fractionation which led to the isolation of three active antioxidant, antimicrobial and cytotoxic compounds from AHE and one active compound from AHB fraction besides active isolates. The compound structures were interpreted by ESI-MS and NMR spectroscopic analysis. Three AHE compounds were identified as 7-O-galloyl catechin (GC), (+) catechin (C), methyl-gallate (MG) while catechin-3- gallate (CG) was extracted from AHB fraction. In antimicrobial testing, MG was the most active compound. Isolated compounds GC, CG and MG showed more potent activity against DPPH, hydroxyl radical, nitric oxide radical, showed highest xviii antioxidant index and FRAP (649.5±1.5 μM Fe (II)/g) potency compared to standard reference rutin and gallic acid. In vitro models of prostate (PC-3) and breast (MDA-MB-231) cancer cells were used to evaluate the anticancer potential of isolated compounds (AHCs). AHC resulted in specific effects against tested prostate and breast cancer cell lines. GC, C, CG and MG inhibit cell proliferation in PC-3 cells in a dose dependent manner, whereas CG and MG negatively affected MDA-MB-231 cell growth. Compounds induce cell death via suppressing various signal transduction pathways that regulates cell proliferation and survival. Chromatin condensation, cell shrinkage and apoptotic bodies were observed by phase contrast microscopy. Compounds significantly inhibited cell survival and colony growth in both cell lines. Staining with acridine orange, ethidium bromide, propidium iodide and DAPI demonstrated that cell death occurred at least partly through induction of apoptosis in both PC-3 and MDA-MB- 231 cells. GC, C, CG and MG repressed the expression of anti-apoptotic proteins Bcl- 2, Bcl-xL and survivin in dose and time dependent manner, which futher validate the apoptotic effect of isolated compounds. Further analysis of signaling pathways indicated that compounds treatment induced a dose and time dependent suppression of JAK2, NFκB, p-Akt (Ser473and Thr308), NFκB p65 P-Ser 529, phospho-IκBa Ser32/Ser36, p-ERK1/2 (Thr202/Tyr204) in PC-3 cells. In MDA-MB-231 cells, CG and MG treatment significantly suppressed the expression of CK2α, PI3-K and JAK2. Expression of NFκB total protein and its phosphorylation at p65 P-Ser529 was significantly inhibited following treatments. Furthermore, Bcl-xL, Survivin and xIAP expression was also inhibited in MDA-MB-231 cells in a time and dose dependent manner. These findings provide strong indication that A. hydaspica compounds may be favorable therapeutic candidates against highly invasive triple negative breast cancer and androgen insensitive prostate cancer cells.

افسوس ہے کہ ہماری بزم احباب کی ایک شمع دل فروز اٹھ گئی یعنی مولانا یعقوب الرحمن صاحب عثمانی نے ۵۴ سال کی عمر میں ۱۰/فروری کواپنے وطن دیوبند میں انتقال کیا۔مرحوم دیوبند کے عثمانی خاندان سے تعلق رکھتے تھے اوراس تقریب سے مولانا مفتی عزیز الرحمن اورحضرت الاستاذ مولانا شبیر احمد عثمانی کے بھتیجے تھے۔ ذہانت وفطانت ان کا خاندانی ورثہ اورعلم وادب کے ساتھ لگاؤ ان کا آبائی ترکہ تھا۔ اسلامی علوم وفنون کی تکمیل دارالعلوم دیوبند میں کی اورہمیشہ اپنی جماعت میں ممتاز رہے۔ فراغت کے بعد دوایک سال یہیں معین المدرس رہے اور پھر حیدرآباد چلے گئے، جہاں پہلے عرصہ تک ورنگل کالج میں پروفیسر رہے اور اس کے بعد جامعہ عثمانیہ میں شعبۂ دینیات کے استاذہوگئے۔ آخرایک سال ہوا کہ یہیں سے پنشن لے کر وطن مالوف میں آبسے۔ عمائد دارالعلوم دیوبند نے مرحوم کی اہلیتوں اورصلاحیتوں کے پیش نظران کو جمعیۃ طلبائے دارالعلوم کانگراں کار مقرر کردیا۔اس عہدہ پرکام کرتے ہوئے ابھی چندماہ ہی ہوئے تھے کہ اجل کا پیام آپہنچا۔

This study was basically designed to explore the role of young companions of Holy Prophet (ﷺ) in the defense and development of Islamic state Madīnah. The approach applied for the collection and analysis of data was qualitative and descriptive. The review of literature uncovered that the young companions, progress the in role cardinal a played (صلى الله عليه وسلم) Muhammad Prophet of advancement, defense, and development of the society. The Holy Prophet young of education and training the towards attention special paid (صلى الله عليه وسلم) companion which enhanced their capacity and level of motivation and led them to work with enthusiasm. The kind behavior of our Prophet Muhammad (ﷺ) enabled them to transform their selves, their families, Meccah, Madīnah and the surroundings of the these sacred cities. Within the shortest possible period they spread the peaceful message of Islam and brought a positive change in the society and particularly established the peace in Madīnah. They built a strong defense of Madīnah from internal and external challenges and laid strong foundations for social, moral, economic and political development of the Islamic state Madīnah. It is, therefore, recommended to encourage the youth of Pakistan to make them realize their duties towards Pakistan and Islam so that they can play a significant role in the development and progress of Islam and Pakistan.

Prediction of Breast Tumor Size by Ultrasound: A Comparison With Pathology

Background: Tumor size is one of the most important prognostic factors in breast cancer and its accurate assessment is critical in planning appropriate treatment. Although histology is often regarded as the definitive way of establishing tumor size, radiological techniques, particularly ultrasonography, are usually relied upon to give an estimate of the tumor size for prognostication and treatment planning. Correlations between mammography, ultrasonography, magnetic resonance imaging (MRI) and pathology size have been performed in different populations with varying results however no prospective study has been done to establish the agreement between sonographic tumor size and histopathological tumor size in our population. Objectives: The main objective of this study was to determine whether or not there was a difference between the tumor size on ultrasound and tumor size on histopathology. The relationship between tumor size on ultrasound and tumor size on histopathology was also assessed. Methods: This was a prospective, observational survey. Women undergoing preoperative ultrasound for suspicious breast masses followed by lumpectomy or mastectomy were recruited into the study. The tumor sizes measured to the nearest millimeter on ultrasound and on histopathology were recorded side by side in a data sheet. Analysis: The mean difference between the two sets of measurements was assessed using a paired t-test. The paired measurements were also plotted on a scatter diagram and linear regression analysis performed to establish their degree of correlation. An equation for predicting the histopathological tumor size from the sonographic tumor size was given along with 95% confidence intervals. Outcomes: Ultrasound tended to underestimate the tumor size and the mean difference between ultrasound and histopathology was 4.3 mm (95% confidence interval of 0.8 mm to 7.8 mm; p-value of 0.0196). The equation y = 1.1758x + 0.126 best predicted the histopathology size from the ultrasound size although this did not differ significantly from the line of equality. Discussion: This study demonstrated that tumor size on ultrasound differed significantly from that on histopathology, similar to findings from previous studies. However, due to the wide variability in the size differences, it remains difficult to accurately predict the histopathological size for an individual tumor. Recommendations: Clinicians managing patients with breast cancer need to be aware that while ultrasound may be used to estimate tumor size, this estimate might differ significantly from the histopathological measurement.
