Present study was conducted on plant-microbe interactions in wheat (Triticum aestivum L.) under wheat-rice and wheat-cotton rotations. Soil samples were collected from rhizosphere of wheat and used for isolation of bacteria on LB medium as well as on selective media for phosphate solubilizers and nitrogen fixers. A total of 29 isolates were obtained and identified on the basis of 16S rRNA gene sequence analysis as Azospirillum (2 strains), Acinetobacter (2 strains), Actinobacteria (1 strain), Arthrobacter (3 strains), Bacillus (5 strains), Enterobacter (3 strains), Microbacterium (2 strains), Pantoea (one strain), Pseudomonas (4 strains) and one strain each of the genera Sphingobacteria, Terribacillus and Xanthomonas. In the present study, pqqE (a gene known to have a role in P-solubilization) was PCR amplified and sequenced. Sequence analysis of pqqE gene amplified from Arthrobacter sp. WP-2, Pseudomonas spp. T-27 and NN-4 showed maximum (80-81%) sequence similarity with that of Pseudomonas putida and that of Pantoea sp. WP-5 with Klebsiella (84%). Phosphate solubilization (a plant beneficial trait) activity was detected in 12 bacterial strains. Among the tested strains, high P- solubilization activity (207-311 μg/mL) was detected in the pure cultures of Arthrobacter sp. WP-2, Azospirillum sp. WS-1, Bacillus sp. T-34, Enterobacter spp. T-41 & T-42, Pantoea sp. WP-5 and Pseudomonas sp. T-27. The most efficient indol-3-acetic acid producing (11-31 μg/mL ) strains among the isolates were Bacillus sp. T-34, Enterobacter sp. T-41, Pseudomonas sp. WP-1, Arthrobacter sp. WP-2 and Azospirillum sp. WS-1. Selected bacterial isolates were tested as inocula for wheat grown in sterelized sand, in earthen pots (filled with non-sterelized soil) and finally evaluated in field trials under wheat-rice and wheat-cotton crop rotation. Maximum increase in grain yield over control was noted in plants inoculated with Azospirillum spp. WS-1 & WB-3 and Bacillus sp. T-34 under wheat-rice rotation (10-15%) and under wheat-cotton rotation (7-14%). Bacterial population determined at different growth stages of inoculated plants indicated maximum number of bacteria (108-109 cfu/g dry soil) at booting stage. Organic acids (acetic acid, citric acid, malic acid and oxalic acid) and sugars (sucrose and glucose) were detected in rhizosheaths of wheat grown under both crop rotations. Diversity of bacteria in the rhizosheath of wheat under both crop rotations was studied through direct soil DNA analysis of 16S rRNA using barcoded pyrosequencing. From the soil samples a total XIV of 46,971 cleaned sequences were obtained with read length of 319 bp. Among these sequences, 48.8% sequences were obtained from wheat-cotton rotation while 51.2% from wheat-rice crop rotation. Out of total 46,971 sequences, 11,729 (24.97%) showed 97% similarity with phylotypes having PGPR activity. The results showed that in wheat-cotton and wheat-rice rotation, Proteobacteria were dominant (25.1% and 35.7%, respectively), followed by un-classified bacteria (20.5% and 17.1%, respectively) and Actinobacteria (17.7% and 13.6%). However, Chloroflexi, Firmicutes, Acidobacteria, Planctomyctes, Bacteriodetes, Cyanobacteria, Verrucomicrobia and Nitrospora were also present. It was also observed that out of 495 different phylotypes detected, 280 phylotypes were common in both the crop rotations while 96 were only abundant in wheat-rice rotation and 41 were only present in wheat-cotton rotation system. Diversity of diazotrophs was determined in rhizosheath of wheat under both crop rotations by nifH sequence analysis amplified from soil DNA. A total of 41,287 nifH sequences were obtained with read length of 339-345 bp. The results showed that in wheat-rice and wheat-cotton cropping system, Proteobacteria were dominant (61.3% and 59.9%, respectively). However, sequences related to Cyanobacteria, Chlorobi, Firmicutes, Verrucomicrobia, Euryarchaeota, Spirochaetes, Actinobacteria and Fibrobacteres were also detected. Sequence analysis indicated presence of nifH sequences belonging to 150 different nitrogen fixing genera. Among these 150 genera, 22.6% genera were present only in wheat-rice rotation, 12.6% genera were found in wheat-cotton rotaion and 40.6% genera were present in both the cropping systems.

Development and Validation of Extended Multi-Dimensional Scale of Entrepreneurial Ecosystem in the Context of Pakistan

The purpose of this research is to validate the multi-dimensional scale of EntrepreneurialcEcosystem in the context of Pakistan. This research is based on 7 constructs with 54 items that affect the entrepreneurial ecosystem in any given region. The sample of 244 respondents are the owners of companies and, startups who participated in this research. The Confirmatory factory analysis showed factor loadings of all constructs greater than 0.40, while partial least square structural equation modeling showed acceptable values of construct reliability, composite reliability, however, average variance extracted was shown to be greater than 0.40 and less than the acceptable value of 0.5 for some constructs, while the HTMT ratio established discriminant validity of the constructs another criterion i.e. Fornell-Larcker criterion also established the discriminant validity of the constructs with some constructs having values less than 0.705, while some of the item outer loadings were found to be between 0.6-0.70 however, within the acceptable range. This research has validated the multi-dimensional scale of the entrepreneurial ecosystem with new sub-domain i.e. Support professions and support finance. This scale can be used to measure the strength of the entrepreneurial ecosystem of any region with appropriate homogeneous sample

Evaluation and Optimization of Mixed Wheat Flour in Relation to End Product Quality

Lack of soft wheat variety and classification of wheats according to end product, baking industry faces continuous problems with the stability and quality of products. The current research work was undertaken to characterize different (six) Pakistani wheat varieties for physicochemical, rheological, nutritional characteristics and to check multivariate association among these characteristics with special reference to cookie quality prepared from wheats of varying extraction rates (study-I). In study-II, different levels of chemical treatments (sodium stearoyl 2-lactylate (SSL), L-cysteine and Lecithin) and shortenings (fat, oil, and margarine) were applied on mixed wheat flour to choose best level on the basis of sensorial and physical characteristics of cookies. Furthermore, selected levels of chemical treatments, shortening and physical treatments (heat) were compared regarding sensory, physico-chemical and nutritional attributes of cookies (study-III). Significant variation among wheat varieties were observed with respect to particle size index (PSI), thousand kernel weight (TKW), test weight (TW), pearling value, water solvent retention capacity (WSRC), sodium carbonate solvent retention capacity (SCSRC) and dough rheological parameters. Fatty acids like, Stearic, Elaidic, Caprylic acids were observed abundantly in wheat flours, and comparatively better fatty acid profile was observed in “Punjab 2011” and “Galaxy 2013”. Cookies prepared from flours of varying extraction rate imparted significant variation on color and overall acceptability of cookies while interaction of wheat variety and extraction rate imparted significant (p<0.05) variation on taste and texture of cookies. In most of the sensory parameters, it was observed that cookies prepared from high extraction rate i.e. 72-74% (stream split flour) gained high score and among wheat varieties, cookies prepared from flour of “Millat 2011” attained better score than cookies prepared from flour of other wheats. Spread factor (SF) of cookies was correlated with PSI (r = -0.63), pelshenke value (r = -0.62), water absorption (WA) (r = -0.60) and mixographic peak height (MPH) (r = 0.85).Principal Component Analysis (PCA) showed that PC1 accounted for 30.68% of variation for MPH, SF, peak viscosity, breakdown viscosity and damaged starch being the major factors. While, PC2 accounted for 21.29% variation with flour rheological properties like dough development time, dough stability, pasting temperature, trough viscosity, falling number and TW were the major factors. Based on sensory and physical characteristics (diameter, thickness, spread factor and textural hardness), it was concluded that “T1” (SSL @ 0.5%), “T4” (L-cysteine @ 0.5%), “T9” (oil + margarine) and “T11” (lecithin @ 1%) provided better quality cookies when applied on mixed wheat flour during study-II. In study–III, significant deviation in physical parameters like diameter (3.96-4.66cm), thickness (0.99-1.21cm), spread factor (37.32-46.03), color like L* (58.99-70.01), a* (0.80-2.61), b* (15.36-22.37) and hardness (2.09-2.89) of cookies was observed due to selected treatments. Among these, cookies prepared from Lecithin (@ 1%) and SSL (@ 0.5%) got high score in sensory evaluation, in textural hardness cookies prepared with SSL showed better results and regarding spread factor of cookies best results were observed in SSL (@ 0.5%) and in lecithin (@ 1%). Among fatty acids of cookies, abundantly found fatty acids were palmitoleic acid, stearic acid, myristoleic acid and oleic acid. Highest amount of palmitoleic acid (9.84%) and stearic acid (8.81%) was observed in “T4” (Lecithin @1%) while least amount was observed in “T3” (oil + margarine) and “T1” (SSL @ 0.5%) respectively. Therefore, current study suggested that SSL @ 0.5% and lecithin @ 1% can be incorporated in cookie recipe to minimize variation (occurred due to mixed wheat flour) in quality attributes of cookies.
