This study shows that strain of thermotolerant Kluyveromyces marxianus was used for the production of ethanol and invertase (fructofuranosiadse, Ffase). This strain (D-67283) was collected from Shakkar Gunj Sugar Mills, Jhang, selected through Gamma rays on 1.5 % (w/v) deoxy-Dglucose (DG) in liquid medium after growth at 60 °C for 5 days and designated as K. marxianus M15. The selected mutant strain produced maximum ethanol and Ffase at 48 h of cultivation on different substrates including glucose, sucrose, and molasses each at 10, 12, 15 and 17% total sugars in 23 L fermentor (working volume 15 L). Optimized studies on different carbon sources displayed that product formation rate (Qp) was greater on glucose- followed by molasses- medium but was found to be lower on sucrose medium. Product yield (Yp/s) and specific product yield (Yp/x) were also significantly higher on glucose (15%) whereas found as lower on sucrose medium. Specific product formation rate (qp) was also recorded higher on glucose medium and remained lower on sucrose medium. Nitrogen sources like ammonium sulphate, corn steep liquor (CSL), and urea were added to the growth medium to enhance growth and ethanol formation. All these sources were used at the rate to contain 0.11, 0.16 and 0.21 % nitrogen in the growth medium. The best results were observed for fermentation kinetic parameters of growth and product formation by ammonium sulphate using 0.75% (w/v). In further studies, temperature of fermentation was optimized for maximum ethanol production, and substrate utilization, for both wild and mutant strains of K. marxianus. For this purpose, they were grown at different temperatures ranging from 20- 65 oC. The study further revealed that maximum ethanol production on molasses medium supplemented with ammonium sulphate was used 0.75% at pH 5.5 after 48 h at 40 oC. xx Effect of various agitation rates from 250-450 rpm on production of ethanol by K. marxianus cells was carried out and the maximum amount of ethanol produced on the sugar based used was more than 89.96 %. Further increase in agitation intensity did not increase ethanol production in both organisms. Hence, agitation rate of 300 rpm was optimized. The production of ethanol is an anaerobic fermentation process; therefore supply of oxygen to the yeast culture is of great importance as it is needed to support an initial amount of cell mass for maximum ethanol production. At a constant rate of oxygen supply, agitation rate supports uniform distribution of cells and maintain a constant temperature by uniformly stirring the media to dissipate excess heat. Effect of supplying air to the fermentor at different aeration rates (0.25 - 1.5 LL-1min-1) on ethanol fermentation, amount of ethanol produced was 72.5-74.8 gL-1 when the aeration rate was kept at 1.0 LL-1min-1 for 8 h and then between 0.25 - 0.50 LL-1min-1. Further increase in aeration rate, resulted in lower production of ethanol and greater amount of cell mass in the fermented broth but up to a certain extent. The maximum amount of ethanol (75 gL-1) produced when the aeration rate was kept at 0.30 LL -1min-1. Maximum ethanol specific or volumetric productivity increased with the increased temperature up to 40 oC and 45 °C in the case of wild and mutant respectively. The activation enthalpy for ethanol formation (DH* = 55.6 KJ/mol) pathway was lower than that for phytase production ((DH*70-80 KJ mol-1).The estimated values of enthalpy of Ffase formation (13.1 kJ mo-1l) network was lower as compared with the product inactivation (12.0 kJ mol-1 ) network. This usually happens in the case of thermotolerant organisms as reported earlier. Over all these studies revealed that mutant strain acquired significantly better changes in the genetic make up and qualifies for its evaluation at industrial scale ethanol production.

ہندوستان کے کہنہ مشق اور فارسی کے مسلم الثبوت شاعر حضرت گرامی نے ۲۶؍ مئی ۱۹۲۷؁ء کو چند روزہ علالت کے بعد اس دنیائے فانی کو الوداع کہا، مرحوم پنجاب کے ضلع جالندھر کے رہنے والے تھے، فارسی شاعری سے ان کو فطری لگاؤ تھا، کچھ دنوں امرتسر کے ایک اسلامی مدرسہ میں معلم رہے، پھر اعلیٰ حضرت نظام سابق مرحوم کی قدر شناس نگاہ نے ان کو تاکا اور اپنے دربار کا فارسی شاعر مقرر کیا، اخیر عمر میں حیدرآباد سے جالندھر آکر جب قیام کیا تو ان کی صحبت اور فیض اثر سے متعدد نوجوان اردو شاعر پیدا ہوئے، جن میں ابولاثر حفیظؔ اور سالکؔ کے نام سب سے اونچے ہیں، ڈاکٹر اقبال نے بھی جب سے فارسی میں کہنا شروع کیا، ان سے استفادہ میں دریغ نہیں کیا، زبان کے معاملہ میں وہ ان کی سند تھے، افسوس ہے کہ اب کشورِ ہند ایسے یگانہ نامور کے وجود سے خالی ہوگیا۔
مرحوم سے صرف ایک دفعہ آل انڈیا شعراء کانفرنس دہلی منعقدہ ۱۹۲۳؁ء میں ملاقات ہوئی تھی، بے حد ملنسار، متواضع اور مرنجان آدمی تھے، ایک سال پہلے تک ان کے اکثر خطوط میری عزت بڑھاتے رہتے تھے اور کبھی کبھی معارف کے صفحوں کو بھی اپنے نغموں سے معمور کیا کرتے تھے، مولانا شبلی مرحوم کے تعلق اور ان سے حیدرآباد کی یک جائی اور شاعری کی ہم پیشگی کا اثر یہ تھا کہ وہ مولانا مرحوم کی اس یادگار کو بزرگانہ محبت کی نگاہوں سے دیکھا کرتے تھے، افسوس کہ یہ فیض اب ہمیشہ کے لئے بند ہوگیا۔

TO COMPARE THE EFFECTIVENESS OF TRUNK STABILIZATION EXERCISE AND GENERAL EXERCISE IN NON-SPECIFIC CHRONIC LOW BACK PAIN-A COMPARATIVE STUDY

Aim of study: To identify the effectiveness of particular trunk stabilization versus a general exercise in low back pain management. Methodology: An experimental study was conducted at the physiotherapy department of Dow University of Health Sciences, 52 participants with low backache were enrolled and assessed for pain intensity using Visual Analog Scale (VAS) and disability by using the Modified Oswestry Low Back Disability Index (MOLBDQ-I). Through equal randomization one group got their low back pain treated through trunk stabilization exercises while the other with general exercises, 3times/week* 4weeks. Data were analyzed using SPSS version 25.0 by applying non-parametric Mann-Whitney U-test. Results: This study demonstrated that males and females are equally affected by chronic low back pain. Trunk stabilizing and general exercise regimes both significantly reduced the pain and disability in the study population but the effectiveness of trunk stabilizing exercises were significantly superior in reducing pain. Limitations and Future Implications: Study did not include a control group that received no intervention. It would be valuable to assess the cost-effectiveness of trunk stabilization exercises compared to general exercises or other interventions. Originality: Trunk stabilizing exercises are superior in reducing pain, disability, and restoring functional mobility than general exercises in chronic back pain. Conclusion: Trunk stabilizing exercises are superior in reducing pain, disability, and restoring functional mobility than general exercises in chronic back pain.

Genetic Divergence Estimation Through Agro-Morphological, Bio-Chemical and Molecular Markers in Sunflower Helianthus Annuus L.

Sunflower (Helianthus annuus L.) is an important oilseed crop. In Pakistan, it is the third biggest source of edible oil. Every year Pakistan spends huge amount of its foreign reserves on import of edible oil and now Pakistan is the third largest importer of edible oilseeds in world after China and India. Our local production has decreased over the years and now our local edible oil production is only the 13% of our demand. To bridge the gap between supply and demand of edible oil in Pakistan, government has to focus on Sunflower crop. For increasing the sunflower production new and improved local sunflower hybrids should be developed. Information of Genetic diversity study is a prerequisite for designing any breeding program. In the first experiment genetic diversity among 109 sunflower lines was estimated at morphological, bio-chemical and molecular level. For morphological characterization 109 sunflower lines were evaluated during Spring-2015 and Spring-2016 at National Agricultural Research Center, Islamabad following Augmented Design. The studied genotypes showed significant amount of variation on the basis of results obtained by cluster analysis and Principal component analysis. SDS-PAGE technique was used to study the variability among the sunflower genotypes at bio-chemical level. Genetic variability at molecular level was studied using SSR markers. Morphological and molecular based characterization showed considerable amount of genetic variation among the studied sunflower genotypes, while these genotypes showed less than 3% dis-similarity through SDS-PAGE based bio-chemical genetic study. The second experiment was for the identification of heterotic groups among the studied material. Heterotic groups were identified by combing the morphological and molecular data sets as, combining different data sets has been found useful in increasing the resolution of experiment. Two major clusters one of CMS and maintainer lines and other of restorer lines were observed. Six sub-groups in each major cluster were observed and one best performing genotype from each subgroup was selected. In third experiment, 12 parents (6 CMS and 6 Restorer) selected from heterotic groups was crossed in LxT mating design during autumn, 2016 to develop crosses and study the efficacy of identified heterotic groups and gene action. 36 crosses developed along with their parents and checks were sown in Randomized Complete Block Design (RCBD) at two locations (NARC and URF) during spring-2017. Three best performing hybrids (RHP-41 x CMS-HAP-56, RHP-71 x CMS-HAP- 111, RHP-71 x CMS-HAP-12) were found to be best for seed yield and oil contents over three check hybrids and four best combining parents i.e., CMS-HAP-12, CMS-HAP-56, RHP-41 and RHP-71 were also identified. Results of our study showed that heterotic grouping among sunflower could be developed and strengthened on the basis of morphological and SSR markers to pace up the hybrid development in Sunflower.
