HCV develops a chronic infection in humans, which ultimately leads to liver failure. Discovery of direct-acting antivirals (DAAs) has initiated the era of well tolerated medications. While these treatments are useful but still encounter certain limitations including drug resistance mutations, selective immune pressure and various side effects. Besides, no effective vaccine for the prevention of HCV infection is yet available. Therefore, for the development of efficient antiviral treatment, comprehensive knowledge of viral proteins characterization and pathogenesis is essential. The current study attempted to use integrated approaches to characterize the HCV major drug target proteins NS3/4A, NS5A and NS5B. Here, we provide a detailed analysis of the drug and immune driven variations among these viral proteins using systems virology and proposed a mechanistic insight highlighting the importance of these mutations on the therapeutic and immune response. In NS3/4A, DRMs such as A36V, Q80K, M175L, I132L, S138T, and R123T were observed in epitopes associated with HLAB*57, HLA-B*27, DRBl*ll04, and DRB1*0101 alleles. Within NS5A, DRMs such as L31M, Q30K/R, L28V, F28L, Q54H, and H58P were found in epitopes related to DRB1*0701, DRBl*ll04, HLA-A*68, and DRB1*0101 alleles. Similarly, DRMs including D168V, M423I, M419M, V494A, V499A, V138I, and I482T were frequently found in epitopes associated with DRB1*0101, DRB1*0701, HLA-B*57, HLAB*27, and DRB1*1104 alleles within NS5B. Among these alleles DRB1*0701, DQA*0201,DRBl*ll04, DRB1*0101, DRB*5701, DRB*5703, Cw*0102, DQBl*O301, HLA-B*57, HLA-A*03, HLA-A*68, and HLA B*27 are involved in HCV protection or clearance. Moreover, the efficacy of four prioritized drugs with no drug and immune driven variations, Danoprevir, Balaprivir, Narlaprevir, Samatasvir was compared with Sofosbuvir using in vitro analysis and highlight the significance of these drugs as more efficacious and potential therapeutic targets. This study also attempted to investigate the evolutionary conservation of these proteins (NS3/4A, NS5A and NS5B) via global consensus sequence profiling of all HCV genotypes (Thio et al.). This comprehensive analysis finds out many conserved drug targets and post translational modification sites (PTMs) that could be a target for the development of universal drug and vaccine. This study also aimed to propose a conserved pan genotypic multi-epitope vaccine by using structural modeling and epitope-epitope compatibility as a promising strategy to combat HCV infections, effectively. Multi-epitope vaccine construct was designed by using sixteen linear conserved epitopes, to induce better antigenic responses than a univalent subunit vaccine. Thus, surface-exposed, conserved and antigenic epitopes from the selected viral proteins were screened to design broad spectrum multi-epitope based subunit vaccines. While stable and substantial interactions were also observed with Toll-like receptor 3 and 8. This study showed that integrated lines offer various opportunities to amass the incomplete mystery of HCV biology in a meaningful way. It will increase our comprehension of how HCV roots liver diseases and how different hidden an unanticipated mechanism including immune driven variations could affect its therapeutic response. It also provides efficient screening stratagem to effectively extract worthwhile insights from multidimensional molecular datasets and helps in improving our understanding of the development of possible therapeutic targets against HCV

منشی میراں بخش جلوہ

منشی میراں بخش جلوہ انیسویں صدی کے ربع آخر میں سیالکوٹ میں اردو میں شعرو شاعری کرتے تھے۔ انجمنِ حمایتِ اسلام کے جلسوں میں شریک ہوتے ہوئے نظمیں پڑھتے تھے۔ آپ سراج الاخبار(جہلم) کے سیالکوٹ میں نمائندہ تھے۔ جلوہ کے پانچ شعری مجموعے گلشنِ نعت‘ جلوہ حق‘ تحفہ جلوہ‘ نوحہ جلوہ‘ دیوان جلوہ اور ایک نثری کتاب جو جلوہ کی شعری تصانیف کے بارے میں معلومات فراہم کرتی ہے۔ شائع ہو چکی ہیں۔(۵۳) جلوہ کی مذکورہ بالا کتب نایاب ہیں۔

مولانا عبد المجید سالک اپنی تالیف’’ذکرِ اقبال‘‘ میں جلوہ سیالکوٹی کے بارے میں لکھتے ہیں:

ایک شاعر منشی میراں بخش جلوہ سیالکوٹی تھے جو اکثر انجمنِ حمایتِ اسلام میں بھی آ کر نظمیں پڑھا کرتے تھے۔ نہ جانے کہاں سے شعر کہنے کی لت پڑ گئی۔ شعر کیا تھے پکوڑے تل لیا کرتے تھے۔ ان دنوں خزانے کے ایک کلرک اہلِ زبان تھے جلوہ صاحب ان کو اکثر شعر سنایا کرتے تھے۔ ایک روز انہوں نے تنگ آ کر کہا بھائی جلوہ تمہارے شعروں سے چھیچھڑوں کی بو آتی ہے۔ جلوہ صاحب تاؤ کھا کر شاہ صاحب کی خدمت میں حاضر ہوئے اور ان کو اپنے اشعار سنا کر پوچھا کہ یہ اشعار کیسے ہیں شاہ صاحب۔ شاہ صاحب سے مراد مولوی سید میر حسن ہیں ‘ نے فرمایا سچ پوچھتے ہو تو تم نے شعروں کا جھٹکا کر دیا ہے۔(۵۴)

میراں بخش جلوہ فن تاریخ گوئی میں مہارت رکھتے تھے۔ شاعرِ کشمیر منشی محمد دین فوق کے چچا منشی غلام محمد خادم کا بیٹا محمود فوت ہوا تو جلوہ نے کئی تاریخیں کہیں جن میں سے ایک یہ ہے:

مر گیا جلوہ جو خادم کا پسر                              نام تھا محمود اور تھانیک خو            

کیوں نہ خادم روئے سر کو پیٹ کر                 مل گیا ہے خاک میں

Issuing Fatwa is much important in the field of Islamic Theology. At least one of the contemporary famous three methodologies in the field of Fatwa for the mufti to adopt is necessary; as it leads mufti to extract ruling from the text of Holy Qur᾽ān or Sunnah of the Holy Prophet (ﷺ) and from what the Muslim Jurists have agreed upon. A thorough study of the book Fatāwā Ahl Ḥadīth has been conducted in this study in order to highlight the characteristics and main features which distinguish the method of the author, ‘Abdullāh Muḥaddith Rōpaṟi, a prominent scholar of his time in the main stream of Ahl e Ḥadīth, from other scholars of his time in issuing fatwa. The study approves that the author has adopted the depth has and (صلى الله عليه وسلم) Prophet Holy the of companions the of method understanding of the primary sources i. E. Holy Book Qur᾽ān, Sunnah and Ijmā‘, and secondary sources i. E. Qiyās and custom and vice versa and he has given best solutions to the matters posed to him at his time on the basis of textual and rational evidences which ultimately influenced people and made them to accept the author as an authority in his field.. His prominent work also tells us that he has expertise in the fields of Islamic literature, Islamic Jurisprudence, Ḥadīth and Tafsīr. Therefore, his book regarding fatwa has got admired by the scholars of the Subcontinent of all the main streams. Also in the court of Law in the country the book has been considered as a referencing book.

Analysis of Single and Two-Layer Coating Flows

Modern optical fibers require a double-layer coating on the glass fiber in order to provide protection from signal attenuation and mechanical damage. The most important plastic resins used in optical fiber and wires are plastic polyvinyl chloride, low- and high-density polyethylene, nylon, and polysulfone. One of the most important things that affect the final product after processing is the design of the coating die. This thesis presents the theoretical analysis of double layer optical fiber coating extrusion process inside a pressure type die. From the modeling of Single and Double-layer fluids in coated process, a number of guiding results in general may be identified. Generally, it is concluded that coating thickness is directly controlled by die design, boundary of the die, polymer used for coating the optical fiber and the dragging velocity of the optical fiber. The analysis concentrates on flow conditions around the die exit: particularly so on shear and strain rates, and identification of the influence of slip. It is analyzed that the bond between the polymer and the continuum can become stronger by applying high pressure gradient and using the shear thickening polymer for coating. Moreover, it is observed that the thickness of coated fiber optics increases with the increase in slip parameter, dilatant parameter, elastic and Deborah number, while the temperature distribution decreases with the increase of these quantities. Shear stress decreases with increase of slippage at the boundary of the die only for a limiting value of the boundary slip coefficient. It is also found that for , the thickness of coated wire increases and decreases for It is also established that as we increase lies in the range the distribution of temperature increases while, decreases when we increase the power law index in the domain . The flow rate, force on coated fiber optics and the thickness of coated fiber optics increases as the pressure gradient through the annular gap is increased. The force on the surface of coated fiber optics and thickness of coated fiber optics increases as the elasticity of the polymer increases. It is observed, that the elasticity of the liquid reduces the speed of flow. Furthermore, it is concluded that the elasticity of the liquid decreases the temperature within the region.
