Ajwain (Trachyspermum ammi) belongs to the family Umbelliferae, is commonly used in traditional, and folk medicine due to its carminative, stimulant, antiseptic, diuretic, antihypertensive, and hepatoprotective activities. Non-specific lipid transfer proteins (nsLTPs) reported from various plants are known to be involved in transferring lipids between membranes and in plants defense response. Here, we describe the complete primary structure of a monomeric non-specific lipid transfer protein 1 (nsLTP1), with molecular weight of 9.66 kDa, from ajwain seeds. The nsLTP1 has been purified by combination of chromatographic techniques, and further characterized by mass spectrometry, and Edman degradation. The ajwain nsLTP1 comprises of 91 amino acids, with eight conserved cysteine residues. The amino acid sequence based predicted 3D structure is composed of four α-helices stabilized by four disulfide bonds, and a C-terminal tail. The predicted model was verified by using different computational tools; i.e. ERRAT, verify 3D web server, and PROCHECK. The docking of ajwain nsLTP1 with ligands; myristic acid (MYR), and oleic acid (OLE) was performed, and MD (Molecular Dynamics) simulation was used to validate the docking results. The findings suggested that amino acids; Leu11, Leu12, Ala55, Ala 56, Val15, Tyr59, and Leu62 are pivotal for the binding of lipid molecules with ajwain nsLTP1.

ڈاکٹر محمد ایوب قادری سے میری دیرینہ ملاقات تھی، جب جب کراچی جانے کا موقع ملا، وہ بہت ہی عزیزانہ طور پر ملے، ہر ملاقات میں پہلے سے زیادہ اپنی بھلمنساہت، شرافتِ طبع، عجز اور انکسار کا ثبوت دیتے، بہت سی کتابوں کے مصنف ہوئے، جیسا کہ حسب ذیل مضمون سے ظاہر ہوگا، ان کے قلم میں بڑی برق وشی تھی، کسی کتاب کو لکھنا یا ترجمہ کرنا شروع کرتے تو بڑی کم مدت میں یہ کام ختم کردیتے، وہ جس بلند مرتبہ کے مصنف تھے، اپنے عجز و انکسار کی بدولت اس سے اپنے کو کم ہی ظاہر کرتے، وہ اپنی ہر کتاب دارالمصنفین ضرور بھیجتے اور اس کو یہ راقم شوق سے پڑھتا، جب انھوں نے کراچی کے قیام میں شاہنواز خاں کی تصنیف مآثر الامراء کی تین جلدوں کے ترجمے ہدیہ کیے، تو یہ تینوں جلدیں میری میز پر برابر رہیں، اور جب کبھی ان میں سے کسی اقتباس کو اصل فارسی سے ملایا، تو ان کو ہر طرح صحیح، سلیس اور فصیح پایا، اس سے ترجمہ کرنے میں ان کی مہارت اور قدرت کا معترف ہوا، جب وہ طبقات اکبری کا ترجمہ کررہے تھے تو ان سے یہ گفتگو آئی کہ اکبرنامہ کا ترجمہ ایک انگریز نے انگریزی میں کردیا ہے، لیکن یہ بڑی ندامت اور شرم کی بات ہے کہ اب تک اس کا اردو میں ترجمہ نہیں ہوسکا، گویہ بہت مشکل کام ہے، لیکن جب اس کا ترجمہ انگریزی میں ہوسکتا ہے، تو کوئی وجہ نہیں کہ اردو میں نہ ہوسکے، جناب ایوب قادری صاحب نے کہا کہ طبقات اکبری کے ترجمہ کے بعد انشاء اﷲ اکبرنامہ کی جلدوں کا ترجمہ کرکے لوگوں کی ندامت کو دور کردوں گا، اس سے مجھ کو بڑی خوشی ہوئی، گزشتہ نومبر میں میرے قیام کراچی ہی...

Islamic Perspective of Governance and Judiciary: A Literature Review

The aim of this research is to accumulate the literature related to an Islamic ideology for two of the basic components of a State, Governance and Judiciary; both have a strong impact on society. Pakistan is an Islamic ideological state, the only Islamic rule created on the name of religion after the state established by the Prophet Muhammad (Peace be Upon Him) in Medina. Its constitution and the style of governance should have its foundation based upon the principles of Islam; this is the main concept behind the creation of “Islamic Republic of Pakistan”. The paper will review some of the basic requirements for an administrative system to be qualified as Islamic way of governance and the judicial system in Islam includes the formulation of a comprehensive ideology of human equality and justice; the creation of a powerful incentive to adopt this ideology; and to establish a living example of equality and justice at all levels of life by defining the Islamic Judicial System & Its Legal Basis. Different Quranic verses, sayings and acts of Muslim jurists in this perspective are also studied. The paper has literature review in two major parts, first we will cover governance and Islam along with some basic terminologies from Islamic point of view and in the later part Judiciary, the concept of Justice and Judicial system in Islam will be discussed.

Physiological and Molecular Studies of Tomato under Salinity Stress

The aim of the present investigation was to determine the effect of salt stress on the physiology of twelve genotypes of tomato (Solanum lycopersicum L.), to study the genetic diversity among these genotypes and to transfer salt tolerant gene in the selected genotype through Agrobacterium mediated gene transformation method. The seeds of different tomato genotypes were obtained from Ayub Agricultural Research Institute (AARI), Faisalabad and from Horticulture Research institute (HRI), NARC, Islamabad. First experiment comprises study of plant growth, ionic content, ABA content of shoot and root and proline content at four different NaCl salinity level. The genotypes Avinash, NTH242 and Gol performed better at higher salt concentration as indicated by less inhibition of plant height, shoot and root fresh weight and shoot and root dry weight. There was less translocation of Na + to shoot but greater accumulation of K + and Ca 2+ concomitant with higher production of osmoregulant (proline) and endogenous ABA accumulation in shoot was recorded. Second experiment was conducted to study the genetic diversity among the twelve tomato genotypes through randomly amplified polymorphic DNA analyses. Twelve tomato genotypes were grouped in four clusters at similarity coefficient value of 0.88 on the basis of randomly amplified polymorphic DNA analysis. Cluster-I consisted of a single tomato genotype “NTH-242”. Cluster-II of three tomato genotypes Lyp No.1, BARI, Roma and Cluster-III grouped Pakit and Riogrande together. Cluster-IV composed of maximum tomato genotypes i.e. 88572, Gol, Nagina, Checo, Pant Bahar and Avinash. Third experiment deals with Agrobacterium mediated transformation of DREB 1A gene in tomato using tissue culture. Prior to transformation three tomato genotypes were subjected to tissue culture studies to select the best genotype for callus induction and regeneration. Hypocotyls and leaf discs were used as explant source and inoculated on Murashige and Skoog media having different hormonal combinations. The callus induction and regeneration frequency varied among genotypes and it was concluded that hypocotyls of tomato genotype Avinash produced maximum callus induction and regeneration frequency on MS media xiiisupplemented with IAA (0.5 mg/l) and Kin (1.5 mg/l). Agrobacterium mediated gene transformation was carried out in tomato genotype Avinash to transfer DREB 1A gene tolerant to salt stress. During the study it was observed that two min infection time with Agrobacterium, two days for co-cultivation, Acetosyringone @ 50 μM in co-cultivated media and 50 mg/l hygromycin in selection media was optimum for high frequency transformation. Polymerase chain reaction (PCR) analysis was conducted to confirm trasfomants by amplification of 500 bp size DNA fragment for hygromycin resistance gene.
