CMCases are important enzymes that can hydrolyze lignocellulosic mass efficiently. Apart from this, CMCases are used in industries pertaining to textile, food processing, wine and brewery, pulp & paper, animal feed, agro based products, detergents, waste management, olive oil extraction and carotenoid extraction. The aim of this study is to explore the diversity of the indigenous thermophilic bacteria that are capable of degrading cellulosic biomass. A total of thirty-seven bacterial strains were isolated from stove ash samples. All the isolates were Gram positive and on the basis of physico-biochemical characterization identified and named as Bacillus sp TLW-1 to TLW-37. Plate screening was performed for ten different industrially important hydrolytic enzymes. Almost all the isolates were able to produce all the tested enzymes (except pectinase) with different profile. Positive CMCase producers were quantitatively screened by shake flask method. The isolate Bacillus sp. TLW-3 showed the highest CMCase production and was selected for further study. Isolate was identified on the basis of 16S rDNA sequencing as Bacillus licheniformis TLW-3. Sequence was submitted to NCBI (with KY440432 accession number). According to the growth curve, the isolate entered into log phase after 6 hrs of incubation and the log phase ended after 30 hrs incubation. Afterwards, the stationary phase began which ended after an incubation period of 72 hrs then the decline phase started. Highest enzyme production was found during the stationary phase. Optimization of production and growth conditions was performed by using two techniques i.e. OFAT and RSM. According to the OFAT approach, the highest growth and CMCase production were obtained in the medium containing 1% CMC, 1% peptone and 0.5% yeast extract with pH 7.0 at 50˚C and 150 rpm after the incubation period of 72 hrs. When compared with RSM optimization, the best growth was found at 70˚C in the medium having 0.5% peptone, 2.3% yeast extract and 2.52% CMC with pH 9.0 after 49.2 hrs of incubation. RSM optimized production conditions for highest CMCase production was found to be 1.8% peptone, 1.4% yeast extract and 1.6% CMC with pH 7.0 at 70˚C and 69.63 hrs of incubation period. Co-production of the enzyme was obtained in the medium containing 1% CMC and 1% starch as the sole carbon source. For the purification purpose, various precipitation and chromatography techniques were employed. CMCase was successfully (partially) purified with 8.250-fold and 13.75 % yield. Molecular weight of CMCase was estimated to be 28 kDa. Enzyme kinetics of partially purified CMCase demonstrated that enzyme was found active in the reaction mixture containing 1% CMC with pH 7.0 and at temperature 70˚C in the presence of Co+2, Ca+2 and Hg+2 ions. When the glucose was added in the reaction mixture enzyme activity was inhibited. The stability conditions of CMCase include, pH 6.0 and at temperature 4-50˚C; however, the stability was totally lost in the presence of the tested metal ions. The Vmax, Km and activation energy for CMCase were found to be 42.553 IU/ml/min, 9.587 mg/ml and 2.649 KJ/mole respectively. Endoglucanase gene was amplified, sequenced (11888 bp) and submitted to NCBI (with MF953225 accession number). Expasy-protparam online server was used for amino acid translation and in slicio physico-chemical characterization of the sequence. Secondary and tertiary structure of the protein revealed using SOPMA and ITASSER online server respectively.

Enlightened Message of Sufism Towards Peaceful Pakistan

In today’s world mankind seems to be skidding towards chaos and violence. Mankind is divided on the lines of faith, race, sect, colour, cast, language and creed. Odium and intolerance have become the orders of the day. This condition pushes to take the refuge of religion but the religious radicalism, intolerance and discrimination are also in the fold. In this situation Sufi message comes to give shelter for peace, love and humanity. The enlightened Sufi message is not for one nation, race, or community but it is for the whole of humanity. Pakistan has faced terrified situations due to dangerous trends of religious extremism, radicalism and social intolerance. In order to understand this problem, the concept of Sufism has been applied. This has been used as an approach / methodology to evade from hated and violent tendency to reorient Pakistan back on the track to peace and prosperity. Sindh and Khyber Pakhtunkhwa provinces of Pakistan have been taken as case study to give the empirical evidence. This study demonstrates that the enlightened message of Sufism has enough potential to bring back peaceful environment.

Adaptation, Modification and Development of Norms of Hand Test in Pakistan

The present study aims to Adapt, modify and develop norms of the Hand Test in Pakistan. A sample of 500 participants (270 males & 230 females) age ranged from 11 to 80 years (M=34.44 & SD=17.34) was recruited from various cities of Pakistan. The sample belonged to various economic classes which includes earnings from up to 20,000 to above 1 lac Pakistani Rupees. Participants were further bifurcated into four groups which include 350 normal who were further divided age wise into four groups. Group one age ranged from 11 to 20 years. Group two age ranged from 21 to 40 years. Group three age ranged from 41 to 60 years. Group four 61 years and above. The second group consisted of 50 maladjusted. The third group consisted of 50 neurotic/anxiety related disorders who were categorized into generalized anxiety, obsessive compulsive and phobic. Whereas, the fourth group consisted of psychotics who were further categorized into schizophrenic, major depression and bipolar. Cross sectional research design was implied, purposive sampling technique was used for data collection. The study was carried out phase wise, in phase one scoring booklet of Hand Test (Wagner, 1983) was translated from English into Urdu language using back translation method, comprising fourteen stimuli (10 original and Four adapted). A pilot study was carried out to adapt four new stimuli out of a set of 10 on the criterions (a) their ability to generate responses in more scoring categories (b) varied range of responses and these were placed from stimuli nine to thirteen. Licence to use the Modified Hand Test was obtained from Western Psychological Services. In phase two reliability and validity studies were carried out to establish psychometric properties of the Modified Hand Test with promising results test-retest reliability (r =.39 to .83), inter-scorer reliability (r =.87), split- half reliability was determined while dividing stimuli into odd and even categories and it was found in the range of α=.04 to .84 for failure (in neurotic/anxiety related disorder group) to bizarre (in psychotic group). Construct Validity came out for interpersonal with environmental (r =.43). Predictive validity of socially desirable traits with academic grades remained moderate. In phase three normative data, percentile ranks and z scores were computed for various scoring categories which facilitated in determining normal ranges of scores across group of samples for age groups and gender for normal group. Qualitative analysis based on content was computed through frequency counts; certain new areas were explored like religiosity, oppositional, introjection, projection need for altruism and affiliation. One-way analysis of variance (ANOVA) was computed for different groups of participants, age groups and economic classes for normal group. Results were significant across main domains as well as sub categories at .01 and .05 level of significance. Analysis of results depicted that the Modified Hand Test provided confirmation to normative data and studies reported by its author and other researchers. Furthermore, it explored new dimensions of personality.
