Growing perception concerning diet and health has extended the need to exploit the nutritious, biologically active and sustainable food products. The present study was designed to evaluate the potential role of water-soluble peptides (WSPs) extract derived from buffalo and cow milk Cheddar cheeses with special reference to anti-hypertensive, anti-thrombotic, anti-oxidant, anti-inflammatory and anti-cancer activities. Purposely, the WSPs fractions collected at different stages of cheese ripening were subjected to assess the degree of proteolysis, peptides profiling by RP-HPLC and in vitro assays for bioactivities of interest. The ripening period was accompanied by a significant (p<0.01) increase in the level of water-soluble nitrogen and non-protein-nitrogen. The cheeses expressed peptide peak development during ripening, however few qualitative differences were observed in chromatograms of their WSPs extract. There were slight differences concerning the number, area and height of peaks during whole investigation period. The angiotensin converting enzyme- inhibitory and anti-thrombotic activity of WSPs increased progressively during 180 days of cheese maturation. Regarding the in vitro radical scavenging potential, the WSPs extracts were demonstrated to be potent scavenger of DPPH and ABTS radicals and the activities increased progressively with ripening period. Moreover, results indicated the dose-dependent inhibition of radicals. Comparatively, the highest activities were noticed in the peptide extract obtained from buffalo milk Cheddar cheese. The protective role of WSPs extract on cell viability and cellular antioxidant activity in RAW-264.7 macrophages (LPS-stimulated) and Caco-2 (tert-butyl hydroperoxide-induced) cell lines, indicated that the intracellular ROS production decreased significantly upon treatment with the selected doses of WSPs extract. However, maximum activity was noticed at 150 and 180 day of ripening in a dose-dependent manner. The in vitro anti-inflammatory potential of cheese extract was expounded using RAW-264.7 macrophages on the basis of nitric oxide (NO) production in these cells. The extracts effectively inhibited the NO production in cells without affecting their viability. The cell viability, cell cycle arrest and apoptosis were performed to explore the anti-cancer activity using lung (H1299) and colon (HT-29 & HCT-116) cancer cell lines. Cheese extracts of 120, 150 and 180 of ripening days showed marked anti-proliferation activity towards cancer cells in dose-dependent fashion. The extracts also caused significant changes in cell cycle distribution in comparison to the control cells. The substantial dose-dependent increase in the percentage of cells population in G0/G1 phase was observed in colon cells while WSPs extracts induced G2/M phase cell cycle arrest in lung cancer cell line at rate of 400 μg/mL and 500 μg/mL. Moreover, these extracts also induced extensive early and late apoptosis in all cancer cells. In conclusion, the promising health potential of Cheddar cheese can offer a perspective to reduce the risk of disorders associated with cardiovascular diseases, oxidative stress, inflammation and cancer.

Return Migration to Pakistan during COVID19 Pandemic: Unmaking the Challenges

In order to contain the spread of corona virus (COVID-19) disease, strict border closure measures have been taken globally. Migrants and refugees are affected across the globe due to such measures. Amid the COVID-19 pandemic, the economic recession escalated across the globe which is expected to have serious implications for the migrant workers and laborers. Potohar region was selected to explore implications of return migration from abroad, but the fieldwork halted immediately due to the pandemic outbreak. Since the data on Pakistani migrants is dispersed and scant, it was nearly impossible to stick to the locale for returnees. Therefore, the data for this study was collected qualitatively using exploratory methodology. In-depth interviews were conducted using interview guide as a tool of research. The paper is based on analysis of the narratives based on the experiences of the returnees especially. It sheds light on the state of the stranded Pakistani migrants who have been returning or awaiting repatriation due to layoffs. It delves into long-run and short-run challenges due to return migration, in Pakistan which highly depends on remittances from abroad. Challenges created by sudden end to remittances, entrepreneurial setups and returnees’ reintegration in the society need immediate attention. It is recommended that the state should support and monitor the migrants living abroad, ease out issues in repatriation of the laid off workers, pave way for investment, offer insurance plans, reduce reliance on remittance flow and engage them in Public Private Partnerships for sustainable reintegration.

Low Frequency Perturbations in Inhomogeneous Plasmas The waves and instabilities produced by inhomogeneous currents J 0z (x) in plasmas along the initially constant magnetic field are investigated. The total zero- B order magnetic field is twisted and becomes space dependent . In heavier ion (like barium) plasmas the ion dynamics can be ignored in the presence of sheared flow of electrons. Electrostatic current-driven wave with the frequency near / (where | ln / | , / and is the perpendicular wavenumber) can exist in such plasmas and it becomes unstable under certain conditions. The nonlinear equations for this mode have been derived and stationary solutions in the form of vortices and solitons have been obtained. Due to sheared flow of electrons and ions in the presence of stationary dust, a low frequency electrostatic current-driven drift like wave with real frequency / has also been proposed to exist in dusty plasmas. The nonlinear equations for this wave including the effects of density gradient have been derived. The effects of kappa distribution of electrons have also been investigated. D''Angelo''s mode is modified in the presence of superthermal electrons. In the nonlinear regime the wave can give rise to dipolar vortex structures if the shear in flow is weaker and tripolar vortices if the flow has steeper gradient. The results have been applied to Saturn''s magnetosphere corresponding to negatively charged dust grains. The very low frequency wave at dust time scale can also be produced by the sheared currents if dust is assumed to be dynamic. These very low frequency waves and instabilities with small growth rates can be important in the planetary and cometary magnetospheres due to the interaction of solar wind. In the nonlinear regime these waves give rise to electrostatic structures such as vortices and solitons. A comparison of these very low frequency current-driven dust waves with the current-driven ion waves is also presented to point out analogous physical pictures with different temporal and spatial scales. The theoretical model is a general one and here it is applied to Saturn''s magnetosphere for illustration. ix The global drift mode is also studied in a plasma bounded in a cylinder having Gaussian density profile. The effect of magnetic shear on the wave propagation along density gradient is studied in a Cartesian geometry assuming absorbing boundary. It is found that the wave amplitude is reduced when two-ion species are present (with the same concentration) compared to pure electron-ion plasma.
