Photodynamic therapy (PDT) is an emerging treatment modality that employs the photochemical interaction of three components. These are light of suitable wavelength, photosensitizer, and singlet oxygen. The main objective of this work is the determination of pharmacokinetics of different photosensitizers after laser irradiation in different biological samples like cell lines and tissues to improve the efficacy of PDT. Present study investigates the dynamic behaviour of different photosensitizers under laser irradiation e.g. (a) biodistribution of Photofrin® (b) laser induced effects (c) depth of necrosis under exposure of different wavelengths of light sources (d) synergistic effect of toxicity of ZnO nanostructures bare and conjugated with photosensitizers like Aminolevulinic acid (5-ALA), Photofrin® and protoporphyrin dimethyl ester (PPDME) determined for the treatment of localized cancer cells. Firstly, biodistribution of Photofrin® into Sprague Dawley rat has been investigated by methodology for radiolabelleing of Photofrin® with labelling efficiency of Photofrin® with 99m Tc. In addition, it was explored that 99m Tc is more than 95%. The current technique is simpler and efficient as compared to the earlier published protocol. In the second part of this experimental study, different photosensitizer‟s uptake was explored in the in vitro model e.g. different cell lines (HepG2, RD, Hep2C, AG1518 and FM55P). Study of optimal dose of laser, cellular totoxicity and phototoxicity were also the part of our experiment. The third aspect of study relates to in vivo study, normal rat liver were treated as biological sample, PDT under illumination of different wavelengths of light (630 nm, 660 nm, 600 nm and its alternate combinations) has been performed and proved that effectiveness of 630 nm of wavelength is more efficient for depth of necrosis as compare to other wavelengths of light source. Finally, the synergistic effect of toxicity of zinc oxide nanorods (ZnO NRs) conjugated with 5-ALA, Photofrin® and PPDME was determined for the treatment of localized cancer cells. Cell toxicity due to microinjection and free standing drug delivery was observed by detection of reactive oxygen species (ROS) liberation, and verified by MTT assay. It is successfully demonstrated that UV-A irradiation increased toxicity and caused significant production of ROS which leads to cell necrosis within few minutes. Zinc oxide nanorods are toxic for both normal as well as malignant cells under exposure of ultraviolet (UV) containing 240 nm of wavelength.

افسوس ہے کہ گذشتہ مہینوں میں دنیائے علم و ادب کی دو نامور شخصیتیں پروفیسر محمد الیاس برنی اور خلیفہ عبدالحکیم ہم سے جدا ہوگئے، پروفیسر الیاس برنی علی گڑھ کالج کے نامور فرزند تھے، حصول تعلیم کے بعد کالج ہی میں معاشیات کے لکچرار ہوگئے تھے، پھر دارالترجمہ کے رکن کی حیثیت سے حیدرآباد چلے گئے اور کچھ دنوں تک اس کے ناظم بھی رہے، پھر جامعہ عثمانیہ میں معاشیات کے استاد مقرر ہوگئے، اور اسی عہدہ سے سبکدوش ہوئے، تعلیم و تدریس کے ساتھ تالیف و تصنیف کا سلسلہ بھی برابر جاری رہا، جامعہ عثمانیہ کے لیے معاشیات کی کئی کتابیں لکھیں، اس فن کو اردو میں سب سے پہلے ان ہی نے روشناس کیا تھا، ان کی پہلی تصنیف علم المعیشت ایک زمانہ میں بہت مشہور تھی۔ شعر و ادب کا مذاق بھی رکھتے تھے، معارف ملت، جذبات فطرت اور مناظر قدرت کے نام سے کئی حصوں میں اردو نظموں کا ایک نہایت عمدہ انتخاب مرتب کیا تھا جو بہت مقبول ہوا۔

Considering the vast importance of time management in the light of discipline, the following article throws a great deal of light on it in the Islamic perspective.

Analysis, Inhibition and Degradation of Mycotoxins in Foodstuffs

Mycotoxins are toxic natural food contaminants, mostly produced by moulds of genera Aspergillus, Penicillium and Fusarium. Cereal grains are the most susceptible food material amenable to contamination by mycotoxins. Current study adopted a multiprong approach to develop a better understanding of this major public health concern as well as devise strategies for management and control of cereal mycotoxin contamination. Initially, a comprehensive screening for aflatoxins and ochratoxin-A in various cereals from Pakistan was conducted. The results showed that 43% of samples were found contaminated with Aflatoxins. Furthermore, multi-mycotoxin contamination profile of Pakistani cereals (rice, maize and wheat) has shown prevalence of aflatoxins, trichohecence, fumonisins, zearalenone, sterigmatocystin, roquefortin-c and ochratoxin-A in cereals. The study examined the reduction of 35-51% of AFB1 during cooking of cereals following typical Pakistani recipes. The dietary exposure values for aflatoxins have exceeded the tolerable daily intake at all levels. The margin of exposure of AFB1 was remarkably lower than the recommended safe limits. The mean cancer risk values for adults and children were 0.1613 and 0.266 cancer cases/year /105, respectively. Surveillance of aflatoxins (AFs) contamination in different maize varieties can contribute to the identification of susceptible and resistant varieties, particularly for cultivation in the Pakistani agro-climatic environment. Among tested five short-season maize varieties (Pioneer, Neelam, DK-919, Desi and Hi-Corn), maximum aflatoxin levels were found in Desi (14.5±0.02 μg/kg), while Hi-corn variety demonstrated significant resistance to A. flavus and aflatoxin contamination. Myco-flora from contaminated cereal posing high risk (rice) was isolated and identified. Among the five isolated fungal genera, Aspergillus was found to be the most dominant genus. Eight essential oils were studied to inhibit A. flavus and its toxigenic potential associated with rice. The essential oils of Cuminum cyminum and Eucalyptus citriodora exhibit the maximum anti-fungal and anti-aflatoxigenic activity. The study also investigated that 71-88% of aflatoxin B1 degraded in real food medium at minimum xvii thickness (2mm) of sample exposed to UV-light after 4 hr and 48 hr of sun light. The toxicity of degraded products was much less than that of untreated samples. This study will contribute significantly to manage the mycotoxin problem in Pakistani foodstuffs.
