Image processing is being successfully applied in many areas medical research such as computer aided diagnosis, tumor imaging and treatment, angiography, and carotid artery plaque detection. For medical image analysis, segmentation is an intermediate step to segregate region of interest from the background. The ultimate goal of segmentation is to identify the part of the data array that makes up an object in the real world. Many imaging modalities are in practice for disease diagnosis. Among those, owing to noninvasive nature, ultrasound imaging provides an invaluable tool for disease diagnosis. Major limitations faced by ultrasound imaging modality include low quality, inherent noise, and wave interferences. Consequently, a substantial effort from radiologists is required to extract constructive information about a particular disease. In this regard, an efficient and accurate computer aided diagnostic system for ultrasound images is highly desirable for disease (plaque) diagnosis. Carotid arteries are vital arteries that supply oxygen rich blood to the brain. Carotid artery stenosis is the process of narrowing the carotid artery due to the presence of atherosclerosis. The plaque may partially or fully block the blood flow to the brain and the probability of cerebrovascular stroke becomes high. Ultrasound imaging is used for detection of plaque in carotid artery. Due to lower quality and other degradations, segmentation of carotid arteries ultrasound images becomes a challenging task. In this thesis, several segmentation techniques are proposed, which successfully segment the carotid artery ultrasound images. Firstly, we have proposed spatial fuzzy c-means modified (sFCMM) clustering technique and also investigated effectiveness of ensemble clustering. The proposed sFCMM technique assigns weight to each pixel in a sub-window according to the pixel’s contribution. The proposed scheme required image pre-processing for noise reduction and hence segmentation has been performed on filtered image. In another approach, we propose information gain based fuzzy c-means clustering (IGFCM) algorithm that avoids the preprocessing step and still yields better results compared to sFCMM technique. The IGFCM approach exploits the concept of information gain to automatically update the xvii fuzzy membership function and cluster centeriods. However, from IGFCM segmented images, it has been observed that some of the pixels of arterial walls are mislabeled by IGFCM. In order to overcome this problem, a semi-supervised clustering approach named robust segmentation and classification of ultrasound images (RSC-US) has been proposed to segment carotid artery ultrasound images. The RSC-US approach is composed of three phases. In the first phase, the fuzzy inference system (FIS) is generated. In second phase, carotid artery ultrasound images are segmented based on the generated FIS. Finally, a decision making system has been designed to segregate the segmented images into normal or abnormal subjects. The RSC-US approach did not utilize the spatial information of pixel’s which plays a vital role in segmentation. Consequently, the spatial information has also been explored and a new approach named robust fuzzy radial basis function networks (RFRBFN) has been proposed to segment carotid artery ultrasound images. The RFRBFN segments the carotid artery ultrasound images with high precision. Due to the Lagrange function and a smoothing parameter, the RFRBFN might be computationally expensive. Finally, an automatic active contour based segmentation technique for carotid artery ultrasound images is proposed. This technique can successfully segment natural scene as well as medical images.

Implementation of Gaussian Process Regression in Estimating Motor Vehicle Insurance Claims Reserves

This study aims to calculate the allowance for losses by applying Gaussian Process regression to estimate future claims. Modeling is done on motor vehicle insurance data. The data used in this study are historical data on PT XYZ's motor vehicle insurance business line during 2017 and 2019 (January 2017 to December 2019). Data analysis will be carried out on the 2017 - 2019 data to obtain an estimate of the claim reserves in the following year, namely 2018 - 2020. This study uses the Chain Ladder method which is the most popular loss reserving method in theory and practice. The estimation results show that the Gaussian Process Regression method is very flexible and can be applied without much adjustment. These results were also compared with the Chain Ladder method. Estimated claim reserves for PT XYZ's motor vehicle business line using the chain-ladder method, the company must provide funds for 2017 of 8,997,979,222 IDR in 2018 16,194,503,605 IDR in 2019 amounting to Rp. 1,719,764,520 for backup. Meanwhile, by using the Bayessian Gaussian Process method, the company must provide funds for 2017 of 9,060,965,077 IDR in 2018 amounting to 16,307,865,130 IDR, and in 2019 1,731,802,871 IDR for backup. The more conservative Bayessian Gaussian Process method. Motor vehicle insurance data has a short development time (claims occur) so that it is included in the short-tail type of business.

Synthesis and Applications of Piperazine, Bis-Indole Derivatives and Gold Nanoparticles of Thiolated Ligands

This dissertation has been divided into four chapters and each chapter has its own numbering of compounds and references. General introduction related to the importance of heterocycle based drugs, drug designing and value of lead molecules in drug designing. This research work describes synthesis and bioactivities of different class of heterocycles such as piperazine, bisindole analogs in search of important therapeutic agents and also includes functionalized gold nanoparticles for catalytic study. During this research study, a variety of piperazine, and bisindole analogs were synthesized and screened for enzyme inhibition studies (thymidine phosphorylase, urease, antioxidant, β-glucuronidase and α-glucosidase, anti-Leishmania). Functionalized gold nanoparticles were synthesized for modulating their catalytic ability. The results obtained from this study are encouraging which are discussed separately in the forthcoming chapters 1, 2, 3 and 4. In first chapter, piperazine analogues are studied. We have synthesized eighteen piperazine derivatives (1-18) and evaluated for thymidine phosphorylase, urease, antioxidant, b-glucuronidase and α-glucosidase activity. All analogues showed potent thymidine phosphorylase inhibitory potential with IC50 values ranging between 0.2 ± 0.01 to 42.20 ± 0.70 μM when compared with standard 7-Deazaxanthine (IC50 value of 38.68 ± 1.12 μM). Compound 17 was found to be the most active among the series while compound 18 was found to be the least active. All these analogues showed varying degree of urease, antioxidant, b-glucuronidase and α-glucosidase activity. xi In second chapter, bis-indole analogues are studied. We have synthesized twenty seven bis-indole analogues (1-27) and evaluate their anti-leishmanial, urease, antioxidant, and b-glucuronidase activity. All compounds showed outstanding anti-leishmanial inhibitory potential with IC50 values ranging from 0.7 ± 0.01 to 13.30 ± 0.50 μM respectively when compared with standard pentamidine with IC50 value of 7.20 ± 0.20 μM. All analogues showed greater potential than standard except 23 when compared with standard. All these compounds showed varying degree of urease, antioxidant, b-glucuronidase and α-glucosidase activity. In third chapter, functionalized gold nanoparticles are studied. We report the fabrication of a family of nanozymes comprised of bioorthogonal ruthenium catalysts inserted in the protective monolayer of gold nanoparticles. By introducing simple modifications to the functional groups at the surface of the nanozymes, we have demonstrated control over the kinetic mechanism of our system. Cationic nanozymes with hydrophobic surface functionalities tend to replicate the classical Michaelis Menten model, while those with polar groups display substrate inhibition behaviour, a key mechanism present in 20% of natural enzymes. The structural parameters described herein can be used for creating artificial nano-systems that mimic the complexity observed in cell machinery. In fourth chapter we discussed the different bioassay protocols in detail.
