The purpose of this study was to understand the impact of the Visiting Teacher (VT) Mathematics secondary program by inquiring into the pedagogical approaches of selected teachers in Karachi. Furthermore, the challenges they encountered during the implementation of the new instructional strategies in the classrooms were studied. The field work was undertaken in three of Aga Khan University, Institute for Educational Development (AKUIED)' s co-operating schools, from which two male and one female VTs had attended the program in 1998. The data was gathered from VTs, headteachers (HTs), colleagues, students, and course tutors through interviews, observation and documents within a qualitative paradigm. The major findings of the study were that the VT program in Mathematics (secondary) had impacted on VTs to at least some extent, resulting in increased student participation in the classroom. The VT program also contributed to the raising of awareness about the subject of Mathematics and the teaching and learning processes associated with it. This study revealed that prior to joining the program VTs had a lack of conceptual understanding of the subject matter, as well as a lack of awareness of new pedagogical approaches. After the program it was generally found that the teachers were employing new pedagogical approaches. These were found by them to be useful in order to enhance understanding of the concepts to be taught by relating them to life experiences. The program also helped the teachers to develop thinking skills and taught them to place an emphasis on meaningful learning. They developed reflective practice and had enhanced their course content knowledge but punishing students was found to be a common practice. An important finding was that VTs had made a major shift in their perception of teaching and learning and moved from a traditional mode of teaching towards progressive instructional approaches. Challenges faced by VTs in the implementation of new approaches were discussed. In order to improve the program, some recommendations have been proposed.

ظفر اقبال27 ستمبر، 1932ء کو بہاولنگر ، پاکستان میں پیدا ہوئے۔ ان کے والد چک نمبر 49/3Rضلع اوکاڑہ کے ایک معزز زمیندار تھے۔ ظفر اقبال نے ابتدائی تعلیم بہاولنگر سے حاصل کی اور میٹرک ایم سی ہائی اسکول اوکاڑہ سے 1950ء میں کیا۔ انٹرمیڈیٹ کا امتحان ایف سی کالج لاہور اور بی اے گورنمنٹ کالج لاہور سے کیا۔ ظفر اقبال نے ایل ایل بی کا امتحان لا کالج جامعہ پنجاب سے پاس کیا۔
وکالت کا امتحان پاس کرنے کے بعد انہوں نے اوکاڑہ کچہری میں پریکٹس شروع کر دی۔ وہ ایک بار اوکاڑہ ایسوسی ایشن اور دو مرتبہ پریس کلب اوکاڑہ کے صدر بھی رہے۔ اس دوران انہوں نے قومی سیاست میں بھرپور طریقے سے حصہ لیا۔ 1977ء کے انتخابات میں ظفر اقبال نے نیشنل عوامی پارٹی کی طرف سے راؤ خورشید علی خاں (پاکستان پیپلز پارٹی کے نامزد امیدوار) کے مقابلے میں الیکشن لڑا لیکن کامیاب نہ ہو سکے۔

پرائمری کے دوران ان کی طبیعت شاعری کے لیے موزوں ہو چکی تھی، کیونکہ ان کے استاد نور احمد انجم قریشی جو خود بھی شاعر تھے بچوں کو بطور املا اشعار لکھ کر دیتے۔ ظفر اقبال آٹھویں جماعت تک کلیات میر اور دیوانِ غالب کا بھرپور مطالعہ کر چکے تھے۔ شفیق الرحمن کی تحریریں پڑھ کر ان کے اندر لکھنے کی تحریک پیدا ہوئی۔ انہوں نے غزل کے پیرائے میں فنی اور موضوعاتی سطح پر روایت شکنی کے حوالے سے اپنی ایک الگ اور بھرپور پہچان بنائی۔ اْن کے پہلے شعری مجموعے آب رواں کو عوام اور خواص، ہردو حلقوں میں بے حد پزیرائی ملی۔ اس کے بعد انہوں شعری تجربات کا سلسلہ نہ صرف جاری رکھا بلکہ اسے بام عروج تک پہنچایا۔ 1973ء میں انہوں نے پہلا کالم سرور سکھیرا کے پرچے دھنک کے لیے لکھا۔ ان کے مختلف اخبارات میں...

Islam is considered to be the religion of nature due to its originality and suitability to human nature for all times or centuries. It continues to be successful in the developed world, and elsewhere, because its call is in accordance with the fitrah or natural inclinations of mankind. Allah, subhanahu wata'aala, is the creator of mankind and therefore knows his nature more intricately than mankind himself. Allah has therefore chosen for us a religion best suited to the nature of mankind, a religion that goes neither to the extremes of hardship nor of laxity, but instead provides a middle path; in other words, a religion of ease. Islam does not lay on people tasks that they cannot do or they will have difficulty in doing. Whenever, there is any difficulty performing any religious obligation faced by Muslims, they have provided an element of ease and comfort. In this regard, the focus of this paper is to throw light on the notion of ease and leniency of Islam.  

Effect of Site-Directed Mutagenesis on the Efficiency of the Amylase Gene of B. Licheniformis

Bacillus α-amylases are widely used in industrial processes. Nevertheless, there is a need to improve the temperature tolerance of α-amylases as high temperature conditions are used in many industrial processes. In this study, various isolates of Bacillus licheniformis (B. licheniformis) were recovered from soil using potatoes (Solanum tumberosum) and sweet potatoes (Ipoema batatas) for baiting. Isolates were identified using common morphological techniques and biochemical tests with an API kit. B. licheniformis isolates for further study were selected on the basis of high α-amylase production. The α-amylase gene was cloned in plasmid pTBSG1 and characterized. Mutations to be introduced for enhancing α-amylase stability were predicted by in-silico design using various computational tools: Rosetta, the disulfide design server and the RosettaVIP tool. The disulfide design server identified five double mutants for mutagenesis to cysteine residues as follows: L64C/K80C, E119C/S130C, G433C/G474C, T139C/W157C and Y396C/D416C. A sixth mutant, V283I, was predicted with the RosettaVIP tool. The various mutants were prepared with the Phusion site-directed mutagenesis kit with the help of modified primers. Wild-type and mutants were then recombinantly expressed in E. coli BL21 (DE3) and characterized and compared to wild-type enzyme. Wild-type and mutant enzymes were pre-incubated at 45 ºC to 85 ºC and residual activities were found to not vary significantly till 55 ºC (P-value > 0.05). On the other hand, at 65 ºC, 75 ºC and 85 ºC, mutant V283I showed significantly improved thermostability (P-value < 0.05). Among the double mutants, G433C/G474C was found to have highest thermostability in terms of thermal inactivation (P < 0.05). This study shows that the rational design and introduction of substitution mutations leading to the formation of disulfide bonds and increased hydrophobic interactions has high potential for improving the thermal stability of α-amylases.
