Teacher absenteeism is a big dilemma in many educational institutions in Pakistan. The current work intends to extend our understanding of the causes of teacher absenteeism and the effects of absenteeism on students? performance at public secondary schools in district Kech (Turbat), Balochistan, Pakistan. The main purpose of the research was to find out the causes of teacher absenteeism and the effects of absenteeism on students? performance at public secondary schools of district Kech, Balochistan. This research adopted descriptive survey design to collect data from 10 headmasters, 5 headmistresses, 147 teachers and 646 students of Grade 9th and 10th from the public secondary schools of district Kech, Balochistan. For this research, 10 questionnaires for headmasters, 5 questionnaires for headmistresses and 147 for teachers were used as research instruments. In addition, 646 questionnaires for students of class 9th and 10th were also used as research instruments. and document analysis were used as research instruments as well. Reliability of the questionnaires was tested and enhanced coefficient of .876. Stratified sampling technique was used for selecting 10 males and 5 female public secondary schools out of 60 public secondary schools of district Kech, Balochistan. Random sampling technique was used for the selection of students and the purposive sampling technique was used to select teachers. Data were analyzed using both qualitative and quantitative methods. The quantitative data were coded and analyzed using descriptive statistics of frequencies and percentage in (SPSS) version 23.0. The study revealed that the teacher absenteeism has negative effects on students? performance. The study established that teachers illness, lack of regular supervision and assessment, high workload, teachers strike, teachers unions, other administrative duties, poor salaries, private business, attending test or interviews of other jobs, job dissatisfaction, lack of visit in schools by higher concern authorities, long distance or far from district education offices, lack of interest and non-availability of in-service teachers training, political approach ,poor infrastructure are the key causes of teachers absenteeism. In addition, this study also established that teacher absenteeism has negative effects on students? performance. The given course of the students cannot be completed. Low-grade marks in annual exams, the high dropout rate of students, violence among students in the classroom, poor performance, slow learning process, low quality of education are the effects of teacher?s absenteeism on students. This is also supported by this research that when a teacher is absent from the classroom, students learning is disrupted. When the teacher is repeatedly absent, students? performance can be significantly affected in a negative way. The results of the analysis also support the previous research findings that higher teacher absenteeism leads to lower student scores on standardized tests. The present study recommends that in order to reduce teacher absenteeism there is need to address the factors causing teachers absenteeism effectively. This research established that regular supervision, handsome salaries, good and conducive environment, adequate teachers, good infrastructure, guidance and counseling program, in-service teachers training can reduce the rate of teacher?s absenteeism in public secondary schools of district Kech, Balochistan. Moreover, Future teacher absenteeism studies also need to be conducted to identify the key determinants with more explanatory variables and computing the teacher absenteeism of the different level of schools in the entire Balochistan and in other parts of the country.

شیخ الحدیث مولانا ابوالحسن عبیداﷲ رحمانی ۵؍ جنوری کو رحلت فرماگئے، اناﷲ وانا الیہ راجعون۔ ان کے نام سے میں بچپن ہی میں آشنا ہوگیا تھا، میرے والد مسلکاً اہل حدیث ہیں، وہ جریدۂ اہل حدیث (امرتسر) اور رسالہ محدث اور اس مسلک کے بعض دوسرے رسالوں کے خریدار تھے، محدث مولانا نذیر احمد رحمانیؒ کی ادارت میں دارالحدیث رحمانیہ دہلی سے شایع ہوتا تھا، اس میں فتاویٰ اور مضامین مولانا عبید اﷲ رحمانی کے بھی برابر چھپتے تھے، میں ۱۹۴۷؁ء میں پرائمری درجات میں پڑھتا تھا، اس وقت ’’محدث‘‘ میری سمجھ میں کیا آتا؟ تاہم اسے پڑھنے کی کوشش ضرور کرتا، ایک روز والد صاحب نے اسے الٹتے پلٹتے دیکھا تو فرمایا کہ ’’میں تمھیں اسی مدرسہ میں پڑھنے کے لیے بھیجوں گا جہاں سے ’’محدث‘‘ شایع ہوتا ہے‘‘۔ مگر افسوس

Fashionable dressing is a very sensitive issue for females, it creates sometimes confusion that what are the limits and orders of “Shariah” for it. So I try to inform all females a proper dress code in the light of Islamic “Shariah”. Islam is not against the fashion but it says that it should be only for “Mahrams” and it should not be out of limits. So the article deals to clarify needs and importance of dress, dress codes in Islam as well as the usage of different type of dressings like thin, fitted, expensive and costly, male dresses, uneven (not according to Islam) etc. It will clarify the confusion which makes us confused in fashionable dressing and how much it is allowed to keep them in use. Islam has provided guidance in dressing like in any other fields of life as well as fashion is allowed by Allah as blessing but according to the rules and regulation of Islamic “Shariah” and do not try to go against it. That is why we have to be aware and careful while fashioning.

Inhibition of Mitochondrial Respiration Induces Paused Pluripotent State in Mouse Embryonic Stem Cells

Embryonic stem cells are the pluripotent cells that act as a root of embryonic development to engender the specialized cells of the body. This property can be captured in vitro to cultivate indefinitely, providing a remarkable model to study early development and diseases. In vitro, mouse pluripotent embryonic stem cells (mESCs) exist in two different states, naïve and primed. The naïve state mimics in vivo inner cell mass of blastocyst with the pluripotency genes expressed more uniformly and have more developmental potential than the primed state. The primed state corresponds to a later embryonic developmental stage than the naïve state. Both states of cells express similar level of some key pluripotency genes but the signaling pathways that support their self-renewal and pluripotency are strikingly different. Mitochondrial metabolism, among others, is a major difference between the two states. The primed state has more mature mitochondria but doesn’t use them for ATP generation, which is generated solely through glycolysis. The naïve state however, has a bivalent metabolic state, i.e. uses both glycolysis and mitochondria to generate ATP. What role mitochondria play in these two states remains a very interesting and relevant question. Here in this study, we investigated the function of mitochondrial respiration in naïve mESCs by blocking the electron transport chain (ETC) with specific pharmacological inhibitors, Rotenone and Antimycin, and genetically with shRNAs against mitochondrial specific transcription factor A (TFAM) and a helicase, Twinkle. Both inhibitors and shRNAs resulted in blockade of cell proliferation and when released, the cells resumed self-renewal without affecting the pluripotency. The paused phenotype by ETC inhibition also appeared in the blastocysts cultured in vitro, which naturally die within 2-3 days of the development while Rotenone treatment extends survival of blastocysts for 3 more days. Moreover, the treated blastocysts were able to give live pups when injected back into surrogate mother, confirming a normal developmental potential. Upon ETC inhibition with either compounds or shTFAM, the mitochondrial mass and membrane potential increased and level of reactive oxygen species (ROS) decreased. The shRNAs against TFAM and Twinkle greatly reduced the mitochondrial DNA (mtDNA) copy number and expression of mtDNA encoded mRNA, leading to complete blockade of mitochondrial respiration. Paused mESCs stopped oxygen consumption and enhanced glycolysis instead to maintain a steady level of ATP generation, which is essential for the cells to be alive. Mechanistically, ETC inhibition induced pause is distinct from the pause induced through either mTOR or Myc inhibition. Neither pyruvate, aspartate nor nucleosides supplementation could rescue the self-renewal of mESCs which were shown to have the ability in other cell types upon ETC inhibition. The total non-targeted metabolomics showed that the ETC regulates the carbohydrates, proteins, lipid and nucleic acid metabolism. Specifically, lysosomal-related pathways are found to be significantly and commonly changed from both metabolomics and proteomic analyses. We could confirm the blockade of autophagy and lysosomal pathways in ETC inhibition-induced paused cells. These analyses suggest that an intimate link exists between mitochondria and lysosome in mESCs and highlights the potential important function of lysosomes for mESCs self-renewal. This work reported a new type of pause by ETC inhibition in mESCs and identified some unique metabolic roles of mitochondrial ETC for mESCs self-renewal. Furthermore this study first time shows the effect of Ursolic acid on mouse embryonic stem cells.
