Cardiovascular complications, such as myocardial infarction, atherosclerosis, cardiomyopathy and heart failure are amongst the major causes of morbidity and mortality among diabetic patients. Diabetic cardiomyopathy (DCM) is a progressive disease of the heart muscle mediated by hyperglycemia and oxidative stress. Endothelial progenitor cells (EPCs) have shown promising potential to repair the diabetes induced cardiac damage. EPCs differentiate into functional and mature endothelial cells. However, challenges such as deteriorated functional abilities and survival of EPCs derived from diabetic subjects limit the possible outcome of autologous EPCs based therapies. Diazoxide (DZ), a highly selective mito-KATP channel opener, has been shown to improve the ability of mesenchymal stem cells and skeletal myoblasts for the repair of damage to heart muscles. In the present study, effect of DZ preconditioning was evaluated in determining the ability of diabetes affected bone marrow-derived EPCs to repair streptozotocin (STZ) induced DCM in rats. Diabetic EPCs (DM-EPCs) were characterized by immunocytochemistry, flow cytometry and reverse transcriptase PCR for endothelial markers: vWF, VE Cadherin, VEGFR2, PECAM, CD34 and eNOS. Preconditioning was performed in vitro, by incubation of DM-EPCs in 200 μM DZ for 30 minutes, followed by H2O2 induced oxidative stress and hyperglycemic stress. Oxidative stress to preconditioned and non preconditioned DM-EPCs was induced by exposure to 200 μM H2O2 for 2 hours whereas for hyperglycemic stress induction, the cells were exposed to 30 mM glucose media. Non-preconditioned EPCs with and without exposure to H2O2 were served as controls. IX The treated cells were evaluated for survival (XTT cell viability assay), senescence, paracrine potential (by ELISA for VEGF) and alteration in gene expression (VEGF, SDF-1α, HGF, bFGF, Bcl2 and caspase-3). DZ preconditioning of DM-EPCs significantly increased survival accompanied by enhanced release of VEGF and reduced senescence in DZ DM-EPCs+H2O2 group as compared to DM EPCs+H2O2 group under oxidative and hyperglycemic stress conditions. Furthermore, DZ preconditioned DM-EPCs exhibited upregulated expression of prosurvival genes under oxidative stress (VEGF, SDF-1α, HGF, bFGF and Bcl2) and under hyperglycemic stress (VEGF and Bcl2) while down regulated the expression of Caspase-3 in DZ DM-EPCs+H2O2 group as compared to DM-EPCs+H2O2 group under oxidative and hyperglycemic stress conditions as determined by reverse transcriptase PCR and real time PCR. For in vivo studies, diabetes was induced in Wistar rats by injecting 40 mg/kg streptozotocin. After eight weeks, 2x106/70 μl of serum free medium, each of DZ preconditioned DM-EPCs and non-preconditioned DM-EPCs were transplanted into the left ventricle (LV) of diabetic rats (n꞊6 rats per group). After four weeks, DZ preconditioned DM-EPCs transplanted group showed improved contractility of diabetic heart as compared to untreated DM-EPCs group. There was decrease in collagen content estimated by Masson''s trichrome and Picrosirius red staining in DZ DM-EPCs transplanted group as compared to untreated DM-EPCs transplanted group. Furthermore, reduced cells injury were observed in DZ DM-EPCs group as evidenced by decreased expression of caspase-3 and increased expression of prosurvival genes Bcl2, VEGF and bFGF by semi-quantitative real time PCR. X In conclusion, the present study demonstrated that DZ preconditioning enhanced DM-EPCs survival under oxidative and hyperglycemic stresses and their ability to treat DCM.

عقل سے ماورا ہو کر کسی کو چاہے کا نام عشق ہے۔ عشق محبت کی انتہا ء کو کہتے ہیں، جہاں محبت کی انتہا ہوتی ہے وہاں سے عشق کی ابتداء ہوتی ہے۔ کوئی کسی سے مالی منفعت کے حصول کے لیے عشق کرتا ہے، کسی کی محبت کی انتہا سیم وزرکے لیے ہوتی ہے، کسی کاعشق دنیاوی غرض و غایت کے لیے ہوتا ہے۔ کسی کے جسم و جان سے اُٹھنے والی محبت کی مہک جیسے ہی خواہش کی تکمیل ہوئی ، ختم ہوتی ہے۔
صدرِذی وقار!
عشق و محبت کا معیار ہر ایک کا اپنا ہی ہوتا ہے۔ شاگرد استاد سے جب عشق کرتا ہے تو اس کی حصول علم کے راستے میں آنے والی جملہ رکاوٹیں ختم ہو جاتی ہیں۔ اس کے دماغ کے آنگن میں علم وحکمت کے پھول کھلنا شروع ہوجاتے ہیں، اس کے گلستانِ علم و ادب میں بہار آجاتی ہے ، مرید جب اپنے پیر سے محبت کرتا ہے تو اس کے لیے سلوک کی منازل آسان ہوجاتی ہیں۔

If you have a relationship with the past, a sustainable plan can be done for future. Islamic personalities, especially the introduction of the scholars, intensify the path of progress in the field of scientific and research as well as the continuous hard work in the practical world. Similarly, when introduction of the people with historical roles have reached to the next generation, it is easy to define pathways and take lessons from the opponents to work hard for them. Introducing individuals with a specific region is worth a king's ransom investment for all its geographically entrusted and external roles and a leading rule for a new generation of races. In the underlying article, efforts are being made to present brief introduction of the scholars of the District Haripur who have written in various sciences and fictions, so that people of this region can know that this area also remained the base of Islamic scholars. Due to this article, depicting the introduction of the scholars of different school of thoughts will undermine an enormous and sectarian ideology, and the strength of tolerance and brotherhood will be strengthened.

The Dilemma of Energy Security: Political and Institutional Hazards in Case of Pakistan 1999-2013

„Energy security‟ is a unique blend in the realm of International Relations. The later part of the term has dominated the entire sphere of Realist theories of International Relations. Security has been the prime objective of state and fulcrum of its existence. Theories of Terrence and Deterrence, Power and Balance of Power have strived to bring about the required security for the nations. In recent times, it has been explored that security is just a meaningless phenomenon unless tools to acquire sufficient security are absent from the menu of the „elements of power‟. The oil embargo of 1970s has revealed the genuine position of energy resources amid the multiple strategies of regional and global power politics, tools and tactics. Originally energy security has emerged out of this catastrophic instance. Energy security theory is quite nascent and its contours are yet under development. The Realist theories and their tenants have contributed in shaping the concept to. Classic Realism and its off-shoot theories believe in a nation‟s self-interest and supremacy of the state. Liberalists and Neo-liberalist stand for collective security, competitive environment and mutually beneficial cooperation. Institutionalism stresses upon the legalization of IR and environment of economic development. Theory of energy security is an amalgamation of all basic tenants of Realist theories. The application of Energy Security theory provides a wide range of competitive and beneficial horizon where states can secure not only their own security but also contribute to the collective security parameters. But this application of the theory varies from region to region. A congenial political environment may give it a viable shape of cooperation and non-congenial environment may turn it into a hostile competitive phenomenon. South Asian region presents some interesting case studies of energy insecurity. Pakistan is one such example that has been engulfed in the energy insecurity since its inception. Although some issues related to the division of Sub-continent can be held responsible for the energy inefficiency but in the later part of its political history subtle miscalculations, negligence and mismanagement have added in the prevailing energy deficiency turmoil. There has never been one long-term, well-planned and coherent energy policy. Switching the balance of energy mix from hydropower to thermal power, delays in the construction of mega storage dams, political non-consensus and institutional disarrays have ruined the basis of energy sector. According to the Realist perspective, it was required to secure state interest and indulge actively in securing and utilizing most of the energy resources but most of the natural endowments have been mismanaged severely. After concluding IWT with India, there was no logic left in showing negligence towards hydropower management sector. There could have been better conditions if the hydro resources were accurately managed. Likewise, thermal resource management could have been done in a better way. Natural Gas utilization, coal mines exploration, oil sector policies and civil use of nuclear energy required some serious and in-depth plan. Amid the grim thermal resource management, creation of IPPs proved additionally troublesome in the long run. Evasion of long-term policies and adoption of piecemeal policies for frittering away the situation ruined the basis of energy sector management. Disintegration of WAPDA and non-coherent institutional set up has messed up the entire management of energy sector. Energy security is conditional with the domestic policies at one hand and dependent on the regional and global cooperation on the other hand. In this regard South Asian region has been lagging behind. Pakistan‟s share in the regional energy trade is quite low. Entire region including Pakistan is in dire need of viable energy cooperation and technological coordination under the umbrella of regional organization SAARC. Beside domestic energy sector management, unless the issues of all South Asian states on the regional and global front are resolved, attainment of energy security for this region shall be a far cry. In case of Pakistan, it is indispensible to form a prudent policy and a realistic execution. Unless hydro and alternative energy resources are fully utilized, thermal resources are well calculated, institutional efficiency is acquired, political consensus over key energy decision is attained, regional energy trade parameters are utilized and global aspirations are met, no energy security can be attained in the true letter and spirit.
