The research work presented in this thesis deals with the chemotherapeutic (antiproliferative) and cancer chemopreventive activity of a medicinal plant from Pakistan; Fagonia cretica L. (Zygophyllaceae), based on the traditional claims for its use in folklore medicine in the treatment of different types of cancers and inflammatory conditions. Through a bioactivity guided fractionation approach, the crude extract (FCC), resultant two organic fractions i.e. n-hexane and ethyl acetate (FCN and FCE, respectively) and an aqueous fraction (FCA) were tested for their anti-proliferative and cancer chemopreventive activity by employing different standard bioassays. In this investigation, it was demonstrated that F. cretica extract and fractions have antiproliferative activity towards human breast cancer cells and it can exert cancer chemopreventive effects via NFκB inhibition, iNOS inhibition and QR induction. The most potent fraction was ethyl acetate (FCE) fraction followed by n-hexane fraction (FCN) in this study. Six compounds in total (FCEE1, FCEK2, FCEK3, FCEM4, FCEN5 and FCNC6) were isolated from potent fractions of this medicinal plant using repeated flash column chromatography. Structural elucidation was carried out through a series of spectroscopic experiments (1-D and 2-D NMR). Among these six compounds, one compound (FCEE1) is a triterpene acid, two compounds (FCEK2 and FCEN5) belong to the triterpene saponin class, one compound FCEM4 belongs to steroidal saponin class and two compounds (FCEK3 and FCNC6) belong to phytosterol class of chemical compounds. All the isolated compounds have known structures but these are isolated from this plant species for the first time. All the compounds were evaluated for their anti-proliferative activity against breast cancer cell lines (MCF-7 and MDAMB- 231), non-tumorigenic breast epithelial cell line (MCF-10A) and cervical cancer cell line (HeLa) by using sulforhodamine B (SRB) assay. Cancer chemopreventive potential of compounds was also estimated in different bioassays i.e. TNF-α activated nuclear factor-kappa B (NFκB) assay, inhibition of lipopolysaccharide (LPS)- activated nitric oxide (NO) production in murine macrophage RAW 264.7 cells (iNOS) assay, Aromatase inhibition assay and Quinone reductase 1 (QR1) induction assay. A serial dilution of eight concentrations (0.39, 0.78, 1.56, 3.12, 6.25, 12.5, 25, 50 μM) for each compound was tested. xix Purified compounds were able to reduce cell viability significantly in two phenotypically different breast cancer cell lines (MCF-7 and MDA-MB-231). This activity was markedly reduced in normal mammary epithelial cells (MCF-10A) thus indicating selective toxicity between malignant and normal cells. Among compounds, in NFκB inhibition assay, FCEE1 showed maximum inhibition (IC50, 5.26 μM) while in iNOS inhibition assay, FCEM4 demonstrated best activity (IC50, 0.38 μM). For QR1 induction, FCNC6 was proved to be best compound (CD, 17.07 μM). None of the compounds were active in inhibiting aromatase enzyme. Compound FCEE1, a triterpene acid was the most potent compound in this study followed by FCEM4. Elucidation and characterization of the cytotoxic mechanism was undertaken by cell cycle status, apoptosis and expression of transcription factors and their targets i.e. p53, p21, Bax, γ-H2AX and FOXO3a. Pure compound treatment was able to induce a time and dose-dependent decrease on cell viability of two phenotypically different breast cancer cell lines (MCF-7 and MDA-MB-231), while demonstrating a markedly decreased cytotoxic effect of triterpene acid (FCEE1) and derivatives i.e. triterpene saponins (FCEK2 and FCEN5) on normal mammary epithelial cells (MCF-10A). Cell cycle arrest (expressed by sub-G1 peak) and apoptosis was induced in both MCF-7 and MDA-MB-231 cells. Overall all the purified compounds were more effective against estrogen receptor negative breast cancer (MDA-MB-231) cells. Loss of breast cancer cell viability is associated with induction of DNA double strand breaks which were detected in both cell lines by pure compounds’ treatment. In MCF-7 cells, expression of p53 by FCEE1 seems to induce upregulation of the CDK inhibitor, p21, and pro-apoptotic Bax. In current study, pure compounds’ treatment to human breast cancer cells induced double strand breaks to DNA associated with activation of DNA repair protein γ-H2AX. Compound FCEM4 treatment was able to induce FOXO3a expression in both MCF-7 and MDA-MB-231 cells. This suggests that activation of FOXO3a in the absence of functional p53 plays a vital role in induction of cytotoxicity. The results of this study suggest that FCEE1 is cytotoxic through p53 dependent as well as independent pathways while FCEM4 uses only p53 independent pathway.

ہاشم شاہ(۱۷۵۶ء۔۱۸۲۱ء) میں موضع جگد یوکلاں تحصیل اجنالہ میں پیدا ہوئے۔ آپ کے والد حاجی محمد شریف اپنے عہد کے پیرِ طریقت تھے۔ ہاشم نے انھی کے ہاتھ پر بیعت کی۔ آپ کے والد مختلف علوم اور زبانوں کے ماہر ہونے کے علاوہ حکیم حاذق بھی تھے۔ مہاراجہ رنجیت سنگھ جب ان کے علاج سے صحت یاب ہوا تو خوش ہو کر تھرپال نزد ریہ نارووال ان کو انعام میں دیا۔ ہاشم تا دمِ مرگ ۱۸۲۱ء یہیں رہے(۶) سرور ارمان نارووال کے شعرا کے تذکرے میں ہاشم شاہ کے بارے میں رقمطراز ہیں:

ہاشم شاہ نے فارسی‘ پنجابی اور اردو میں شاعری کی مگر ان کی شہرت کا بڑا سبب ان کی لکھی ہوئی عشقیہ داستاں سسی پنوں ہے۔ جس سے ان کا شمار پنجابی کلاسیکی شعرامیں ہوتا ہے۔ اس کے علاوہ ہاشم شاہ نے سوہنی مہینوال‘ شیریں فرہاد‘ لیلیٰ مجنوں ‘ ہیر رانجھا اور دوہے بھی پنجابی زبان میں لکھے۔(۷)

ہاشم شاہ عشقِ حقیقی کے قائل تھے۔ ان کے آباؤ اجداد میں سب پیر اور روحانی پیشوا تھے۔ آپ نے باطنی طور پر حضرت پیر عبد القادر جیلانیؒ سے روحانی فیض حاصل کیا۔ پیر عبد القادر کی منقبت سے چند اشعار ملاحظہ ہوں :

اس گناہیں کے لیے یا پیر غفاری کرو                             اب تو کچھ پردہ نشیں پر حال ستاری کرو         

دستگیری کجیو ڈرتا ہے ہاشم پر گناہ                  دستگیر بے کساں یا غوث الاعظم بادشاہ         (۸)

دنیا کی بے ثباتی بھی ہاشم کی اردو شاعری کا موضوع ہے۔ ہاشم سمجھتے تھے کہ دنیا کی زندگی عارضی ہے اور ہر کسی انسان نے دنیا سے آخرت کی طرف سفر کرنا ہے۔ اس حوالے سے اشعار ملاحظہ ہوں:

کہاں سکندر کہاں ہے دارا جام کہاں ہے جم کا                 جن کی تیغ سے دیو بھی کانپے دل دہلے رستم کا               

ان کی راکھ ملے نہ ڈھونڈے دنیا گھر ہے غم کا                 ہاشم جان...

منهج الوسطية لإمام الأشعري في علم الكلام

Although the Muslim theological schools have tried to relate their ideologies to the companions of the Prophet (Peace be upon him), the Asha’arah stressed upon this the most so as to ensure that their school of thought remains attached to the Sunni creed. Al- Ash’ari in his school and approach, always tried to keep the middle way between the Ḥanbali and M’utazili approaches, and tried to stay connected to the way of the Salaf or early generations. His moderation in approach lied in his accommodative attitude towards Sunnah as well as incorporating reason in his thought, whence he tried to avoid the two extremes followers by M’utazilah and Hanabilah, pure reason and pure tradition, in issues like God’s attributes, creation of Quran and others. The present paper tries to highlight the main features of Al- Ash’ari ’s moderate theological approach.

Effect of Induced Spawningon Hormonal, Histological and Spermatologicalchanges in Indian Major Carps

Induced spawning has gained much importance due to the increasing demand of fish seed. The seed gathered from rivers does not fulfill the requirements of the aquaculture sector. The effects of induced spawning are under discussion because utilization of Ovaprim, a synthetic hormone also raises the question of fish health. The present research was conducted to know the effects of induced spawning on hormonal, histological and spermatological parameters of Indian major carps. The results depict that Ovaprim administration result in abrupt change in testosterone, LH and FSH level in Indian major carps. The change in LH, FSH and testosterone level is not permanent and it decreases after the induced spawning. After induced spawning fish weight also decreases that is due to the release of milt and egg. The weight change also depicts the weight of milt and eggs released. Histological examination exposed Necrosis and infiltration of mononuclear cells in liver of males of Indian major carps. Another change in liver of male fishes is detachment of hepatocytes from one another. Major changes that occurred in liver of Indian major carps (Female) include cellular swelling and fatty acid changes in females. The histological examination of kidney of male fish showed clear atrophy in glomerulus. Moreover interstitial nephritis was very much evident. In addition infiltration of lymphocytes was also observed with macro phages. Interstitial nephritis and other changes that were found in kidney cells of fish (male) include permeation of mono nuclear cells & shedding of tubular cells. Histological examination of male gonads showed Lobules of testis having residual spermatids in Labeo rohita. In case of Cirrhinus Mrigala testis were observed with active stage of spermatogenesis. Lobules were found to be packed with highly active spermatids in Catla catla. Female Fish gonads were observed normal. There were found diverse phases of follicles in Catla catla. Fish Ovary was found to be with immature follicles and germinal epithelium and presence in Cirrhinus mrigala and Labeo rohita. Spermatological parameters observed showed increased sperm density, sperm motility duration and sperm motility percentage after administration of Ovaprim. Moreover percentage of live and dead sperm showed that there was increase in live sperm percentage in fishes administered with Ovaprim. So, it is concluded that induced spawning influence the fish health in many ways including the percentage of sperm motility and sperm density.
