The subject matter of the present dissertation deals with isolation, characterization and evaluation of biological activities of selected species belonging to families Solanaceae, Guttiferae and Pinaceae. The enclosed research data of the thesis is divided into following parts. PART A: Phytochemical Studies of the Selected Species of Family Solonaceae PART B: Phytochemical Studies of the Selected Species of Family Guttiferae PART C: Phytochemical Studies of the Selected Species of Family Pinaceae PART D: Evaluation of Biological Activities PART A Part A describes the phytochemical investigation on Witahinia coagulans and Physalis divericata (Solanaceae). Six new (45-50) and ten known (51-60) withanolides have been isolated from W. coagulans of Pakistani origin, whereas withaphysanolide A (61), a novel withanolide together with five known physalins (62-66) and four withaphysalins (67-70) were isolated from the P. divericata. Various experimental techniques and extensive spectroscopic studies were used for the structural elucidation of the compounds. The isolated withanolides were evaluated for inhibition activity on lipopoly-saccharide (LPS) induced B-cell, concanavalin A (ConA)-induced T-cell proliferation and against human colorectal carcinoma HCT-116 and human lung cancer NCI-H460 cells PART B Part B includes the isolation and characterization of constituents from Hypericum species (Guttiferae). Six new (105-110) and fourteen known xanthones (111-124) along with nine other compounds (125-133) have been isolated from H. oblongifolium, while six known compounds (134-139) were isolated from H. dyeri. These components were evaluated for respiratory burst inhibitory (anti-inflammatory), enzyme inhibitory and antioxidant activities. PART C Part C contains the the GC and GC-MS analysis of various extracts from conifers belonging to family Pinaceae. The amount and composition of lipophilic and hydrophilic extractives as well as proanthocyanidins in the bark of seven Pakistani conifers were analyzed. The bioactive polyphenols and other known compounds were found interesting in order to find a potential value-added use of local tree species. Gravimetrically these extracts were analysed for lipophilic and hydrophilic extractactives. The predominant lipophilic extractives were common fatty and resin acids, fatty alcohols, and sterols. Different known lignans, stilbenes, ferulates, and flavonoids were generally predominant among the hydrophilic extractives. Pinus species e.g. P. wallichiana, P. gerardiana and Picea smithiana showed large amounts of lipophilic and hydrophilic extractives compared to the other examined conifers. Pinus roxburghii was found different from the other pine species having smaller amounts of both types of extractives. A. pindrow and T. fuana were also found to have the smallest amount of hexane extracts. The proanthocyanidin content and composition revealed that especially Pinus wallichiana and Abies pindrow could be rich sources of such compounds. PART D Part D is concerned with evaluation of biological activities of crude extracts, fractions, semi-pure and pure constituents. Different solvents soluble fractions of the selected plants belonging to family Guttiferae (H. perforatum, H. oblongifolium, H. monogynum, H. choisianum and H. dyeri), Pinaceae (bark and knotwood of Picea smithiana, Abies pindrow, Pinus wallichiana, P. geradiana, P. roxburghii and Cedrus deodara) and Taxus fauna from the north west of Pakistan were screened for their possible antioxidant activity. Anticancer (anti-proliferative) and enzyme inhibitory activities of Hypericum species as well as the cytotoxic, anti-inflammatory and urease inhibitory activities of pure compounds isolated from Hypericum, Physalis and Withania species were also studied. Four complementary test systems, namely phenolic compounds, free-radical scavenging capacity, measuring of reducing power and total antioxidant activities by Phosphomolybdenum method were used for analysis. We report here for the first time the antioxidant and antimicrobial potential of the various extracts and fractions of the listed plants for the first time except Hypericum perforatum which has been the subject of many investigations. The objectives of this study were to explore the biological and medicinal value of the extract/fractions of the above mentioned plants.

افسوس ہے کہ گزشتہ اپریل کی ۲۹؍ تاریخ کو علم و عمل اور فضل و کمال کی ایک اور شمعِ فروزاں گل ہوگئی، مولانا مفتی سید محمد مہدی حسن شاہجہانپوری نے ۹۶ سال کی عمر میں بعارضہ فالج داعی اجل کو لبیک کہا مرحوم اس عہد میں اگلی صحبتوں کی چند بقیۃ السلف یادگاروں میں سے تھے، وہ اتباع سنت، تبحر علم، وسعت نظر، طہارت و تقویٰ زہد و ورع اور کتاب و سنت کی تفسیر و تعبیر میں یگانہ عہد تھے، دارالعلوم دیوبند کی مسند درس و افتا ان کے فیضان کمال سے ایک عرصہ تک بارونق رہی ہے اور سیکڑوں تشنگان علم ان کے منبع فیض سے سیراب ہوئے، وہ بلاشہ معلومات کے دریا، حافظہ کے بادشاہ اور وسعت علم کی ایک نادر مثال تھے، ہر مجلس و محفل میں یکساں ان کی قدر و منزلت تھی، حدیث کے ساتھ فقہ کے جزئیات پر ان کی وسعت نظر مسلم خیال کی جاتی تھی۔

EFFECTS OF CRANIOCERVICAL FLEXION EXERCISES AND SCAPULAR STABILIZATION EXERCISES IN NECK PAIN AND FORWARD HEAD POSTURE AMONG FEMALES WEARING HEADSCARVES

Background: In neck related problems, limited cervical ranges are considered a major contributor leading to forward head posture and increased disability. Methodology: The study design was a randomized clinical trial. Total of 50 subjects were recruited that fulfilled the selection criteria. Subjects were divided into two groups. Craniocervical flexion exercises group A (n=25) and Scapular stabilization exercises group B (n=25). Numeric Pain Rating Scale (NPRS) and Neck Disability Index (ND1) were subjective while the goniometer and plumb line were used as objective modes of assessment. Baseline and after 4 weeks of treatment values were assessed by using SPSS 21. Results: Both groups showed improvement in plumb line values, forward head posture and cervical spine mobility. NPRS, and NDI score values after the 4-week exercise program showed significant results (p-value<0.05). Outcomes except for plumb line value and NDI score indicated statistically significant improvement in the Craniocervical flexion exercise group than the scapular stabilization exercise group (p-value<0.05). However, in NPRS no statistical difference (p-value>0.05) was found between the two groups. Conclusion: Only females were recruited. Further studies can correlate with spatiotemporal features. The idea and conceptual framework were original.

Studies on Bacteriocins from Environmental Bacteria

The present study is primarily concerned with bacteriocins from environmental bacteria. Different bacterial strains were isolated from various environments including hospital air, soil and sewage etc. A total of 51 isolates were screened for bacteriocinogeny, of which 41% (21 isolates) were exhibiting bacteriocinogeny. Two isolates, one of Pseudomonas sp. 40 and Bacillus subtilis strain15 were selected for further studies. Antimicrobial substance from Pseudomonas sp. 40 was neither sensitive to proteolytic enzymes nor showed any antibacterial activity on SDS-PAGE indicating its possible non-proteinaceous nature and hence further studies were not carried out on this strain. Only Bacillus subtilis stain 15 was further studied as it was producing some antimicrobial substance of protein in nature. BLIS of Bacillus subtilis strain 15 was produced during the late exponential-phase or at the beginning of stationary-phase. The inhibitory substance/BLIS was partially purified by ammonium sulphate precipitation. The antimicrobial substance was characterized as a bacteriocins-like inhibitory substance with a molecular weight ranged between 3-5 kDa as determined by SDS-PAGE. BLIS showed the inhibitory activity against locally isolated Bacillus species as well as Bacillus subtilis ATCC 6633 and Bacillus cereus ATCC 11778, Listeria monocytogenes, Staphylococcus aureus, Staphylococcus epidermidis and also showed slight activity against Salmonella typhi and Shigella dysenteriae and those involved in the food-poisoning and spoilage such as Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Micrococcus luteus were inhibited that indicates its possible application in food preservation. BLIS acted bacteriostatically rather than bacteriocidally, on the sensitive cells. It may exhert bactericidal action at higher concentrations. BLIS under study was heat stable up to 80°C as its activity was not affected at this temperature. The BLIS retained its activity over a wide range of pH i.e., from 3-9. It was sensitive to proteinase-K, and α-chymotrypsin indicating about its nature as protein. Its activity remained unaffected in presence of organic solvents like acetone, chloroform, methanol and EDTA except ethyl-alcohol. BLIS from Bacillus subtilis strain 15 showed the effective inhibition of superoxide production in both neutrophils and macrophages. It also had inhibitory effect on whole-blood. The inhibition of superoxide production shows that BLIS may have anti- inflammatory or immuno-suppressive effect. In addition to immunomodulatory activity, BLIS from Bacillus subtilis strain 15 had moderate anti-oxidant activity indicating its probable role in anti-tumorogenic activity.
