The present study was designed to compare the sensitivity and specificity of different parasitological and molecular techniques for the detection of Trypanosoma sp. during different seasonsof year 2013 in equines and camels of Dera Ghazi Khan District and to determine the risk factors associated with the spread of trypanosomiasis. The objective of this project was to demonstrate the effect of Trypanosoma sp. on complete blood count (CBC) and selected serum biochemical parameters in camels, horses and donkeys from Southern Punjab (Pakistan). A total of 858 blood samples (291 camels, 284 horses and 283 donkeys) were collected and were subjected to blood smear examination, hematocrit centrifugation technique and Polymerase Chain Reaction (PCR) (by amplifying kinetoplast maxicircle DNA) to determine the prevalence of trypanosomiasis in collected samples. Blood samples were also analyzed for complete blood count and selected serum biochemical parameters (Total Protein, Creatinine, Alanine Transferase (ALT), Aspartate Transferase (AST) and Triglycerides). Out of 291 camel blood samples, 28 (9.62%) generated a 164 bp DNA fragment specific for Trypanosoma sp. Only 6 blood samples from camels (2.1%) were found positive through microscopic examination while 13 (4.46%) were positive for Microhematocrit centrifugation technique. Out of 284 blood samples from horses 16 (5.63%) samples were found Trypanosoma sp. positive by PCR. Only 5 blood samples (1.8%) were found positive through microscopic examination while 9 (3.17%) blood samples from horses were positive for Microhematocrit centrifugation technique. Out of 283 blood samples of donkeys, 19 (6.71%) samples amplified a 164 bp DNA fragment specific for Trypanosoma sp. while only 7 blood samples (2.5%) were found positive through microscopic examination and 9 (3.18%) were found positive for Microhematocrit centrifugation technique. Seasonal prevalence through PCR was 6.9% (5/72) in spring, 13.7% (10/73) in summer, 9.7% (7/72) in autumn and 8.1% (6/74) in winter in camels. Seasonal prevalence through PCR was 4.2% (3/72) in spring, 5.9% (4/68) in summer, 7.04% (5/71) in autumn and 5.48% (4/73) in winter in horses. In donkeys, seasonal prevalence through PCR was 4.76% (3/65) in spring, 8.22% (6/73) in summer, 8.45% (6/71) in autumn and 5.41% (4/74) in winter. No significant association (P˃0.05) of disease was observed with age of all studied animals. Trypanosomiasis was also found not to be associated (P˃0.05) with the gender of equines and camels during present study. A significant increase was detected in white blood cells (WBC) (P˂0.001), neutrophils(P˂0.004) and ALT(P˂0.028) in Trypanosoma sp. positive as compared to the Trypanosoma sp. negative blood samples while a significant decrease in red blood cells (RBC) (P˂0.001), hemoglobin (P˂0.001), lymphocytes (P˂0.003) and packed cell volume (P˂0.001) was observed in blood samples of camels. In horse blood samples, there was also significant increase in WBC (P˂0.001), neutrophils (P˂0.001) and ALT (P˂0.032) and a significant decrease (P˂0.001) in RBC, hemoglobin (P˂0.001), lymphocytes (P˂0.021) and packed cell volume was observed in parasite positive as compared to the parasite negative blood samples. In donkeys, a significant increase was detected in WBC (P˂0.002), neutrophils (P˂0.001) and ALT (P˂0.019) in Trypanosoma sp. positive as compared to the Trypanosoma sp. negative blood samples while a significant decrease in RBC (P˂0.001), hemoglobin (P˂0.003), lymphocytes (P˂0.001) and packed cell volume (P˂0.003) was observed. In conclusion, PCR was found more reliable and sensitive technique than the blood smear examination and microhematocrit centrifugation technique for Trypanosoma sp. detection in blood samples of equines and camels and it is recommended to be used for Trypanosoma sp. detection in epidemiological surveys and control policies in livestock sector to minimize economic losses.

PENDIDIKAN AGAMA ISLAM (PAI) MULTIKULTURAL PERSPEKTIF PEMBELAJARAN INTEGRATIF

Tulisan ini mengkaji tentang PAI multikultural sebagai model pembelajaran integratif yang memadukan antara ilmu pengetahuan dan agama. Dari aspek konten, multikulturalisme mengkaji keragaman bangsa, suku, warna kulit, bahasa, agama dan keragaman lain yang terbentang dalam realitas sosial kehidupan manusia sebagai ayat-ayat Allah yang bersifat kauniyah. Sedangkan PAI mempelajari normativitas ajaran Islam dan dimensi historis yang banyak termuat dalam al-Qur’an sebagai ayat-ayat Allah yang bersifat qauliyah. Pembelajaran kedua bidang keilmuan tersebut sama-sama sebagai proses pencarian kebenaran yang merujuk kepada Allah sebagai episentrum kebenaran Hakiki dan sumber ilmu pengetahuan. Dari konsep pembelajaran integratif yang dikemukakan pemikir Islam dan Barat, PAI multikultural berada pada pola relasi antardispliner, yaitu integrasi antara ilmu umum dan ilmu agama. PAI multikultural sebagai pembelajaran integratif tergambar pada aspek: tujuan, materi, metode, media dan sumber belajar dan penilaian yang saling terintegrasi dalam membentuk kompetensi beragama peserta didik secara integral, yaitu: keterpaduan antara unsur duniawi dan ukhrawi, antara dimensi spritual dan intelektual, antara ranah personal dan sosial dalam konteks membangun harmoni kehidupan warga sekolah, masyarakat dan bangsa Indonesia yang pluralistik dari segala aspeknya.

Pomegranate Seed Residue As Waste Exploitation to Augment Shelf Life Stability and Neutraceutical Prominence in Edible Oils

The present study was aimed to investigate and utilize the pomegranate seed residues that considered as waste after processing. The current study was an attempt to explore the functional and nutraceutical potential of pomegranate seed extracts i.e. pomegranate seed oil (PSO) and defatted pomegranate seed residues (DPSRs) after juice extraction. Three different varieties (Desi, Kandhari, and Bedana) of Pakistani pomegranate varieties were selected and analyzed comparatively. The comparative study of pomegranate seed oil showed the highest lipid contents (15.90±1.40%) in Desi variety followed by Kandhari (11.00±1.25%) and Bedana (8.70±1.10%). The six different fatty acids were found in each variety. The highest contents of conjugated linolenic acid (punicic acid) as 76.66±1.00, 75.50±1.20 and 68.52±1.63% in Desi, Kandhari and Bedana variety, respectively followed by linoleic acid, oleic acid, palmitic acid, stearic acid and myristic acid. Data also showed the significant difference between physico-chemical parameters in all oil samples. The highest antioxidant activity and oxidative stability was observed in Desi seed oil as compared to other varieties. Defatted pomegranate seed extract (DPSE) was prepared in different organic solvents. A significant difference was observed in proximate and mineral contents of defatted residues of all varieties with maximum antioxidant potential in Desi variety extract with promising anti-inflammatory activity with no cytotoxic effect on RAW264.7 macrophages. According to the results of 1st phase of study, Desi variety was selected for further study. The pomegranate seed extracts (PSO + DPSE) were blended with plant-based oils (canola and sunflower oil) at different concentrations and subjected to six month storage and frying study. The treatments and storage showed significant (p≤0.05) effect on physico-chemical attributes of oil blends. The oxidative stability and antioxidant activity decrease with storage. A decrease was observed in unsaturated fatty acids and tocopherol contents with storage. The French fries prepared in TC7 and TS7 treatments showed even oil distribution as compared to other treatments. Oil uptake decreased in French fries by increasing the DPSE in oil blends while increased with storage. Treatments had non-significant (p≤0.05) on sensory attributes. The experimental rats modeling comprised of three segments including normal, diabetic and hypercholesterolemia Sprague Dawley rats. Each study was divided into further 4 groups D1, D2, D3 and D4. The D1 and D3 were normal diets of canola and sunflower oil, respectively while D2 and D4 were experimental diets of canola and sunflower oil with 7% PSO + 800ppm DPSE, respectively. The maximum cholesterol reduction was observed in canola oil groups in all studies as 1.20, 11.38 and 16.30% following 0.80, 9.80 and 12.68% in sunflower oil group. Similarly, significant reduction was observed in LDL and triglycerides level, while an increase was observed in HDL level as 11.26 & 8.02% and 12.84 & 10.05% in study II & III in canola and sunflower oil groups, respectively. As far as, antidiabetic perspectives are concerned, a substantial decrease in serum glucose level was observed as 7.04 & 6.01% and 4.59 & 3.93% in study-II and III of canola and sunflower oil groups while an increase in insulin level was noticed. The percent increase in insulin level was observed as 1.70 & 0.92%, 3.97 & 3.19% and 2.31 & 2.15% in study-I, II and III, respectively. Moreover, for the assessment of the anti-oxidative markers, glutathione level was improved and thiobarbituric acid reactive oxygen substances (TBRAS) level was reduced by utilization of pomegranate seed extract addition. The analysis of liver and kidney functioning tests ensured the safety of pomegranate seed extracts. It is obvious from the data of efficacy study that canola oil with pomegranate extracts as natural anti-oxidants is more effective as compared to sunflower oil. It is inferred from the present exploration that Desi variety of pomegranate is more effective as compared to other varieties available in Pakistan
