Streptokinase (SK) is a fibrinolytic protein which has been produced by several species of β-hemolytic Streptococci. It has been extensively used to treat cardiovascular disorders by activating plasminogen into plasmin and eventually dissolving the blood clot. The objective of the current research project was to exploit indigenously isolated Streptococcus agalactiae EBL-20 for the production of SK and enhancing its productivity by Ultraviolet (UV) irradiation and ethyl methanesulfonate (EMS) induced mutations, optimized culture conditions and nutritional requirements. Four mutants namely, S. agalactiae EBL-31, S. agalactiae EBL-32, S. agalactiae EBL-33 and S. agalactiae EBL-34 were selected by different screening procedures using triton X-100, casein plasminogen overlay assay and clot lysis assay. The optimum levels of four process variables (pH, temperature, incubation time and inoculum size) were obtained by Response Surface Methodology (RSM). According to statistical results, a temperature value of 37.5⁰C and pH 7 of the fermentation medium with 2.50 mL inoculum size for 36 hours of incubation was considered as optimum for enhanced SK production. After optimization, mutant derived strains EBL-31, EBL-32, EBL-33 and EBL-34 resulted in 2.01, 2.30, 3.70 and 2.48 fold increased activity as compared to parent S. agalactiae EBL-20, respectively. The best mutant obtained after performing optimization studies was found to be S. agalactiae EBL-33 with 3.70 fold increased activity, hence it was purified and exploited for characterization studies with reference to S. agalactiae EBL-20.The purification of SK by DEAE-cellulose and Sephadex-G100 resulted into specific activity of 447.6 and 1278.87 U mg⁻¹ from parent S. agalactiae EBL-20 and mutant derived S. agalactiae EBL-33, respectively. The molecular weight of SK obtained from both the strains of S. agalactiae was found to be 47 kDa. Study of kinetic and thermodynamic parameters revealed that SK obtained from S. agalactiae EBL-33 has more potential for utilizing its substrate i.e., plasminogen with high thermostability, in contrast to SK derived from S. agalactiae EBL-20. The study concludes that both UV rays and EMS are effective mutagenic agents for enhanced production of SK from S. agalactiae. Moreover, enhanced SK yield can be obtained by optimizing the nutritional necessities and fermentation/cultivation conditions of the microorganism.

This research addresses Islam's view of the social ties that man creates by virtue of his living among the people or in which he borns as its member, and aims to answer several questions about the relationship of these social ties with the Islamic brotherhood bond that Islam has brought. Does Islam recognize the social ties? On which basis does Islam recognize them and why? What is their position in front of Islamic brotherhood? How does Islam invest these ties to achieve religious, psychological and social security? Are there any conditions that Islam has developed to recognize and nurture these ties? The answer to these questions comes through the Qur’ānic texts, the prophetic Hadiths and the events of the Prophet's biography, based on open and direct reference, and away from the ambiguous interpretations or weak evidences; in order to clarify this matter clearly, and to check the validity of the results of the study.

Crisis and Emerging Regional Order in the Middle East

The strategic importance of Syria in the Middle East is mainly due to its location. Syria has been a kind of resistance front of the Arabs against Israel and the significance of the Syrian question needs to be discovered in Syria's indispensible role in the regional geostrategic pattern. When Arab Spring brought political changes in Tunisia, Libya and Egypt, Syria was never thought to be immune to the tinge of those changes. The mass demonstrations in Syria, driven by the desire of change, in late 2010, turned to violent protests, leaving destruction, disorder and chaos behind them. The crisis in Syria is still continuing ferociously with abysmal repercussions for the Middle East. Today, Syrian crisis has different dimensions which are ranging from involvement of different regional and trans-regional states to various armed, religio-political and jihadists groups, turning the crisis to a serious regional conflagration. The Assad regime is struggling for its survival against the rebels, who are struggling for regime change in Syria. The involvement of the pro and anti-regime regional states and pro and anti-regime trans-regional states is the significant point, driving the crisis on grave sectarian, ethnic and social lines. In fact, it would be safer to say that Syrian crisis has created deep political divisions not in only amongst the world community but also on regional political horizons. Syrian crisis has created far reaching social, political, strategic and economic implications in Syria and the entire Middle East. The crisis is not only claiming priceless lives of people, crippling many for life, rendering numerous homeless but also altering the traditional security dynamics, strategic calculations and above all regional order of the Middle East. To sum up, Syrian crisis has become one of the gravest flash points in the Middle East, turning the social and political fabric of the society there. It has given birth to a regional order in Middle East where Iran is emerging as the most powerful regional reality, shaping the future political contours of the region on its own version of interests.
