Monoamine oxidase inhibition offers potential therapeutic target for the management of depression and Parkinson’s disease. Keeping this in view, the present study was designed to select and synthesize potential scaffolds followed by monoamine oxidase inhibition, computational studies, behavioral and neurochemical assessments. Five different types of scaffolds (pyrazolobenzothiazine-based carbothioamides, chromones, chalcones and quinoline carboxylic acid derivatives) and a synthetic compound oxatomide were selected for this study based on their potential to interact therapeutically with MAO enzyme. Their various derivatives were synthesized and structurally characterized using spectroscopic methods i.e., mass and NMR. All compounds were subjected to monoamine oxidase A and B inhibition assay using the kynuramine or the amplex red method. The data showed that pyrazolobenzothiazinebased carbothioamides held significant potential to inhibit both MAO-A and B. Compounds 2-(4-(3,4-dimethyl-5,5-dioxidobenzo pyrazolo[4,3-c][1,2]thiazin-2(4H)yl) benzylidene)-N-(p-tolyl) hydrazine carbothioamide (3b) (IC50 = 0.003 ± 0.0007 mM) and N-(4-ethylphenyl)-2-(4-(4-methyl-5,5-dioxido-3-phenylbenzo pyrazolo[4,3c][1,2]thiazin-2(4H)-yl) benzylidene) hydrazine carbothioamide (4d) (IC50 = 0.02 ± 0.001 mM) inhibited the enzyme with efficacy comparable to that of the standard inhibitors, clorgyline and deprenyl. Computational docking studies revealed that 3b showed interaction to Tyr407 and Tyr444 pi-interactions from 4-methylphemyl zone of molecule, and the bezothiazine ring was oriented to hydrophobic pocket of the active site. The amino acids Ile325, Ile335, Phe352, Tyr69 and Tyr197 showed Van der Waals interactions while amino acids Tyr407, Gln215, Val210, Phe208, Ile180, and Cys323 showed polar interaction to flavin adenine dinucleotide (FAD). Kinetic data revealed compound 3b as a noncompetitive inhibitor. Phenothazine ring of 4d showed pi-stacked-interactions to Tyr398 and Tyr435. Ethylphenyl-side was oriented to entrance cavity. Van der Waals interactions were found with Phe103, Leu164, Phe168, Ile199, Tyr326 and Phe34. Apart from π-π interactions with the Tyr398 and Tyr435, FAD and other residues such as Gln206 and Ile316 showed polar interactions with 4d. Thus aryl substitution was necessary for the observed function of these molecules. Enzyme kinetics showed 4d to be a competitive inhibitor for MAO-B. These compounds were tested for their efficacy in-vivo. The compound 3b caused significant decline in the immobility time in tail suspension test, while compound 4d significantly reversed the effect of rotenone as suggested by the remarkable increase in time spent on Rotarod. Hence, it can be deduced that both of this compounds have the ability to reach CNS to produce their desired effects. Their effects were further confirmed by neurotransmitter analysis. The brains from 3b treated animals showed significant increase in the levels of norepinephrine and serotonin alone with decline of its metabolite i.e., 5-hydroxyindoleacetic acid. However, the 4d treatment caused significant increase in dopamine levels with corresponding decline in its metabolites i.e., 3,4 dihydroxyphenylacetic acid and homovanilic acid. The aforementioned analysis is suggestive of preferential effect of 3b and 4d on MAO A and B, respectively in vivo. Hence, both of these compounds are suitable candidates for the development of MAO inhibition based therapy for the management of depression and Parkinson’s disease.

میں ندیم صادق صاحب سے حال ہی میں متعارف ہوا ہوں۔مجھے ان کے پہلے مجموعہ کلام کا مسودہ دیکھنے کا موقع ملا ہے۔ یہ میرے لیے ایک بہت خوش گوارتجربہ تھا۔ میرے لیے دل چسپی کی خاص بات یہ ہے کہ حصہ غزل کی تمام غزلیںناصر کاظمی کی’’ پہلی بارش‘‘کی طرح ایک ہی زمین میں ہیں۔ ان کی بحر بھی وہی ہے اور قافیہ بھی، ردیف البتہ مختلف ہے۔ شاعر نے کئی اشعار میں بھی ناصر سے اپنے لگائو کا اظہار کیا ہے۔ یہ کلاسیکی روایت سے متاثر اور وابستہ شاعر کا کلام ہے۔ کتاب کا عنوان میر کے شعر سے اخذ کیا گیا ہے اور انتساب ناصر کاظمی کے نام ہے۔
ایک ہی زمین میں اتنی غزلیں کہہ لیناکوئی آسان کام نہیں۔شاعر اس کے لیے خصوصی داد کا مستحق ہے۔ پہلی غزل حمدیہ ہے اس کے بعد اکیس غزلیں دو انسانوں کے ملنے بچھڑنے اور ان کے تعلق کے مختلف پہلوئوں کا بیان ہیں۔ چند اشعار دیکھیے:
یاد کے بوٹے سوکھ نہ جائیں
دل دریا پانی دیتا ہے

وہ تجھ کو کیوں یاد کرے گا
صادق وہ مصروف بڑا ہے

ساری گلیاں گھوم چکا ہوں
تیری گلی سے ڈر لگتا ہے

شہر کی سڑکیں تو ٹھنڈی ہیں
لیکن میرا دل جلتا ہے

دل میں کیسا خوف بھرا ہے
پھول کھلے تو ڈر لگتا ہے

اب دل تنہا خوش رہتا ہے
میں نے خود کو بدل لیا ہے

صادق فون نہ کر تُو اس کو

نجيب الكيلاني في مرآة الأدباء والشعراء

Dr. Najib Al- Kailani (1931-1995) is one of the prominent poet and writer of Arabic literature. The multidimensionality of his personality is rare example of the history of Islamic literature. He was a renowned poet, writer, a medical professional, a thinker and overall a great human being. He laid the foundation of the Islamic literature and introduced a sound methodology.  His precious literary works in Arabic and Islamic school of thought made his personality more prominent. He not only discusses the issues and challenges faced by Egyptian Muslims but also covers the whole Islamic world and as well as those Muslims who are in minority in their countries. His literary works are a good source of knowledge for not only the   general readers but for the intellectuals and writers as well. Research works are being carried out on different dimensions of his personality in different ways by the different universities in the world. Research articles and discussions are being carried out and conferences are also organized in different Arab countries in the memory of Dr Najib Al-Kailani. Kailani was paid rich tributes by Arab writers and thinkers for his unique wealth of literary works. In this article we present the views of Arabian writers about his literary status.

Impact of Higher Education On Gross Domestic Product

Impact of Higher Education On Gross Domestic Product Education is the key to the socio-economic development of an economy. It plays a very important role to enhance the capabilities of individuals and to speed up the economic growth of a country. The positive outcomes of education include eradication of poverty and inequality, improvement in health conditions of individuals, enhancement in social status and overall strengthening of a society. Education also provides essential elements for policy framework. Pakistan is a developing country which needs special attention towards education policies in order to enhance and continue the economic growth. The study aims to investigate impact of higher education on Gross Domestic Product of Pakistan from 1981 to 2013 using econometric approach Johansen Cointegration (1995), Granger causality approach in Vector Autoregressive framework and Unit root test. The main emphasis was on to know the impact of Higher Education Commission since its inception. It examines whether higher education effect GDP in Pakistan. The econometric analysis reveals that there is a long run relationship between higher education and GDP, which provide evidence that these variables are necessary for each other. The empirical results of causality test specify that there exists a unidirectional causality running from economic growth to higher education and no other direction of causality found between these variables.
