This study aimed to explore Grade 10 students' perspectives concerning the biology topics that they find the most difficult to learn, the reasons of experiencing difficulties in learning biology, and ways to overcome the difficulties. The study employed both quantitative and qualitative approaches. A survey tool difficulties and effectiveness in Learning Biology questionnaire (DELBQ) was adapted from the literature which has already been used in other context in a similar study (Cimer, 2012). 132 Grade 10 students participated in the cross-sectional survey including girls (n=86; 65%) and boys (n=46; 35%). The sample was recruited from 10 public schools of district Hunza. DELBQ was directly administered to all participants. The results showed that students have identified five concepts of biology - genetic engineering (37%), cell division (31%), Mendel principles and application (28%), defense and immunity (23%) and matter cycle (17%) - as the most difficult concepts to learn. The students proposed multiple reasons to learning hindrances which include: the nature of the topic, teachers' teaching method, students' learning techniques, students' attitudes towards the topics and deficiency of resources and time. Prominently, of five the themes 'the nature of the topic' was found to be the most repeatedly cited reason (99) for difficulty in learning biology. Students have not only identified these hindering factors of learning they have talked about viable suggestions to improve learning of the subject such as reducing the subject content; using visual aids in teaching ; teaching through experiments; students using various study techniques; making biology teaching interesting and effective; and, teaching biology by connecting the topics with daily life. Interestingly, a majority of responses (99) highlighted 'the nature of topic' as one of the primary reasons for making biology learning difficult; however, the suggestions to enhance learning were predominantly related to either 'students' own learning habits' (148) or 'teachers' teaching style' (127). The study provides an insight into 'students' voices' not only in identifying the 'difficult areas in Biology' but highlighting perspectives in explaining the 'reasons for difficulties in learning these concepts' and potential strategies to improve learning. The results could be useful for practitioners and policy-makers in enhancing curriculum and classroom practices to make this fundamentally important subject interesting for students. The tool adapted for the study is a valuable addition to the indigenous literature which can be used by the other researchers in the field to conduct large scale studies.

FUNCTIONAL OUTCOME AMONG PATIENTS WITH ARTHROSCOPIC ANTERIOR CRUCIATE LIGAMENT RECONSTRUCTION WITH ENDO-BUTTON AT GHURKI TRUST TEACHING HOSPITAL POST 6 MONTHS-A SINGLE CENTERED SURVEY

Background of the Study: To assess the functional outcome among patients with arthroscopic ACL reconstruction with endo-button at Ghurki Trust Teaching Hospital post 6 months. Methodology: A single centered survey was conducted on 67 patients with arthroscopic AC Ligament reconstruction using Hamstring auto-graft (Semitendinosus-Gracilis tendons) after 6 months and evaluated for functional outcome. The data was collected from Ghurki Trust Teaching Hospital. The sampling procedure utilized was non-probability sampling. Functional outcome was assessed using subjective form of IKDC score. Data was analyzed using SPSS version 23. Results: Out of 67 patients, all were male. The pre-operative subjective IKDC scoring was less than 30 and post-operative mean IKDC scoring is 73.92. On the basis of findings of study, significant improvement was observed after ACL reconstruction post 6 months. Conclusion: ACL reconstruction shows significant improvement in knee function and significant recovery of preoperative functional status.

Biodegradation of Organochlorinated Pesticides by Indigenous Soil Microorganisms

Biodegradation of Organochlorinated Pesticides by Indigenous Soil Microorganisms Endosulfan is a broad spectrum organochlorinated insecticide extensively used all over the world to enhance the agricultural production. Its widespread application creates severe ill effects on human and environment. The present study was aimed to isolate and identify the bacterial strains that have capability to metabolize endosulfan. Initially, eight bacterial isolates were screened out on minimal salt medium, but two more efficient bacterial isolates i.e., EN-1 and TAH were identified on the basis of 16S rRNA technique. Both isolated bacterial strains had the ability to utilize endosulfan as a sole sulfur source. These isolates were named as Stenotrophomonas maltophilia (EN-1) and Pseudomonas sp (TAH). Degradation study was conducted at various initial concentrations of endosulfan, i.e., 5, 25, 50, 75 and 100 mg/L along with the different incubation temperatures i.e., 25, 28, 32, 37 and 45ºC. The maximum endosulfan degradation by EN-1 and TAH was recorded at 32ºC and 28ºC respectively. It was observed that these bacterial isolates could degrade 80-85 % of endosulfan in 5 days of incubation, but the maximum degradation (80-85 %) was recorded when 100 mg/L of endosulfan was spiked. GC-MS analysis revealed that endosulfan diol and endouslfan lactone were major metabolites accumulated. Therefore, it was confirmed that both isolates followed hydrolytic pathway for endosulfan degradation instead of oxidative. Biodegradation kinetics of EN-1 and TAH were also studied at various initial endosulfan concentrations ranging from 5- 100 mg/L. Growth dependent and independent kinetic models were applied to access the rate of endosulfan biodegradation. The average ratio of µmax/ Ks increased when endosulfan used as a sole sulfur source, while its value decreased when additional sulfur source was added along with endosulfan. So it was found that additional sulfur did not enhance the rate of endosulfan degradation. In the present study plasmid DNA was also cured in order to locate the degrading genes. Endosulfan degrading genes were found on chromosomal DNA in EN-1 bacterial strain while TAH had plasmid borne genes. These plasmid mediated degrading genes were identified by cloning. The resulted clone product showed 97 % similarity with Nta monooxygenase gene. Comparative protein analysis was done by the SWATH MS technique. Thiswas the first studyin which thequantificationand identification of bacterial proteome by using cell lysate.Out oftotal, 48 proteins were similar in both isolated bacterial strains which were suspected to be involved in the endosulfan degradation. Among these 48 proteins, 10 proteins were involved in various enzymatic functions. Thus, there might be 10 proteins involved in the degradation of endosulfan. All other proteins were unknown in their function.
